The book gives information on the usual techniques for derivation of human dendritic cells from precursor stem cells, such as monocytes. In addition it provides data on the difficult tasks of isolating dendritic cells directly from different tissues; whether dendritic cells from precursor cells or from tissues of mouse or human are required, this book contributes practical information. The last section of the book is devoted to functional aspects of dendritic cells ranging from inf- mation relevant to cell migration to antigen uptake and T cell stimulation. But what is a dendritic cell? The explosion of studies on this cell type over the last few years is breathtaking. We now have so-called “myeloid” and “plas- cytoid” dendritic cells; are they of separate lineages or are they related? There are claims that dendritic cells can be derived from B cells and from granulocytes, as well as from “monocytes. ” Lymphocytes can be switched on or off by dendritic cells. Dendritic cells can preferentially cause Th1 or Th2 responses or some may stimulate B cells directly. When there is so much scope for exploring these new developments, it is particularly important that the data that are established and the practical basis for this work are clearly available, a task that this book fulfils.

Radiographic contrast media (CM) are substances whose primary purpose is to enhance diagnostic information of medical imaging systems. The position of CM in pharmacology is unique. First, there is the unusual requirement of biological inertness. An ideal CM should be completely biologically inert, i.e., stable, not pharmacologically active, and efficiently and innocuously excretable. Because they fail to meet these requirements, CM must be considered drugs. The second unusual aspect of CM is that they are used in large quantities, their annual production being measured in tens of tons. It is not in spite of, but because of, the increased use of new radiographic systems, computed tomography, digital radiography, etc., that consumption is on the rise. And, it is not likely that the other emerging imaging modalities - NMR, ultrasonography, etc. - will displace radiographic CM soon; it is quite probable that these remarkable compounds will continue to play an active role in diagnostic imaging in the foreseeable future.

Sir William Osler Over a period of five years, the impact of computed tomography (CT) on pediatric neuroradiology at The Hospital for Sick Children, Toronto, has been, as expected, in the assessment of the brain and its abnormalities. Concurrent with this application was the introduction of Metrizamide (Amipaque, Nyegaard & Co. AS, Oslo, Norway), a water-soluble CSF contrast medium, used primarily as a myelog raphic agent. The subsequent application of the wide-aperture CT scanner to imaging of the spine in children provided remarkable advances in the clinical management of spinal disease since CT is far more accurate than standard neuroradiologic procedures. The com bination of CT and Metrizamide added a further dimension to the imaging of the spine and of the spinal cord and nerve roots. Such spinal CT and CT Metrizamide myelography in children now occupies a significant part of day-to-day pediatric neuroradiologic practice. They have dramatically enhanced our understanding of the normal anatomy and pathologic entities of the spine and its contents in children; have altered and improved the surgical management of such diseases; and have significantly improved the clinical manage ment of such diseases in the specialties of neurosurgery, orthopedic surgery, and genito-urinary surgery . .

Originally, the publication grew outofprotocols used in the Tissue Culture Course given at the University of Saskatchewan. The course was patterned on those given by the Tissue CultureAssociation, first in Toronto, Canada, in 1948, then in Cooperstown, NY, then Denver, CO, and finally in Madison, WI, where the course ended in 1964. The course in Saskatchewan began in 1963 as a month-long international course that included both animal and plant tissue cultures. Over the years the course underwent specialization, first being limited to animal tissue culture, then to an intensive one-week general course. This led to one-week courses especially designed for tissue culture for the study of cancer or of the cardiovascular or the nervous system. In 1989, the Saskatchewan course became part of the Tissue Culture Training Facility of the Neuroscience Network of the Canadian Network of Centres of Excellence. The course and the Training Facility ceased to exist in 1997. The faculty for the Saskatchewan course was drawn from the best labora tories in the world and laboratory protocols from those centers were thoroughly tested in a student laboratory setting for many years.
