The growing approaches to reduce the cost of cellulases production can be achievd viausing of cheap , renewable agriculture wastes which consider an excellent carbon source for microbial enzymes production and also looking for microorganism having the ability to hyper-production of cellulases. Therefore, considering the advantages of SSF, the optimal chemical condition for the production of cellulase enzyme by Apergillus nigerfrom solid -state fermentation of sugarcane bagasse is investigated.

Book excerpt: Cellulase is a key enzyme of industrial interest and plays a crucial role in the hydrolysis of cellulose, a prime component of plant cell walls. Cellulase covers a broad area in the global market of industrially important enzymes and it is considered as the third largest industrial enzyme globally. Additionally, cellulase contributes about 20% of the total enzyme market globally because of its massive demand in various industries such as in biofuel production, pulp, paper, textile, food, and beverages, as well as in detergent industries. Among these, the demand of cellulase may become frequently selected in the commercial production of biofuels in the future and thus will further increase demand of cellulase in the biofuel industry. Because biofuel production is still not realized in a cost-effective, practical implementation due to its high cost (the higher cost of biofuels is due to higher production costs of enzymes), there is a need to introduce these types of approaches, which will help to lower the cost of enzyme production for developing overall economic biofuel production.

In the present study, optimization for cellulase production by Aspergillus niger was investigated by using paddy straw as substrate.Cellulase enzyme was produced by the fungus extracellularly. Optimization of cellulase production was done by using various physical (Temperature, pH, Salinity and Incubation time) and chemical parameters (Carbon sources and nitrogen sources) which could influence the enzyme activity. Cellulase production was maximum at the temperature 20°C and minimum at 40ºC. The optimal pH for the cellulase production was observed maximum in 6.0 and minimum in 7.0. Cellulase production was maximum at 48 hours and minimum at 24 hours. Cellulase production was maximum with when fructose was used as a carbon source and minimum with sucrose. Cellulase production was maximum when Malt extract was used as a nitrogen source and minimum with yeast extract. The cellulase enzyme extract was partially purified and its protein fraction was stained with its molecular weight determination by using SDS-PAGE. It revealed two protein bands with the molecular weights of about 83 kD and 50kD.

Microbial cellulases have become the focal biocatalysts due to their complex nature and wide spread industrial applications. In addition, cellulase production is the most important step in the economical production of ethanol, SCP and other chemicals from renewable cellulosic materials. To date, the production of cellulase has been widely studied in submerged culture processes, but the relatively high cost of enzyme production has hindered the industrial application of cellulose bioconversion. It has been reported that solid state fermentation is an attractive process to produce cellulase economically due to its lower capital investment and lower operating expenses. In this book, authors experimentally analyzed and illustrated the suitability of locally available cheap agro-residues as solid supports for the growth of A. niger in solid state fermentation for production of cellulolytic enzymes. In addition, a variety of leachate methods for maximum recovery of enzyme from the solid-state fermentation of bran were explained in this book.

Book excerpt: ABST ACT The use of waste as raw material is important for government economy and natural balance. The purpose of this work is to study the production of xylanase and CMCase enzyme by Aspergillus niger in solid state fermentation (SSF) using agricultural residues sugar cane bagasse as substrates. The study is focus on the effect of substrate concentration which is 1 g/L, 3g/L and 5 g/L. All the enzymes were incubating in temperature 37 oC and pH 5.0 for 30 minutes. The reaction was quenched by cooling on ice and the amount of reducing sugar was measured by dinitrosalicyclic (DNS) acid procedure with slight modification (Miller, 1965). The mixture was then incubated at 100 oC for 5 minutes. Subsequently, the reaction was stopped by cooling on ice. Distilled water was added into the final volume of 16 ml and the color intensity was measured at wavelength of 575 nm. The concentration of SCB affected the xylanase and CMCase activity. Xylanase and carboxymethylcellulase activity was higher at lower concentration hen using 3 g of SCB.
