The objectives of this study were to develop new or improve existing analytical methods for the quality evaluation of virgin olive oil, which can be used as alternative or assist sensory tests. A new standard for olive oil quality has been proposed that relates the ratio of 1,2-diacylglycerol (DAG) to 1,3-DAG to sensory evaluations of olive oil. The 1,2-DAG % test was found to have the highest correlation to negative sensory results of commercial extra virgin olive oil. However, DAGs and their isomers are difficult to quantitate and characterize by Flame Ionization Gas Chromatography (GC-FID) due to the lack of suitable standards. In this study, a GC Electron Ionization Mass Spectrometry (GC-EI-MS) method was developed to determine the fatty acid composition and molecular structure of trimethylsilyl (TMS) derivatized DAGs present in edible oils. Moreover, a number of reaction mechanisms were proposed to explain the formation of the most abundant mass fragments of DAGs and their isomers. Volatile compounds are responsible for both positive and defective aromas of virgin olive oils. The volatile analysis should directly reflect the sensory perception of oil samples. To further understand the relationship between volatile composition and the sensory quality of virgin olive oils, the volatile profiles of freshly produced virgin olive oils and defective oil samples were determined using solid phase microextraction (SPME)-GC-MS. Both the factors of origin and cultivar showed significant correlations with the volatile composition of fresh virgin olive oils, with geographic origin having the dominant role. In the study of negative sensory attributes, reference standards of each of main sensory defects found in virgin olive oil (rancid, fusty, winey-vinegary and frozen) were developed. The defective reference standards can be reproduced with ease in the laboratory and used for sensory panel training. To the best of our knowledge, there are currently no reference samples available for olive oil though they exist for some other foods and beverages such as wine and beer. To reduce the instrumental limitation on the determination of volatile carbonyls in the oil matrix, which can be useful to evaluate degree of oxidation, a novel purge and trap-ultra high performance liquid chromatography diode array detector (P&T-UHPLC-DAD) method was established. This method was found suitable for the quantitative analysis of volatile carbonyl compounds in an oil matrix. The quantification, accuracy and repeatability of the P&T-LC method were evaluated through the analysis of oil samples spiked with carbonyl standards. The comparison of this new method and traditional SPME-GC method showed the possibility of using HPLC to determine volatile carbonyls when GC is not available.

Book excerpt: The prevention and bioactivity effects associated with the so-called "Mediterranean diet" make olive oil the most consumed edible fat in the food intake of the Mediterranean basin. The road to quality demands that legislation should be followed. Hence, official European Union classifications such as protected designation of origin (PDO), protected geographical indication (PGI), etc. guarantee the quality and the origin of the labeled foodstuff. The profiling of volatile components and the aroma of olive oil are key factors in the quality dimension and are affected by various factors and conditions such as cultivar; atmospheric, pedologic, and fostering conditions; the ripening degree; olive and oil storage; and the technology of oil extraction from drupes, as well as the quality of the pre-extraction procedures. In extra virgin olive oil production, as in all kinds of production, the maintenance of high quality standards is assured by the olive fruits' and the final products' quality. Modern milling technologies can aid in the direction of quality and safety and thus can be employed in the production of extra virgin olive oil (EVOO), which can be directly consumed without any further manipulation. The overall quality of EVOO should be determined by quality characteristics including sensory analysis, stability, and nutritional value and safety (microbiology, absence of contaminants and toxins), along with authenticity. Food authenticity issues are very important for the food industry due to legislation aspects, economics, quality specifications and conformance, safety concerns, and religious matters. Authentic EVOO should comply with the producer's declaration regarding the quality of olive fruits, natural components, the absence of extraneous substances, production technology, the geographical and botanical origin, the production year, and the genetic identity. Hence, olive oil authenticity can be implemented by the validation of the application of accurate specifications for olive fruits and the selection of trustworthy suppliers with a quality assurance system in place. Authenticity methodologies will avoid adulteration but will also aid the control of accidental contaminations, e.g., in factories, where several oils are produced or used at the same time, or cross-contaminations.
