Book excerpt: Organisms are often motivated by goals which are spatially and temporally distant. Animals travel large distances, risking their lives in search of food, water, or a mate. How does the animal remain motivated and focused on the goal? How does the animal change its behavior adaptively in varying environmental conditions? Here, we reveal a role for two major neuromodulators in different behavioral contexts - dopamine when animals seek distant rewards and serotonin when animals behave adaptively in environments with different valence. We measured population calcium activity of dopamine neurons in ventral tegmental area as mice learned a variety of reward seeking tasks. When mice navigated through space toward a goal, dopamine activity progressively increased or ramped upand peaked at the goal. This ramping dopamine activity was sensitive to reward magnitude and emerged quickly within a few trials after the first reward exposure in naïve animals. Physical action was not necessary to produce dopamine ramps; dynamic sensory feedback of progress to reward alone can produce dopamine ramps. However, dopamine ramps driven by sensory feedback were short lived. When the animal was required to use an internal model of distance to reward, in the absence of sensory feedback, the dopamine ramps were robust and did not fade over training. Collectively, these results suggest that dopamine ramps reflect an internal model of goal proximity information. Next, we measured population activity of serotonin neurons in the dorsal raphe nucleus when animals were in a moderate threat or a high threat environment. We found that the serotonin neural activity decreased during movement initiation in a moderate threat environment but increased during movement initiation in a high threat environment. We then measured calcium activity of individual serotonin neurons when animals were exposed to environments at differentthreat levels. We found that individual serotonin neurons switched their movement-related neural activity when mice transitioned from moderate threat to high threat environments, signifying routing of environmental valence information to individual serotonin neurons. Together, this research provides insights into the neural mechanisms underlying environmental valence-dependent adaptive behaviors and sustained goal directed behaviors.

But in addition to asking what attention actually is, decomposing and analyzing its varieties, or delimiting its neurobiological mechanisms and effects, in this volume we want to explore attention somewhat differently. We believe that a full-fledged theory of attention must consider its workings in the context of motivated, goal-directed, and environmentally constrained organisms. That attention is related to goal-directed behavior is not news. What the contri- tions to this volume do suggest, however, is the existence of fundamental links between attention and two key processes that are crucial for adapted conduct: go- directed behavior and cognitive control. Importantly, they show that these relations can be explored at multiple levels, including neurodynamical, neurochemical, evo- tionary, and clinical aspects, and that in doing so multiple methodological challenges arise that are worth considering and pursuing. The reader will find here, therefore, a selection of contributions that range from basic mechanisms of attention at the n- ronal level to developmental aspects of cognitive control and its impairments. Another trend that will become evident is that, in different ways, the authors stress the need to understand these issues as they unfold in natural behavior (both healthy and pathological), thus arguing for a more ecological approach to these questions.

When this fundamental need is not fulfilled, people place greater emphasis on forming social bonds and are motivated to behave in ways consistent with accomplishing these social goals. The present thesis leverages knowledge of the specific cognitive and neural mechanisms involved in the processing of nonsocial rewards and related motivational behaviors to examine how social interactions may similarly be conceptualized as a class of rewards responsible for shaping instrumental learning and evoking approach behaviors. In nonsocial domains, the rewarding properties of stimuli are crucially involved in the development and implementation of goal-directed behaviors, and function to guide animals towards resources that can satisfy unmet needs. Previous research has identified a specific set of brain regions (i.e., the mesolimbic dopamine circuit including the ventral striatum) that are responsible for coding information about rewards and facilitating learned associations between cues and subsequent rewards. The experiments described here provide evidence that 1) the same neural mechanism that facilitates associative learning and reward detection in nonsocial domains similarly functions in the social domain, 2) activity in this neural system tracks the motivational relevance of social stimuli, and 3) these motivational influences bias the perception and interpretation of social cues. Taken together, these results suggest that the neural reward system is highly attuned to social context and functions to advantageously support social relations by translating the value of social cues into motivated action.

Moreover, the ability to integrate diverse incentives to modulate goal pursuit is essential for healthy cognitive function. A potential mechanism of motivational influence may be via cognitive control, the set of processes that coordinate and regulate cognition and action based on currently maintained goals [3,4]. However, it is currently unknown whether and how different types of incentives are combined in the brain to modulate cognitive control, and how this putative integrated value signal influences goal-directed behavior. In the current functional magnetic resonance imaging (fMRI) study, we utilized an innovative incentive integration task paradigm that establishes dissociable and additive effects of liquid (e.g., juice, neutral, saltwater) and monetary incentives on cognitive task performance, and applied innovative fMRI analysis approaches to elucidate the neural mechanisms that underlie the interaction between motivational and cognitive control process. First, we applied univariate parcel-based approaches to test whether a priori regions of interest (e.g., ventromedial prefrontal cortex, striatum, dorsal anterior cingulate cortex) represented the integrated value of the 'bundled incentives', and whether these regions were also associated with variability in cognitive task performance (Aim 1). Second, we applied representational similarity analysis - an innovative multivariate approach - to test whether and how the combined values from diverse motivational incentives are represented in the similarity of neural patterns in fMRI BOLD activity (Aim 2). Moreover, we aimed to examine whether such multivariate approaches were more sensitive to motivational incentive effects compared to univariate approaches, or alternatively provided complementary information to the univariate results in motivational incentive effects. This is the first study, to our knowledge, that investigates the neural mechanisms underlying whether and how value integration of primary/consummatory and secondary/abstract incentives in a cognitive control context guide goal-directed behavior. Importantly, these results provide critical knowledge into the basic neural mechanisms underlying interactions between motivational incentive integration and cognitive control, which can inform subsequent hypotheses about neuromodulatory influences (e.g., dopamine) in such interactions, as well as inform key predictions about targeted neural mechanisms in age-related changes in motivation-cognition interactions as well as maladaptive motivational processes in psychopathology (e.g., depression, addiction).

In the first chapter, by Deepak Pandya and Edward Yeterian, the general architectural features and connections of cortical associ ation areas are considered; as these authors point out, in primates the association areas take up a considerable portion of the total cortical surface. Indeed, it is the development of the association areas that accounts for the greatest differ ences between the brains of primate and non primate species, and these areas have long been viewed as crucial in the formation of higher cognitive and be havioral functions. In the following chapter, Irving Diamond, David Fitzpatrick, and James Sprague consider the question of whether the functions of the as sociation areas depend on projections from the sensory areas of the cortex. They use the visual cortex to examine this question and show that there is a great deal of difference between species in the amount of dependence, the differences being paralleled by variations in the manner in which the geniculate and pulvinar nuclei of the thalamus project to the striate and extra striate cortical areas. One of the more interesting and perhaps least understood of the association areas is the cingulate cortex, discussed by Brent Vogt. Cingulate cortex has been linked with emotion and with affective responses to pain, and in his chapter Vogt gives an account of its cytoarchitecture, connections, and functions.

Book excerpt: Learning to direct behaviors towards goals is a central function of all vertebrate nervous systems. Initial learning often involves an exploratory phase, in which actions are flexible and highly variable. With repeated successful experience, behaviors may be guided by cues in the environment that reliably predict the desired outcome, and eventually behaviors can be executed as crystallized action sequences, or "habits", which are relatively inflexible. Parallel circuits through the basal ganglia and their inputs from midbrain dopamine neurons are believed to make critical contributions to these phases of learning and behavioral execution. To explore the neural mechanisms underlying goal-directed learning and behavior, I have employed electrophysiological and electrochemical techniques to measure neural activity and dopamine release in networks of the striatum, the principle input nucleus of the basal ganglia as rats learned to pursue rewards in mazes. The electrophysiological recordings revealed training dependent dynamics in striatum local field potentials and coordinated neural firing that may differentially support both network rigidity and flexibility during pursuit of goals. Electrochemical measurements of real-time dopamine signaling during maze running revealed prolonged signaling changes that may contribute to motivating or guiding behavior. Pathological over or under-expression of these network states may contribute to symptoms experienced in a range of basal ganglia disorders, from Parkinson's disease to drug addiction.
