Book excerpt: We are pleased to present to our readers the Proceedings of the International Symposium "Proteases: Potential Role in Health and Disease" which was held in WUrzburg (FRG) during October 17-20, 1982. The topics discussed included those dealing with the physi ology and pathophysiology of proteases and their inhibitors, the interactions of proteases and hormones, the kallikrein-kinin system, complement and the coagulation system, the function of proteases in the kidney and the intestinal tract as well as the role of proteases in lung diseases, pancreatitis, arthritis and hypercatabolic states (multiple trauma, septicemia, acute renal failure). The papers presented answered many questions, but raised many more concerning the significance of proteases and their inhibitors in clinical medicine. It was unfortunately impossible in this volume to in clude the extended, lively and extremely stimulating discussions which were enjoyed by the participants during the conference. The meeting has provided a unique framework for close inter action between scientists from various disciplines, including bio chemistry, physiology, surgery, anaesthesiology, endocrinology, hematology, pulmonology and nephrology. We would like to express our thanks and appreciation for all those who have stimulated, encouraged and supported us to hold this symposium in WUrzburg. Book excerpt: There was a time, not all that long ago, when scientific study of the cell was called cytology, and the workers in the field named themselves cytologists. When I was a medical student, lectures in cytology were a special, segregated part of the curriculum in the histology course, given along with general anatomy, and they were, as I recall, the surest of cures for insomnia. I still possess Cowdry's three-volume set entitled Special Cytology, published in 1934, and leafing through these books today is rather like examining a medieval manuscript. You could never have guessed what was going to happen to the field. At that time it was all structure, and all guesswork about the structure. When cells were packed together in various tissues, how did the geometry of packing work? How many sides did a liver cell have, in real life? What on earth were all those granules in side, and what were the best stains for looking at them? One thing about those granules, they never moved. Indeed, nothing moved. Cytology turned into cell biology much later on, and suddenly came alive. As has been the case in so many diSciplines in biology, it was brought to life by techniques. New instruments and cytochemical methods were devised for look ing at cells, manipulating cells, more or less in vivo.

This publication provides an integrated picture of the latest information on the kallikrein kinin system. It contains contributions from morphologists, geneticists, biochemists, pharmacologists, physiologists, and clinicians. The Fifth International Kinin Congress (Nov. 29-Dec. 3, 1987) provided a forum for the exchange of information and ideas on the kallikrein-kinin system. The participation of more than 350 scientists from 23 countries reflects the widespread interest and international scope of research activity in the physiological and pathological functions of the kallikrein-kinin system. A total of 275 papers including posters were presented, attesting to the unquestioned success of this Congress. These proceedings, in two volumes, contain the collective studies presented, studies of high scientific standard that provoked stimulating and fruitful discussions. Also included in these volumes are the two plenary lectures presented by Dr. H.A. Margolius (USA) and Dr. S. Nakanishi (Japan). During the last decade our knowledge of the role of the kallikrein kinin system in health and disease has been greatly advanced by the develop ment of antagonists to bradykinin and the introduction to clinical practice of converting enzyme inhibitors. Symposia on converting enzyme and on recent advances in research on the kallikrein-kinin system are included in the proceedings.

Book excerpt: When phagocytes are exposed to a number of different stimuli, they undergo dra matic changes in the way they process oxygen. Oxygen uptake increases markedly, frequently more than 50-fold; the phagocytes begin to produce large quantities of superoxide and hydrogen peroxide; and they immediately begin to metabolize large amounts of glucose by way of the hexose monophosphate shunt. This series of changes has become known as the respiratory burst. It was first believed that the major function of this respiratory burst was to generate powerful antibacterial agents by the partial reduction of oxygen. It is becoming apparent that the respiratory burst has much wider application, and its physiological function in many different biolog ical areas is clear. In this volume, we have attempted to bring together the work of experts who have published extensively on the involvement of the respiratory burst in different physiological functions. In the first three chapters, Dr. Borregaard and Dr. Berton and co-workers and Dr. Roos and co-workers bring together what is known about the respiratory burst. They present up-to-date versions of the biochemical and metabolic activities associ ated with the burst. In Chapter 4, Dr. Styrt and Dr. Klempner discuss the respiratory burst as it affects cellular ion homeostasis. The rapid progress which has been made and the results achieved in intensive care medicine are based on sound basic research, which is duly reflected in these chapters. Multiorgan failure is the foremost cause of postoperative and posttraumatic death and many complex mechanisms are involved. Only with a good foundation of basic research can abnormalities in the physiological, biochemical, and morphological course of shock be recognized and the necessary conclusions for treatment drawn. Therapy must proceed from profound knowledge of the multi variant physiological events in order to influence shock, sepsis and organ failure. Although numerous possibilities for therapy have arisen from pharmaceutical research in recent years, they are beyond the scope of this book and are not discussed here. To gain a better understanding of the pathophysiological events it was necessary to examine and to describe different models that simulate and reproduce these events. Here we describe the causative agents (shock) and the consequences (sepsis, organ failure) in two main sections, divided on the basis of their pathophysiology.

The Foundation has even been abl~ to beat the inflation and the prizes have steadily increased during the last 20 years. It might interest you to hear that this year's prizes will amount to 681. 000 Sw. crowns each. Due to the favourable financial development and also due to addi tional donations the Nobel Foundation decided to extend its inte rests by promoting the arrangements of symposia, lectures and other international scientific activities.

Book excerpt: Enzymes perform the executive role in growth, energy conversion, and repair of a living organism. Their activity is adjusted to their en vironment within the cell, being turned off, switched on, or finely tuned by specific metabolites according to demands at the physiologi cal level. Each enzyme discovered in the long history of enzymology has revealed its own individuality. Even closely related members of a family differ in specificity, stability or regulatory properties. Despite these, at first sight overwhelming aspects of individuality, common factors of enzymic reactions have been recognized. Enzymes are stereospecific catalysts even when a nonspecific process would yield the same product. Knowledge of the detailed stereochemistry of an enzymic reaction helps to deduce reaction mechanisms and to ob tain insight into the specific binding of substrates at the active site. This binding close to catalytically competent groups is related to the enormous speed of enzyme-catalyzed reactions. The physical ba sis of rate-enhancement is understood in principle and further exploit ed in the design of small organic receptor molecules as model enzymes. These aspects of enzyme catalysis are discussed in Session 1. Session 2 emphasizes the dynamic aspects of enzyme substrate inter action. Substrate must diffuse from solution space to the enzyme's surface. This process is influenced and can be greatly facilitated by certain electrostatic propterties of enzymes. The dynamic events during catalysis are studied by relaxation kinetics or NMR techniques.
