Book excerpt: A preface should justify the existence of the book it precedes and this is invariably done in scientific texts by reference to the explosive growth of the field since the last such volume appeared. In molecular biology, most fields can be justifiably described as growing explosively, as should be the case for a young and vigorous science, but the study of membrane proteins stands out as one which has taken giant strides in the last few years. Ignorance of the structure and function of membrane proteins at the molecular level was certainly not due to lack of interest but rather was a result of lack of appropriate techniques. It has above all been the development of new experimental methods which has wrenched membrane biochemistry out of what Anthony Martonosi fetchingly called its 'romantic phase' (Le. lots of ideas and few facts), into an era when the determination of membrane protein structure and mechanism is a reasonable goal. Membrane proteins are generally classified as peripheral or integral. Peripheral proteins are relatively easily dissociated from membranes by mild treatments whence their study is essentially no different to that of soluble proteins. This book therefore concentrates on integral proteins which are strongly bound to the membrane by hydrophobic interactions with lipids. A crucial step in their study is of necessity the d~velopment of methods of solubilization and purification under non-denaturing conditions.

I began writing this book in the summer of 1975 at Emory University under a grant from the National Library of Medicine. Part of the writing was also done at the Marine Biological Laboratory Library in Woods Hole and in the Library of the Stazione Zoologica in Naples. I wrote this book because in the intervening years membrane noise became a definable subdivision of membrane biophysics and seemed to deserve a uniform treatment in one volume. Not surprisingly, this turned out to be much more difficult than I had imagined and some areas of the subject that ought to be included have been left out, either for reasons of space or because of my own inability to keep up with all aspects of the field. This book is written for biologists interested in noise and for physicists and electrical engineers interested in biology. The first three chapters attempt to bring both groups to a common point of understanding of electronics and electrophysiology necessary to the study of noise and impedance in membranes. These chapters arose out of a course given over a period of six years to electrical engineers from the Georgia Institute of Technology and biologists from Emory University School of Medicine.

The functions of membrane proteins include cell/cell and cell/extracellular matrix recognition, the reception and transduction of extracellular signals, and the tra- port of proteins, solutes and water molecules. Abnormal membrane protein expression has profound biological effects and may, for example, underlie phenotypic and functional differences between normal and tumour cells. Moreover the accessibility, particularly of plasma proteins traversing the plasma membrane of cells, makes them of particular ut- ity to the therapeutic intervention in disease. Indeed, it is estimated that of all currently licensed pharmaceuticals, approximately 70% target proteins resident in the plasma m- brane. In theory, unbiased technologies such as proteomics have the power to de?ne patterns of membrane protein expression characteristic of distinct states of cellular development, differentiation or disease, and thereby identify novel markers of, or targets for intervention in, disease. However, although about 25% of open reading frames in fully sequenced genomes are estimated to encode integral membrane proteins, global analysis of membrane protein expression has proved problematic. Membrane protein analysis poses unique challenges at the level of extraction, solubilization, and separation in particular, and to a lesser extent of identi?cation and quantitation. These challenges have, however, fostered creativity, in- vation, and technical advances, many of which are brought together in Membrane P- teomics.

Structural ana- sis of multiprotein complexes helps to define the role and interaction of each individual component, and can predict the consequences of protein mutation or conditions that promote dissociation and rearrangement of the complex. Determining the three-dimensional structure of a protein requires milligram quantities of pure material. Such quantities are required to refine crystallization conditions for X-ray analysis, or to overcome the sensitivity limitations of NMR spectroscopy. Historically, structural determination of proteins was limited to those expressed naturally in large amounts, or derived from a tissue or cell source inexpensive enough to warrant the use of large quantities of cells. H- ever, with the advent of the techniques of modern gene expression, many p- teins that are constitutively expressed in minute amounts can become accessible to large-scale purification and structural analysis.

The chapters describe water organization at the lipid membrane–water interphase, the water penetration, the long range water structure in the presence of lipid membranes by means of X-ray and neutron scattering, general polarization, fluorescent probes, ATR-FTIR and near infrared spectroscopies, piezo electric methods, computer simulation and surface thermodynamics. Permeation, percolation, osmotic stress, polarization, protrusion, sorption, hydrophobicity, density fluctuations are treated in detail in self-assembled bilayers. Studies in lipid monolayers show the correlation of surface pressure with water activity and its role in peptide and enzyme interactions. The book concludes with a discussion on anhydrobiosis and the effect of water replacement in microdomains and its consequence for cell function. New definitions of lipid/water interphases consider water not only as a structural-making solvent but as a mediator in signalling metabolic activity, modulating protein insertion and enzymatic activity, triggering oscillatory reactions and functioning of membrane bound receptors. Since these effects occur at the molecular level, membrane hydration appears fundamental to understand the behavior of nano systems and confined environments mimicking biological systems.
