Book excerpt: Le mode HCCI (Homogeneous Charge Compression Ignition) est un procédé de combustion qui permet une réduction substantielle des NOx et des particules sans pénaliser le rendement thermique. La combustion HCCI se déroule essentiellement par auto-inflammation instantanée de la charge à différents points de la chambre de combustion. Elle est fortement contrôlée par la cinétique chimique qui détermine en partie la phase d'auto-inflammation. Il est donc indispensable de prendre en compte les détails de la chimie dans un code calcul moteur, afin de mieux reproduire les phénomènes importants de ce type de combustion. L'objectif de ce travail est de développer un modèle d'auto-inflammation prenant en compte les détails de la cinétique chimique ainsi que les interactions entre la chimie et la turbulence pouvant fortement influencer le processus. L'accent est mis dans un premier temps sur l'analyse des effets de la dilution par des espèces résiduelles telles que le CO et le H2 sur le processus d'auto-inflammation. Un nouveau modèle d'auto-inflammation (TKI-PDF) est proposé. Son développement a été divisé en deux phases distinctes. La première concerne la prise en compte de la cinétique chimique d'auto-allumage. La deuxième a été dédiée à l'introduction des effets de fluctuations de la richesse dues à la turbulence sur le taux de réaction moyen. Une méthode de tabulation a priori pour le taux de réaction d'une variable d'avancement est utilisée pour décrire la chimie détaillée. Pour prendre en compte les effets de la turbulence, une méthode de pdf présumée est ensuite appliquée. Le modèle a été validé à plusieurs niveaux. Tout d'abord, les résultats de simulations sans prise en compte des effets de la turbulence ont été comparés à des solutions de calculs avec chimie complexe en configuration homogène. Puis, le modèle complet a été validé par comparaison avec des résultats expérimentaux en cellule haute pression avec injection Diesel haute pression et en moteurs Diesel et HCCI.

La thématique s'inscrit dans le cadre général des systèmes industriels tels que les moteurs diesel, basés sur une combustion turbulente initiée par l'auto-inflammation d'un combustible injecté sous forme liquide. L'outil utilisé est un code de simulation numérique directe (DNS) dans lequel la phase gazeuse est résolue par une approche Eulérienne tandis que la phase liquide dispersée est modélisée par une approche Lagrangienne. Pour rendre compte du phénomène complexe d'auto-inflammation, une cinétique chimique semi-détaillée est directement résolue. Dans un premier temps, l'influence des propriétés du gaz et du liquide sur le délai d'auto-inflmammation est étudiée. Sur plusieurs configurations (réacteurs homogènes et hétérogènes, turbulence homogène isotrope), des comparaisons entre auto-inflammation en milieu diphasique et auto-inflammation purement gazeuse mettent en évidence les modifications induites par le spray en évaporation. L'accent est mis sur la localisation des zones les plus réactives et sur l'existence d'une forte corrélation entre les zones à faible dissipation scalaire et les sites les plus favorables à l'auto-inflammation. Dans un second temps, le modèle de tabulation chimique FPI (Flame Prolongation of IDLM) basé sur la fermeture des taux de réaction à partir de fractions massiques des espèces tabulées a été implanté pour évaluer sa capacité à reproduire les résultats obtenus par l'approche chimie complexe. Les comparaisons avec la chimie complexe résolue ont été effectuées sur des cas homogènes et des cas turbulents en présence de gouttelettes. Il s'avère que la présence d'un spray évaporant modifie fortement les processus d'évolution de la chimie rendant difficile l'utilisation de tabulations "a priori".

Nous souhaitions identifier de nouveaux corégulateurs de PPARg.Le criblage par double-hybride dans la levure d'une banque d'ADNc de tissu adipeux humain avec le domaine DE de PPARg comme appât a permis d'identifier plusieurs corégulateurs potentiels de PPARg. Deux d'entre-eux ont été caractérisés : la sous-unité c du facteur de 60kDa associé à BRG1/Brm, nommée BAF60c et le facteur B1 d'ancrage à la matrice nucléaire, appelé SAFB1. BAF60c fait partie du complexe SWI/SNF de remodelage de la chromatine. Nous avons identifié une deuxième isoforme : BAF60c2. Les deux isoformes sont des protéines nucléaires ubiquistes. Elles se lient à divers facteurs de transcription et coactivent les récepteurs nucléaires. BAF60c n'est impliqué ni dans la différenciation adipocytaire ni dans la prolifération cellulaire. La protéine nucléaire ubiquiste SAFB1 se lie aux récepteurs nucléaires et inhibe leur activité transcriptionnelle. Son niveau d'expression évolue au cours de la différenciation adipocytaire. SAFB1 pourrait influencer le choix entre prolifération cellulaire et différenciation cellulaire. Nous avons identifié et caractérisé de nouveaux corégulateurs des récepteurs nucléaires. Les données obtenues doivent être replacées dans un contexte moléculaire complexe. Des études dans des souris génétiquement modifiées nous aideront à évaluer l'impact physiologique de chaque protagoniste. Cela nous permettra d'obtenir une meilleure vue globale du faisceau de liens moléculaires existant entre la régulation génique et les stimuli externes associés à la nutrition et conduira à terme à l'élaboration de nouvelles stratégies thérapeutiques.

D'autre part, les voies de signalisation des MAP kinases, en particulier p38, participent aussi à l'activation des cellules bronchiques par l'étirement. Finalement, il a été établit que deux autres ARN messagers sont actives en sus de l'IL-8 au cours de l'étirement mécanique parmi les 588 ARNm testés : MCP-1 et GADD45.
