Book excerpt: Thyroid hormone receptors (TRs) are hormone-regulated transcription factors and part of a superfamily of nuclear receptors (NRs) that mediate a diverse array of biological activities. TRs are key regulators of metabolism, homeostasis, and reproduction, and can serve as tumor suppressors. NR insult (i.e. lesions or chromosomal alterations) can upset normal receptor activity and trigger a gamut of physiological maladies. V-Erb A, a highly mutated viral version of TR that contributes to the development of leukemias and sarcomas in birds, can cause hepatocellular carcinoma when ubiquitously expressed in mice. TR mutations have been identified at a high frequency in hepatocellular carcinoma (HCC) and renal clear cell carcinoma (RCCC), and have also been associated with thyroid and gastric cancers, but their role in carcinogenesis is complicated and not well-defined. I examined the functional and molecular characteristics of a panel of RCCC TR mutants, and analyzed the expression profiles of two different TR mutants from HCC and from RCCC to better understand their contribution to oncogenesis. RCCC-associated mutants behaved as dominant negative inhibitors of wild-type receptor activity (similar to their HCC mutant counterparts), and some gained the ability to interact with different corepressor partners. RCCC TR mutants also regulated a set of genes unique from those targeted by the wild-type or HCC-associated receptors; many of these genes have been previously implicated in RCCC. All together our results indicate that cancer-related TR mutants behave similarly, though not identically, and may point to a tissue-specific role for TR mutants in the occurrence or development of different types of cancer.
