Book excerpt: Non-thermal atmospheric pressure plasmas are increasingly attracting more attention from the scientific community for their potential use in surface treatment and biomedical applications. Even though considerable progress has been made, investigation of the species generated at close proximity (1 mm to ~100 nm) to a surface has not been addressed due to limitations of common optical techniques. A novel approach using Optical Emission Spectroscopy (OES) that can probe emissions over this distance away from the surface has been investigated. Additionally, the presence of high energy VUV photons was confirmed and these can play an important role in the propagation of plasma jets. By performing temporally resolved OES, it was determined that excitation of O, N, OH, NO and N2+ is predominantly due to collisions between He* and their precursors, whereas the N2 emissions are due to electron impact that produces electronically excited N2.

Non-invasive approaches of optical emission spectroscopy (OES), schlieren or shadowgraph, invasive methods of active laser spectroscopy including laser-induced fluorescence (LIF), laser or broadband absorption, cavity ring-down spectroscopy (CRDS), and laser scattering are illustrated. Basic plasma parameters of gas temperature, electron density and temperature, electric field strength, and reactive chemical gaseous species (O, H, N, OH, NO, O3, etc.) are able to be monitored. Comparisons and comments of these approaches are provided depending on diagnostic purposes.

Book excerpt: Non-thermal (cold) plasmas at atmospheric pressure have recently found many breakthrough applications in biology, medicine, and food security. Plasmas can efficiently kill bacteria, yeasts, moulds, spores, biofilms and other hazardous microorganisms, including potential bio-terrorism agents. They can be employed for bio-decontamination and sterilization of surfaces, medical instruments, water, air, food, even of living tissues without causing their damage. Direct or indirect plasma interaction with living cells of microorganisms or even humans enables novel bio-medical applications, e.g. treatment of skin diseases and ulcers. Plasma-enhanced blood coagulation coupled with its antiseptic properties proved success in wound healing and opens new possibilities in surgery, emergency medicine and military applications. Plasma treatment allows cell manipulations, their removal and targeted transfer into the injured area, which can accelerate wound healing. Plasma induced apoptosis (programmed cell death) of tumor cells brings forth a great potential for cancer treatment. Besides, plasma enables painless treatment of dental caries, root canal disinfection, and other dentistry applications. This book is a selection of reviewed manuscripts issuing from the NATO Advanced Research Workshop Plasma for bio-decontamination, medicine and food security held in Jasná, Slovakia, on 15-18 March 2011. Book excerpt: This thesis presents a novel, asymmetrical atmospheric pressure plasma jet, that uses air as its working gas, operating in the kilohertz range. The plasma system is designed to be mobile, easily set-up in many environments and has variable controllability, such as voltage, airflow and electrode position. The initial body of work presented in the thesis relates to the design and development of the atmospheric pressure plasma jet used throughout the bulk of the thesis. The key components of the atmospheric plasma jet are introduced. Electrical measurements of the plasma were also preformed and showed that the plasma operated electrically in two modes, the so called noisy and quiet modes. Optical emission spectroscopy was preformed on the plasma jet and showed that the emission spectrum of the jet was complex, containing emissions from many species, both molecular and atomic. These species included but were not limited to atomic species of nitrogen, oxygen and hydrogen and molecular emissions from N2, O2 and OH. In addition to this, it is possible to control the emission intensities from these species by varying the control parameters of the plasma jet such as voltage, airflow and electrode distance. The observed emission spectra were noticed to be lacking in any optical features due to emissions from nitric oxide, while it was believed to be the case that the plasma jet should produce nitric oxide. To test this hypothesis the experimental method laser induced fluorescence was chosen as a means to probe or nitric oxide in the plume region of the plasma. Experimentally it was found that nitric oxide was present within the plume. In addition by again varying the control parameters of the plasma jet the absolute number density of nitric oxide within the plume of the plasma could be varied. Finally, two-photon absorption laser induced fluorescence is used to probe for atomic oxygen in the plume region of the atmospheric pressure plasma jet. Two photon absorption differs from the single photon method in that two photons are simultaneously absorbed by and used to excite the species under investigation, opening up the possibility to probe species with excitation energies that are difficult to reach using conventional lasers systems. Like, the case for nitric oxide, atomic oxygen was confirmed to be present within the plume, although it was not possible to determine an absolute number density. Likewise, by varying the control parameters a change in the trend in the intensity of the fluorescence was observed.

The hydroxyl radical (OH) is of these species and has significant biological importance. An atmospheric pressure plasma jet (APPJ) was constructed in the form of a plasma pencil, and relative and absolute measurements were made of OH in both its first excited ground state—OH(A) and OH(X), respectively—using optical emission spectroscopy and cavity ring-down spectroscopy (CRDS). The total number of OH radicals were found to be constant in the plume and within the range given by relative measurements made on similar devices in the literature.
