Book excerpt: The elucidation of the cellular and molecular bases underlying the inte grated function of the central nervous system, both in disease and in health, must ultimately come from the combined efforts of scientists from many disciplines, including biology, chemistry, histology, pathology, physiology, pharmacology, and psychology. Communication between scientists from these various disciplines-vital to the advancement of our understanding of the function of the nervous system-has become more and more difficult in recent years. Both increasing specialization and the incredible increases in publications pertinent to brain research in a wide spectrum of journals, in symposium volumes, in monographs, in abstracts, and in reviews contrib ute to the problems of cross-communication and even of communication within a scientific discipline. Research on the significance of cyclic nucleo tides to the function of nervous systems is particularly illustrative of the communication problem. Since the initial publications by Sutherland, Rall, and Butcher in the late fifties and early sixties on high levels of adenylate cyclase, phosphodiesterases, and cyclic AMP in brain, the ensuing litera ture of this field has expanded exponentially. At the present time, from five to ten publications relevant to cyclic nucleotides and the nervous system appear each week. Indeed, these are minimal numbers based mainly on examination of literature titles and key index words. Many articles concerned with some aspect of central function contain, buried within their text, experiments with or related to cyclic nucleotides.

The were chosen to emphasize areas in which there has been substantial topics progress in the past 2-3 years and fall into seven major groups dealingwith: cholinergiC receptors; chemistry, histochemistry and enzymology; cyclic nucleotides and cholinergiC mechanisms; storage, compartmentation and release of acetylcholine; regulatory mechanisms in acetylcholine metab olism; modulation of acetylcholine metabolism; and behavioral and clin ical manifestations of cholinergiC function and dysfunction. Each group contains one or more reviews and a number of shorter contributions de scribing current work. This symposium was the third in a series of which the first two were held in Skokloster, Sweden in 1970 and Boldern, Switzerland in 1974. The Proceedings of the meetings indicate a rapid development of knowledge of cholinergiC mechanisms which for many years lagged behind that of other neurotransmitters and neuroregulators. The inclusion of a large section in the present volume dealing with clinical manifestations of cholinergiC dysfunction reflects one of the most important trends in current research on cholinergiC mechanisms, namely the close inter-relationship and mutual support of basic science and clinical investigation. The meeting is related to two previous meetings on metabolic compart mentation in the brain. The first of these meetings took place at Rockefeller Foundation, Bellagio, Italy, July 11-16, 1971 and the proceedings, Metabolic Compartmentation in Brain, were edited by R. Balazs and J. E. Cremer and published by Macmillan in 1973. The second meeting was an Advanced Study Institute on Metabolic Com partmentation and Neurotransmission Relation to Brain Structure and Function, which was held in Oxford, September 1-8, 1974. The proceedings were edited by S. Berl, D. D. Clarke and D. Schneider and published as Volume 6 of the NATO ASI Life Science series by Plenum Press. The object of the present meeting was to review and discuss the present status of amino acids as chemical transmitters. Several issues such as electrophysiological response, localization, synthe sis, release and receptor binding of transmitter candidates were discussed. The possible morphological correlates to these func tions were also reviewed. During the meeting 24 leading papers were given. In addition, several of the participants presented important new findings during the discussion. Some of these have been included as short reports. The main financial support was obtained from NATO, Scientific Affairs Division.

First: As far as we know, already primitive societies had to cope with environmental toxins of many kinds and set up regulations to limit their effects on food and drug use. Modem science, synthesizing tens of millions of new compounds has incredibly magnified this challenge. Today, xenobiotic metabolism has become a crucial task for humans and many other species alike. Second: When reading this book, one is impressed by the extraordinary speed at which neurotoxicology has advanced. Obviously, processing (and endogenous formation) oftox ins has become an extremely relevant topic. When I had the chance, almost three decades ago, to work in chemical pharmacology with Bernard B. Brodie at NIH, the drug metabo lizing system of the liver had just been recognized and characterized. We had just started to work on the biogenic amines, newly discovered cyclic nucleotides in rat brain, human cere brospinal fluid, and on the effects of toxic drugs like amphetamines. Today, biochemical neuropharmacology is a mature field of neuroscience.
