Book excerpt: There have been many important advances in our understanding of biological membrane structure and function over the last decade. Much of this progress has been driven by the development of new techniques for studying membrane components and their interactions. Tradition ally, the investigation of membranes has largely occurred within the domains of biochemistry, physical chemistry, and cell biology; but many of the most significant advances have resulted from an expansion into other disciplines, such as molecular biology, immunology, and the clinical sciences. In these two volumes we have attempted to bring together some of these techniques—to combine the old and the new. Biomembrane Protocols: II. Architecture and Function is concerned exclusively with the architecture and activities of membranes, whereas the companion volume, Biomembrane Protocols: I. Isolation and Analysis, deals with their isolation and compositional analysis. The aim of each chapter is to provide detailed technical and meth odological information that will allow the reader to perform the tech nique successfully, without the need to consult other texts. Detailed reviews of membrane structure and function are not included, except where they are relevant to the choice or efficacy of a particular proce dure. In addition to Materials and Methods sections, each chapter has a Notes section that explains the reasons for taking certain steps and pro vides practical tips for applying the techniques to other systems.

Book Immunocytochemical Methods and Protocols

Download or read book Immunocytochemical Methods and Protocols written by Lorette C. Javois and published by Springer Science & Business Media. This book was released on 2008-02-02 with total page 414 pages. Available in PDF, EPUB and Kindle. Book excerpt: The principle that antibodies can be used as cytochemical agents provided they are tagged with suitable markers has been evident for over 50 years. During this time the use of immunocytochemical meth ods has spread to a wide array of biological disciplines. Early applica tions focused on the detection of microbial antigens in tissues, while more recent applications have used monoclonal antibodies to study cell differentiation during embryonic development. For a select few disci plines, volumes have been published focusing on the specific applica tion of immunocytochemical techniques to that discipline. What distinguishes the present book, Immunocytochemical Meth ods and Protocols, from earlier books is its broad appeal to researchers in all disciplines, including those in both research and clinical settings. The methods and protocols presented here are designed to be general in their application and the accompanying "Notes" provide invaluable assistance in adapting or troubleshooting the protocols. Interspersed throughout the book are chapters providing overviews of select topics related to immunocytochemistry.

Book Membrane Biogenesis

Download or read book Membrane Biogenesis written by Doron Rapaport and published by . This book was released on 2013 with total page 404 pages. Available in PDF, EPUB and Kindle. Book excerpt: Membrane proteins and membrane lipids form complex interactive systems that are highly dynamic and able to be studied only by combinations of different in vivo and in vitro techniques. In Membrane Biogenesis: Methods and Protocols, experts in the field present a broad collection of methods to study the biogenesis and function of cellular membranes. Beginning with how membrane lipids or membrane proteins can be studied, this detailed volume continues with sections covering different procedures to investigate the interaction of membrane proteins among each other or with membrane lipids, methods to study the biogenesis of membrane proteins and the dynamics of organelles, as well as protocols for the analyses of the functions or complex organization of membrane proteins. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Extensive and easily applicable, Membrane Biogenesis: Methods and Protocols provides readers with a comprehensive but still concise collection including both basic protocols of rather general application and more specialized methods for specific and novel techniques.

Book Protein Purification Protocols

Download or read book Protein Purification Protocols written by Shawn Doonan and published by Springer Science & Business Media. This book was released on 2008-02-02 with total page 406 pages. Available in PDF, EPUB and Kindle. Book excerpt: Hans Neurath has written that this is the second golden era of enzymology {Protein Science [1994], vol. 3, pp. 1734—1739); he could with justice have been more general and referred to the second golden age of protein chemistry. The last two decades have seen enormous advances in our understanding of the structures and functions of pro teins arising on the one hand from improvements and developments in analytical techniques {see the companion volume, Basic Protein and Peptide Protocols, in this series) and on the other hand from the tech nologies of molecular genetics. Far from turning the focus away from protein science, the ability to isolate, analyze, and express genes has increased interest in proteins as gene products. Hence, many laborato ries are now getting involved in protein isolation for the first time, either as an essential adjunct to their work in molecular genetics or because of a curiosity to know more about the products of the genes that they have been studying. Protein Purification Protocols is aimed mainly at these newcom ers to protein purification, but it is hoped that it will also be of value to established practitioners who may find here techniques that they have not tried, but which might well be most applicable in their work. With the exception mainly of the first and last chapters, the format of the contributions to the present book conform to the established format of the Methods in Molecular Biology series.

Book Animal Cell Electroporation and Electrofusion Protocols

Download or read book Animal Cell Electroporation and Electrofusion Protocols written by Jac A. Nickoloff and published by Springer Science & Business Media. This book was released on 2008-02-03 with total page 367 pages. Available in PDF, EPUB and Kindle. Book excerpt: The ability to introduce macromolecules into animal cells, includ ing DNA, RNA, proteins, and other bioactive compounds has facili tated a broad range of biological studies, from biochemistry and biophysics to molecular biology, cell biology, and whole animal stud ies. Gene transfer technology in particular will continue to play an essential role in studies aimed at improving our understanding of the relationships between the gene structure and function, and it has impor tant practical applications in both biotechnology and biomedicine, as evidenced by the current intense interest in gene therapy. Although DNA and other macromolecules may be introduced into cells by a variety of methods, including chemical treatments and microinjection, el- troporation has proven to be simpler to perform, more efficient, and effective with a wider variety of cell types than other techniques. The early and broad success of electric field-mediated DNA transfer soon prompted researchers to investigate electroporation for transferring other types of molecules into cells, including RNA, enzymes, antibodies, and analytic dyes. Animal Cell Electroporation and Electrofusion Protocols begins with three chapters that describe the theoretical and practical aspects of electroporation, including a review of the commercially available instrumentation. These introductory chapters will be of particular inter est to those new to electric field technologies and to those developing protocols for as yet untested species or cell types. Nineteen chapters follow that present well-tested protocols for electroporation of proteins and DNA into insect, fish, and mammalian cells.

Book Signal Transduction Protocols

  • Author : David A. Kendall
  • Publisher : Springer Science & Business Media

Download or read book Signal Transduction Protocols written by David A. Kendall and published by Springer Science & Business Media. This book was released on 2008-02-02 with total page 305 pages. Available in PDF, EPUB and Kindle. Book excerpt: As our understanding of the biological sciences expands, the bou- aries between traditional disciplines tend to blur at the edges. Physio- gists and pharmacologists, for instance, now need to embrace techniques that until recently were the strict preserves of biochemists and mole- lar biologists. However, the acquisition of new technologies can be a time-consuming and frustrating business, and unless an expert is on hand to give instruction, precious hours can be spent poring over half-described Methods sections with no guarantee of eventual success. The aim of Signal Transduction Protocols has been to get experts with "hands-on" experience in particular techniques to give detailed accounts of experimental protocols in a recipe-type format, which we hope will circumvent the problems of ambiguity often encountered when reading the literature. The techniques described in Signal Transduction Protocols are those that we think will be most useful in addressing questions in the area of receptor-mediated cell signaling, with particular regard to those receptors that are part of the G-protein-linked superfamily. To keep it to a manageable size, we have omitted any reference to electrophysi- ogy and have instead concentrated on more biochemical approaches.

Book Chromosome Analysis Protocols

    Book Details:
  • Author : John R. Gosden
  • Publisher : Springer Science & Business Media

Download or read book Chromosome Analysis Protocols written by John R. Gosden and published by Springer Science & Business Media. This book was released on 2008-02-02 with total page 508 pages. Available in PDF, EPUB and Kindle. Book excerpt: Chromosomes, as the genetic vehicles, provide the basic material for a large proportion of genetic investigations, from the construction of gene maps and models of chromosome organization, to the inves tigation of gene function and dysfunction. The study of chromosomes has developed in parallel with other aspects of molecular genetics, beginning with the first preparations of chromosomes from animal cells, through the development of banding techniques, which permitted the unequivocal identification of each chromosome in a karyotype, to the present analytical methods of molecular cytogenetics. Although some of these techniques have been in use for many years, and can be learned relatively easily, most published scientific reports—as a result of pressure on space from editors, and the response to that pressure by authors—contain little in the way of technical detail, and thus are rarely adequate for a researcher hoping to find all the necessary information to embark on a method from scratch. A new user needs not only a detailed description of the methods, but also some help with problem solving and sorting out the difficulties en countered in handling any biological system. This was the require ment to which the series Methods in Molecular Biology is addressed, and Chromosome Analysis Protocols forms a part of this series.
