Book excerpt: Melanin and melanin-related materials have become novel and blooming topics for both academia and industry, due to their extraordinary functions and properties. It is found that in jaws of Glycera, melanin increases the stiffness and provides an example of organic-based particles used for reinforcement instead of inorganic materials. Although there are examples where melanin particles are added to polymers for improving mechanical performance, the mechanical properties of melanin are unknown. Here, we have probed the mechanical properties of melanin using Atomic Force Microscopy by indenting individual natural melanosome and analyzing force-displacement curves to measure the mechanical response. We found the Young's modulus obtained from at least five melanosomes for each species were 1.45±0.50 and 1.45±0.78 GPa for crow (Corvus ossifragus) and turkey (Melleagris gallopavo), respectively. The results indicate that both melanosomes have indistinguishable modulus, which are assumed to arise from the fact that they are similar in size and chemistry. Melanin is also known for protecting biological tissues from harmful UV rays. It is known that exposure to UVA results in chemical changes in melanin. Therefore, we have also used AFM to measure if the mechanical modulus is affected by UV radiation. We find that the Young's modulus for crow and turkey melanosomes after UV treatment are 1.59±0.67 and 0.97±0.53 GPa, respectively, which are statistically similar to the results without UV treatment. It was surprising that the mechanical properties were not affected by UV even though the melanosomes are degrading chemically.
