In the first of these, Papahadjopoulos, Poste, and Vail extensively review much of the available data on the fusion of natural membranes, model membranes (liposomes), and natural membranes with liposomes. The authors are led by their review of the experimental methods and their interpretations of the results obtained to a general theory of membrane fusion which they believe is applicable to all systems that have been studied. Arguing that although protein and carbohydrate may serve, in some cases, to bring membranes into sufficiently close proximity for fusion to occur and, in other cases, to remove peripheral and integral proteins from the regions that are to undergo fusion, the authors conclude that membrane fusion per se is solely a property of the lipid bilayer. In their view, all the experimental observations to date can be subsumed under a unifying hypothesis in which membrane fusion is the result of a phase separation in one-half of the membrane bilayer brought about by the interaction - of calcium ions with acidic phospholipids, mostly phosphatidylserine. Where half-membranes already contain sufficient acidic phospholipids, a local increase in calcium ion concentration may suffice to induce fusion (examples might include exocytosis and fusion of intracellular membrane systems). In other cases, natural or experimentally induced events preceding fusion might be necessary to increase the local concentration of the acidic phospholipids in the half-membrane (virus-or fusogenic agent-induced cell-to-cell fusion, or endocytosis, for example).

The objective of this Institute was to survey recent developments and to discuss future directions in the rapidly advancing field of membrane cell biology, with particular emphasis on the dynamical properties and intracellular flow of membranes. A wide range of interrelated topics around the central theme of intracellular trafficking of membranes was covered, including lipid flow, membrane fusion, dynamics of membrane components, protein folding and assembly, vesicular transport in membrane biogenesis, exocytosis and endocytosis. A large variety of experimental techniques and systems, including the application of viruses and model systems, to study these processes was also considered. Membrane cell biology is a broad discipline which encompasses many scientific areas including cell biology, biochemistry, biophysics, virology, immunonology and genetics. Indeed, recent advances in the cell biology of membranes could not have been made without this multidisciplinary approach. Significant progress achieved during the last few years in understanding how newly synthesized lipids and proteins find their way to the cell organelles, how molecular sorting and the continuous flow of membranes allow each cellular membrane to maintain its own distinct molecular composition, and, thereby, the individuality of the various intracellular compartments, was discussed in considerable detail in this Institute.

A recent symposium which took place at the Center for Tomorrow, State University of New York at Buffalo, New York, April 27-29, 1987, was the first meeting of its kind to specifically address the molecular mechanisms of membrane fusion. The Symposium consisted of oral, workshop and poster presentation sessions. The papers presented in the oral and workshop sessions are collected here and arranged approximately in the order presented at the Symposium. These papers are the most up-to-date and representative work at the for front of each aspect of membrane fusion. Although the readers may find some differences in interpretations regarding the molecular mechanisms of membrane fusion, there is an over all concensus that increased hydrophobicity and dehydration of the membrane surface are essential physico-chemical factors for membrane fusion to occur. We trust that these papers will contribute to your further understanding of the mechanisms of membrane fusion.
