Book excerpt: DNA Repair Mechanisms is an account of the proceedings at a major international conference on DNA Repair Mechanisms held at Keystone, Colorado on February 1978. The conference discusses through plenary sessions the overall standpoint of DNA repair. The papers presented and other important documents, such as short summaries by the workshop session conveners, comprise this book. The compilation describes the opposing views, those that agree and dispute about certain topic areas. This book, divided into 15 parts, is arranged according to the proceedings in the conference. The plenary sessions are grouped with the related workshop and poster manuscripts. The first two parts generally tackle repair in terms of its identification and quantification, as well as the models, systems, and perspectives it utilizes. The following parts discuss the various types of repair including base excision, nucleotide excision repair in bacteria, excision repair in mammalian cells, inducible/error-prone repair in prokaryotes, and strand break repair in mammalian cells among others. This reference material looks into the replicative bypass mechanisms in mammalian cells, viral probes, and hereditary repair defects. It explains repair deficiency and human disease, as well as mutagenesis and carcinogenesis. The last part of this book deals with the consequences and effects of DNA repair. This volume is a helpful source of reference for students, teachers, scientists, and researchers in the different fields of genetics, radiology, biochemistry, and environmental biology.

Available in PDF, EPUB and Kindle. Book excerpt: This detailed volume provides a comprehensive set of experimental protocols and useful strategies to examine the repair of damaged bases via the Base Excision Repair (BER) pathway in vitro and in cells. Beginning with multiple molecular and cellular techniques to examine the excision of damaged bases from double-stranded DNA or DNA wrapped in a nucleosome, the book continues with sections covering procedures to detect and quantify the damaged bases, protein DNA crosslinks, and double-strand breaks, experimental procedures to identify DNA repair protein interactome by conventional tandem affinity purification followed by mass spectroscopy analysis, as well as the analysis of genome-wide binding of DNA repair proteins and copy number variations of the DNA damage response gene in tumors. Written for the highly successful Methods in Molecular Biology series, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step and readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Base Excision Repair Pathway: Methods and Protocols serves as a valuable resource for novices and experts trying to examine the repair of various types of DNA lesions in vitro and in cell by the distinct set of proteins in the BER pathway.

Book Molecular Mechanisms for Repair of DNA

Download or read book Molecular Mechanisms for Repair of DNA written by Philip Hanawalt and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 433 pages. Available in PDF, EPUB and Kindle. Book excerpt: An "age" has passed in the 40 years since we first observed recovery from radiation damage in irradiated bacteria. During the early 1930s, we had been discussing the possibility of rapid changes after radiation exposure with Farring ton Daniels, Benjamin Duggar, John Curtis, and others at the University of Wisconsin. After working with living cells, we had concluded that organisms receiving massive insults must have a wide variety of repair mechanisms available for restoration of at least some of the essential properties of the cell. The problem was how to fmd and identify these recovery phenomena. At that time I was working on a problem considered to be of great importance-the existence of the so-called mitogenetic rays. Several hundred articles and a score of books had already appeared dealing with mitogenetic rays, a type of radiation that was thought to exist in the shorter ultraviolet region. Our search for mitogenetic rays necessitated the design of experiments of greatest sensitivity for the detection of ultraviolet. It was vital that conditions be kept as constant as possible during exposure. All the work was done at icewater temperature (3-5°C) during and after exposure. We knew that light was an important factor for cell recovery, so all our experiments were done in dim light, with the plated-out cells being covered with dark cloth. Our statements on the effect of visible light stimulated Kelner to search for "photoreactivation' (as it was later called).

Book DNA Replication Recombination and Repair

Download or read book DNA Replication Recombination and Repair written by Fumio Hanaoka and published by Springer. This book was released on 2016-01-22 with total page 548 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book is a comprehensive review of the detailed molecular mechanisms of and functional crosstalk among the replication, recombination, and repair of DNA (collectively called the "3Rs") and the related processes, with special consciousness of their biological and clinical consequences. The 3Rs are fundamental molecular mechanisms for organisms to maintain and sometimes intentionally alter genetic information. DNA replication, recombination, and repair, individually, have been important subjects of molecular biology since its emergence, but we have recently become aware that the 3Rs are actually much more intimately related to one another than we used to realize. Furthermore, the 3R research fields have been growing even more interdisciplinary, with better understanding of molecular mechanisms underlying other important processes, such as chromosome structures and functions, cell cycle and checkpoints, transcriptional and epigenetic regulation, and so on. This book comprises 7 parts and 21 chapters: Part 1 (Chapters 1–3), DNA Replication; Part 2 (Chapters 4–6), DNA Recombination; Part 3 (Chapters 7–9), DNA Repair; Part 4 (Chapters 10–13), Genome Instability and Mutagenesis; Part 5 (Chapters 14–15), Chromosome Dynamics and Functions; Part 6 (Chapters 16–18), Cell Cycle and Checkpoints; Part 7 (Chapters 19–21), Interplay with Transcription and Epigenetic Regulation. This volume should attract the great interest of graduate students, postdoctoral fellows, and senior scientists in broad research fields of basic molecular biology, not only the core 3Rs, but also the various related fields (chromosome, cell cycle, transcription, epigenetics, and similar areas). Additionally, researchers in neurological sciences, developmental biology, immunology, evolutionary biology, and many other fields will find this book valuable.

Available in PDF, EPUB and Kindle. Book excerpt: The First International Congress on DNA Damage and Repair was held in Rome, Italy, July 12-17, 1987. It was organized by the Italian Com mission for Nuclear Alternative Energy Sources. The subject of DNA damage and repair involves almost all the fields ofbidogical sciences. Some of the more prominent ones include carcino genesis, photobiology, radiation biology, aging, enzymology, genetics, and molecular biology. These individual fields have their own interna tional meetings and although the meetings often have sessions devoted to DNA repair, they do not bring together a wide diversity of international workers in the field to exchange ideas. The purpose of the Congress was to facilitate such an exchange among scientists representing many fields of endeavor and many countries. The 37 manuscripts in this volume, presented by the invited spea kers during the four and half days of the Congress, encompass the field of DNA damage and repair. They cover biological systems ranging from mo lecules to humans and deal with damages and repair after treatment of cells with various types of radiations, chemicals, and exogenous and en dogenous oxidative damages. The Congress and its Proceedings are dedicated to two international leaders in the field of DNA damage and repair, Alexander Hollaender of the United States and Adriano Buzzati Traverso of Italy. Hollaender, who died in December 1986, was one of the first investigators to recognize the damage to DNA was important in cell killing and mutagenesis. His early work indicated that cells could recover from radiation injury.

Available in PDF, EPUB and Kindle. Book excerpt: Ribonucleic acid (RNA) is a macromolecule that plays a central role in cell physiology: RNA molecules act as intermediates between the deoxyribonucleic acid (DNA), where genetic information is stored, and proteins, which perform the necessary functions within the cell. Traditionally, the structural and functional properties of RNA are closely linked to gene expression. However, RNA-based enzymes, called ribozymes, are also involved in catalysis and small RNAs regulate key cellular processes, such as cell growth, division, differentiation, aging and death. RNA is a sensitive macromolecule that can be easily damaged by environmental conditions (ultraviolet radiation, oxidative stress) and biological factors (ribonucleases, ribotoxins, CRISPR-Cas systems). Therefore, cells have developed mechanisms to protect and/or repair RNA molecules. This book presents an overview of the biology of RNA damage, protection and repair in prokaryotes and eukaryotes. Individual chapters cover the expression regulation, enzymology and physiological role of such systems, and link them to important human diseases such as cancer and degenerative diseases.
