Book excerpt: A NATO Advanced Study Institute on "Membrane Receptors, Dynamics and Energetics" was held in order to consider recent developments in membrane receptor research. Presentations and discussions focussed on signal transduction mechanisms (e.g. 2 G-proteins, transducin, Ca +) and on the structure and func tion of some selected receptors (i.e. low density lipoprotein receptor, acetylcholine receptor, growth factor receptors, toxin receptors). These topics were put in the larger con text of current knowledge on the structure and function of membranes; connections between different fields of research were established by in-depth discussions of energy transduc tion and transport mechanisms. Specialized membrane systems were discussed including chloroplasts and chromaffin granules. Functional relationships with specific lipids (e.g. phospho inositides, lipid A) were major topics of discussion. This book presents the content of the major lectures and a selection of most relevant posters presented during the course of the Institute. This book is intended to make the proceedings of the Institute accessible to a larger audience and to offer a comprehensive account of those topics of re ceptor, membrane and bioenergetics research that were at the center of interest during our stay on the Island of Spetsai from August 17 to 30, 1986.

The first two chapters deal with biomembranes, providing an introduction to the membranes of eucaryotes and a description of the evolution of membranes. The following chapters are concerned with different aspects of lipids including the physical properties of model membranes composed of lipid-protein mixtures, lateralphase separation of lipids and proteins and measurement of lipid-protein bilayer diffusion. Other chapters deal with the flexibility of fluid bilayers, the closure of bilayers into vesicles which attain a large variety of different shapes, and applications of lipid vesicles and liposomes. Part B covers membrane adhesion, membrane fusion and the interaction of biomembranes withpolymer networks such as the cytoskeleton. The first two chapters of this part discuss the generic interactions of membranes from the conceptual point of view. The following two chapters summarize the experimental work on two different bilayer systems. The cytoskeleton within eucaryotic cells consists of a network of relatively stiff filaments of which three different types of filaments have been identified. For more than 15 years, mitochondria have served as the paradigm organelle system to study these processes. Although key questions, such as how precisely proteins cross a membrane, still remain to be answered, exciting progress has been made in understanding the basic pathways of protein import into mitochondria and the components involved. In addition to a fascinating richness and complexity in detail, the analysis of mitochondrial protein import has revealed mechanistic principles of general significance: Major discoveries include the demonstration of the requirement of an unfolded state for translocation and of the essential role of molecular chaperones on both sides of the membranes in maintaining a translocation-competent conformation and in protein folding after import. It is becoming clear how a polypeptide chain is "reeled" across the membrane in an ATP-dependent process by the functional cooperation of membrane proteins, presumably constituting part of a transmembrane channel, with peripheral components at the trans-side of the membrane.In this volume, eminent experts in the field take the time to review the central aspects of mitochondrial biogenesis.
