Book excerpt: Optical feedback interferometry (OFI) is an emerging sensing technique which has been studied in fluidic systems. This sensing scheme is based on the modulation of the laser emission output power and/or the junction voltage induced by the interaction between the back-scattered light from a distant target and the laser inner cavity light. Thanks to the Doppler Effect, OFI can precisely measure the velocity of seeding particles in flowing liquids which is much required in chemical engineering and biomedical fields. In the present thesis, optical feedback interferometry performance for microscale flow sensing is studied theoretically and experimentally. A new numerical modeling approach based on multi-physics numerical simulations for OFI signal simulation in the micro-scale flowmetry configuration is presented that highlight the sensor performances. In this model, many factors are involved such as particle concentration and laser-particle scattering angle distribution and flow velocity distribution. The flow rate measurement shows good agreement with the modeling. The implementation of OFI based sensors in multiple fluidic systems, investigating the impact of the fluidic chip specific configuration on the sensor signal.Finally, a compact OFI flowmetry sensor based on Vertical-Cavity Surface-Emitting Lasers (VCSELs) using micro optical fabrication techniques is demonstrated as well. The simulation method for the design and the microfabrication procedures are detailed. After an evaluation of the experimental results, the capabilities of this new OFI sensor in microfluidic measurements are emphasized, thus demonstrating an open path towards ultra-compact microfluidic systems based on the OFI sensing technique.

Book excerpt: Recent progress of interferometric sensors based on the optical feedback in a laser diode have demonstrated possibility for measurement of flow rates and flow-profiles at the micro-scale. That type of compact and embedded sensors is very promising for a research and industrial field -microfluidics - that is a growing domain of activities, at the frontiers of the physics, the chemical science, the biology and the biomedical. However, the acquisition of flow rate or local velocity at high resolution remains a very challenging issue, and the sensors that have been proposed so far did not have been giving sufficient information on the nature of the particles flowing. The present thesis is driven to the implementation, validation and evaluation of the sensing performances of the optical feedback interferometry technology in both chemical and biomedical fields of applications. The elaboration of a new generation of sensors that will provide both a high spatial resolution for 2D Doppler imaging is presented, as well as a methodology that gives further information on the flowing particles concentration and/or dimensions. Then, a new embedded optical feedback interferometry imager for flowmetry has been realized using a 2-axis beamsteering mirror mounted on Micro-Electro-Mechanical Systems (MEMS) thus taking the full advantage of the compactness offered by the optical feedback interferometry sensing scheme. While previous works on optical feedback interferometry flowmetry have been limited to high particle densities fluids in single or multiple scattering regimes, we present also a sensing technique based on the optical feedback interferometry scheme in a laser diode that enables single particle detection at micro and nanoscales through the Doppler-Fizeau effect. Thanks to the proposed signal processing, this sensing technique can detect the presence of single spherical polystyrene micro/nanospheres seeded in watery suspensions, and measure their flow velocity, even when their diameter is below half the laser wavelength. It discriminates particle by their diameter up to a ratio of 5 between large and small ones while most of the technologies for particle characterization is bulk and requires manipulation of the fluid with small volume handling, precise flow and concentration control. Altogether, the results presented in this thesis realize a major improvement for the use of optical feedback interferometry in the chemical engineering or biomedical applications involving micro-scale flows.

Book excerpt: Microfluidics and lab-on-a-chip have, in recent years, come to the forefront in diagnostics and detection. At point-of-care, in the emergency room, and at the hospital bed or GP clinic, lab-on-a-chip offers the potential to rapidly detect time-critical and life-threatening diseases such as sepsis and bacterial meningitis. Furthermore, portable and user-friendly diagnostic platforms can enable disease diagnostics and detection in resource-poor settings where centralised laboratory facilities may not be available. At point-of-use, microfluidics and lab-on-chip can be applied in the field to rapidly identify plant pathogens, thus reducing the need for damaging broad spectrum pesticides while also reducing food losses. Microfluidics can also be applied to the continuous monitoring of water quality and can support policy-makers and protection agencies in protecting the environment. Perhaps most excitingly, microfluidics also offers the potential to enable entirely new diagnostic tests that cannot be implemented using conventional laboratory tools. Examples of microfluidics at the frontier of new medical diagnostic tests include early detection of cancers through circulating tumour cells (CTCs) and highly sensitive genetic tests using droplet-based digital PCR. This Special Issue on “Advances in Microfluidics Technology for Diagnostics and Detection” aims to gather outstanding research and to carry out comprehensive coverage of all aspects related to microfluidics in diagnostics and detection.
