Book excerpt: nd This book stems from the 2 Parkinson's Disease Symposium on "Neurotoxic Factors in Parkinsons disease and related disorders", held on August 6-7,1999 at the University ofUlm Medical School in Ulm at the Danube, Germany. The symposium wasalso asatellite ofthejointmeetingofthe InternationalSocietyfor Neurochemistry (ISN) and the European Society for Neurochemistry (ESN) that followed immediately afterwards in Berlin. The specific topic, neurotoxic factors in Parkinson's disease (PD), was chosen in light of accumulating neurobiological and epidemiological evidence indicating that the causes of this second most common neurodegenerative disorder, and possibly of related conditions such as multiple systems atrophy, in some manner involve environ mental (xenobiotic) and even endogenous toxic agents. This volume brings together much of that neurobiological evidence; and in epidemiology, several recent major studies ofPD - for example, a study of20,000 twins published in the Journal of the American Medical Association in 1999 - conclude that environmental and life style than genetics, appear to be critical in the idiopathic disease. However, factors, rather the precise roles of neurotoxic factors in the pathogenesis of PD and related basal ganglia disorders remain uncertain, despite the cascade of research resulting from the discovery of the prototype MPTP in the early 1980's, and frankly, we have been surprised by the paucity of concentrated attention on specific environmental agents other than MPTP.

Book excerpt: The specific topic, “Isoquinolines And Beta-Carbolines As Neurotoxins And Neuroprotectants – New Perspectives In Parkinson`S Disease Therapy,” was chosen in light of accumulating neurobiological evidence indicating that, in addition to exogenous neurotoxins (e.g., 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine [MPTP]), endogenous compounds may play an important role in the most common neurodegenerative disorders (e.g., Parkinson`s disease). Two groups of amine related compounds, which appeared chemically like MPTP, were detected in human brain and cerebrospinal fluid (CSF): ß-carbolines (BCs) and tetrahydroisoquinolines (TIQs). These are heterocyclic compounds formed endogenously from phenylalanine/tyrosine (TIQs) and tryptophan, tryptamine, and 5-hydroxytryptamine (BCs), respectively and exert a wide spectrum of psychopharmacological and behavioral effects. The TIQs and BCs may bind to their own high affinity sites on neuronal membranes associated with or located close to the receptors of neurotransmitters. Research on TIQs and BCs is stimulated also by their possible role in pathological conditions, especially parkinsonism and alcoholism. Recently, clinical interest has been spurred by their role as neuroprotective, and even neurorestorative, anticonvulsant and antiaddictive, substances. In this book we are going to summarize, for the first time, the results from behavioral, neurochemical and molecular experiments, which demonstrate a wide spectrum of TIQs and BCs effects - from their rather mild neurotoxic actions to the important neuroprotective and antiaddictive properties. Additionally, the recent results of experimental studies in vivo have allowed a much better understanding and simultaneous comparison of the neurochemical and molecular mechanisms underlying the neuroprotective and neurotoxic actions of endogenous TIQs and BCs and have pointed to the possibility of their therapeutic applications in neurodegenerative diseases such as Parkinson's disease.

Book excerpt: This is a program of epidemiologic and neuropsthologic research aimed toward determining neurotoxic and preventive/ ameliorative risk factors for Parkinson's disease (PD), parkinsonism, and other neurodegenerative conditions using existing data and autopsied brain tissue from the Honolulu Heart Program I Honolulu-Asia Aging Study. Major findings from the four years of this include: a) a significant association of coffee and caffeine consumption with reduced risk of PD, b) an association of years Worked on a plantation and occupation in sugar mill processing with increased risk of PD, c) an association of constipation with development of future PD, d) a significant association of increased triceps skinfold thickness measured in mid-life with future development of Parkinson's disease, e) an association of slow reaction time with presence of incidental Lewy bodies found in the subsantia nigra or locus ceruleus at autopsy, f) an association of elevated GFAP in frozen brain tissue with lower cognitive. function, and g) an association of prolonged QT interval with future risk of PD project. A genetics component found no association of CYP2D6 gene, the-CYP1A2 gene, the dopamine transporter gene, the parkin gene; and the adenosine receptor gene with PD. Organochlorine levels from two areas of brain from each of 15-research subjects with the highest exposure to pesticides are complete and indicate measurable amounts of these compounds in most brains. Request has been made for supplement fluids to perform organochlorine levels on all autopsied brains.
