Book excerpt: The data, the information, and even the overarching knowledge necessary for risk assessments of economically important environmental carcinogens come, for the most part, from the applied biological disciplines, e. g. , toxicology, epidemiology, biostatistics, etc. The more fundamental biological disciplines, e. g. , biochemistry, cell biology, molecular biology, molecular genetics of cancer, etc. , have enormous but unrealized potential to improve current cancer risk assessment methods. The objective of this advanced research workshop ARW was to advance the state of the art of cancer risk assessment methods by identifying potential short and long term contributions to such methods from the more fundamental disciplines. Attention was paid to short and long term contributions from research advances in the biochemistry and physiology of oncogenes (oncogenes research) and in the construction and utilization of transgenic animals (transgenics research). In the last 20 years, researchers in the fundamental biological disciplines, i. e. , biochemists, geneticists, molecular and cell biologists, etc. , have, inter alia, advanced spectacularly our understanding of the nature of neoplastic diseases. Their phenomenal progress is the combined result of both advances and refinements of the techniques available to them and of new fundamental discoveries. Among the latter the most significant are the discoveries of oncogenes and of the feasibility of creating transgenic animals, i. e. , of transferring well defined and expressible genes from the cells of one species of organisms to the embryonic cells of another.

It is only in the past decade, however, that molecular genetics and epidemiology have combined to define the role of inheritance in cancer more clearly, and to identify some of the genes involved. The causative genes can be tracked through cancer-prone families via genetic linkage and positional cloning. Several of the genes discovered have subsequently been proved to play critical roles in normal growth and development. There are also implications for the families themselves in terms of genetic testing with its attendant dilemmas, if it is not clear that useful action will result. The chapters in The Genetics of Cancer illustrate what has already been achieved and take a critical look at the future directions of this research and its potential clinical applications.

Book excerpt: The NATO Advanced Research Workshop entitled "The Photosynthetic Bacterial Reaction Center: Structure, Spectroscopy, and Dynamics" was held May 10-15, 1992, in the Maison d'H6tes of the Centre d'Etudes Nuc1eaires de Cadarache near Aix-en-Provence in the south of France. This workshop is the most recent of a string of meetings which started in Feldafing (Germany) in March 1985, soon after the three-dimensional structure of the bacterial reaction center had been elucidated by X-ray crystallography. This was followed, in September 1987, by a workshop in Cadarache and, in March 1990, by a second meeting in Feldafing. Although one of the most important processes on Earth, photosynthesis is still poorly understood. Stimulated by the breakthrough of solving the bacterial reaction center structure at atomic resolution, the field of relating this structure to the function of the reaction center, i. e. the remarkably efficient conversion and storage of solar energy, has been developing vigorously. Once the general organization of the cofactors and some details of the protein-cofactor interactions were known, it became possible to combine a variety of spectroscopic techniques with the powerful tool of site-directed mutagenesis in order to address increasingly incisive questions about the specific role of some amino acid residues in the electron transfer process. Still another promising tool is being developed, namely the exchange of a number of the native bacteriochlorophyll and bacteriopheophytin cofactors by chemically modified pigments.

Such interactions brought together the exchanges of ideas amongst participants and helped clarify the intricate details and formulate new vistas and collaborations. Since the study of nervous system development has focused mostly on the origin of neuron and glia cells, the area of current research was represented by talks on early cellular events including effects of growth factors, BOX and other gene expressions and cell lineage of specific cell type(s). Formation of specific cell types and the specific neuronal connections have been a major theme in the study of the nervous system development. Recent technical advances has resulted in new information at both cellular and molecular levels which have provided new details. Current research was represented by "selective" topics discussed at the meeting.

Book excerpt: Advances of cardiovascular engineering prompt one to consider innovative device technology - that is, the development of new replacement heart valves or engineering of a totally implantable energy source for an artificial heart. However, these kinds of advances have often proved unable to achieve a long-lasting benefit as the cardiovascular field has matured so fast. Cardiovascular engineering has matured to the point where a major innovation must not only function, but must continuously function better than existing devices. This is difficult to accomplish in the complex cardiovasculature system, in which energy source, biocompatibility, compliance, and functionality all must be considered. The maturation of the field is evident from the fact that many engineered prosthetic systems perform well - for example, heart valves function for long periods of time, large-vessel vascular grafts are quite adequate, extracorporeal membrane oxygenation has significantly prolonged the feasible length of heart bypass and other surgical operations, and total artificial hearts can be used as a bridge to transplant without serious complications, yet none of these systems is as good as the natural ones it replaces. The reasons for this are many and incompletely understood. The next stage of progress must be better to alterations understandings of the various components of vasculature and their response by our devices, be they at the micro- or macro-circulatory levels, in the blood, or associated with the vascular wall.

Advanced Study Institute (ASI) of the same title, held at Bishop's University, Lennoxville, Quebec, Canada in August 1991. All the major presentations had been commissioned, so to speak, during the organisational phase. This was done with the view of not only having a structured ASI, whIch is expected to be a high-level tutorial activity, but also the ensuing volume. As will be indicated in the general introduction, there have been a couple of meetings and publications dealing with rhythms in fishes in the past twenty years. However, as in other disciplines, there has been substantial progress in this field also. Further, I wished to cover almost all aspects and come out with a volme which will be as complete as possible. Of course, this failed to materialise. In spite of starting the arrangements two years before the event, several lecturers were not able to attend due to a number of reasons. Some could be replaced while others could not Taking into account their individual specialties, I asked about twenty active workers in the field to provide provoking overviews, not simply reviews of their own work. In some cases estimates of risk under conditions prevailing in the original data are of primary interest; in others predictions of risk under other conditions are required. Estimates of risk may be based on empirical models that provide a reasonable description of the available data or on models developed on the basis of plausible assumptions about the mechanisms of carcinogenesis. Throughout this volume established scientific principles of carcinogenesis are used to support methods proposed for the quantitative estimation and prediction of risk.
