Book MECHANISMS OF LEFT VENTRICULAR REMODELING IN PHYSIOLOGICAL AND PATHOLOGICAL CONDITIONS

Download or read book MECHANISMS OF LEFT VENTRICULAR REMODELING IN PHYSIOLOGICAL AND PATHOLOGICAL CONDITIONS written by Gabriel Grilo and published by . This book was released on 2020 with total page 109 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cardiac remodeling is composed of molecular, cellular, and interstitial changes in the cardiac tissue that affect the size, shape, and function of the heart. There are two types of cardiac remodeling: physiological and pathological remodeling. Physiological remodeling of the heart is an adaptation of the organ based on the body's demand, such as changes due to physical exercises and during aging. Cardiac pathological remodeling can occur due to the evolution of a chronic disorder in the cardiovascular system or after an acute injury, such as myocardial infarction (MI). In this thesis, we investigated both physiological and pathological cardiac remodeling, with particular focus on the role of the extracellular matrix (ECM). Determining the mechanisms involved in cardiac remodeling by changes in ECM provides insight to distinguish the local and functional changes from external risk factors; and it provides identification of novel targets that could be used as therapeutic approaches to reduce cardiac dysfunction. In our first study, we hypothesized that changes in ECM composition during physiological remodeling with age occurs in a sex-specific manner, since cardiac function varies between sexes among cardiovascular disease patients. We assessed cardiac parameters using both conventional echocardiography and speckle tracking echocardiography (STE). Our results suggest that STE allows for early detection of changes in cardiac function between sexes during aging. ECM factors involved in collagen metabolism, such as decorin, osteopontin, Cthrc1, and Ddr1 expression were age-dependent but sex-independent; while periostin, lysyl oxidase, and Mrc2 displayed age-dependent and sex specific differences. These data highlight the importance of including sex-differences analysis when studying cardiac aging. In our second study, we investigated the role of a collagen-derived matricryptin in pathological remodeling. Matricryptins are biologically active peptides, generated from ECM proteolysis, able to regulate cell function and survival. We tested the potential of the matricryptin p1159 to reduce adverse cardiac remodeling using a rodent MI model. A previous study from our lab showed that p1159 plasma levels negatively correlate with left ventricle (LV) filling pressure, suggesting a beneficial role against adverse remodeling. In this thesis, we found that p1159 increases cardiac fibroblast migration by activating RhoA pathways via the membrane receptor integrin alpha 4. Fibroblast migration is an essential step during cardiac healing. In addition, p1159 significantly improved cardiac function post-MI by inducing the formation of a compliant and organized infarct scar, which promoted LV contractility and preserved the structural integrity of the heart. Our data strongly supports matricryptin p1159 as a therapeutic treatment to reduce adverse remodeling post-MI.

Book Cardiac Remodeling New Insights in Physiological and Pathological Adaptations

Download or read book Cardiac Remodeling New Insights in Physiological and Pathological Adaptations written by Leonardo Roever and published by Frontiers Media SA. This book was released on 2017-11-15 with total page 119 pages. Available in PDF, EPUB and Kindle. Book excerpt: The effective management of Cardiac remodeling(CR), remains a major challenge. Heart failure remains the leading cause of death in industrialized countries. Yet, despite the enormity of the problem, effective therapeutic interventions remain elusive. In fact, several initially promising agents were found to decrease mortality in patients recovering from myocardial infarction. Cardiac remodeling is defined as molecular and interstitial changes, manifested clinically by changes in size, mass , geometry and function of the heart in response to certain aggression. Initially, ventricular remodeling aims to maintain stable cardiac function in situations of aggression.

Book Cardiac Remodeling

Download or read book Cardiac Remodeling written by Jerald Sherman and published by Nova Science Publishers. This book was released on 2016 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cardiovascular diseases are the leading cause of death in almost 40% of patients suffering from end stage renal disease (ESRD). Cardiomyopathy and ischemic heart disease are the most frequent causes of cardiac death. The risk of cardiovascular mortality in dialysis patients is 10 to 20 times greater than the general population, particularly in younger patients, taking into account that the relative risk decreases with age. Left ventricular hypertrophy (LVH) is the most common cardiac abnormality in chronic kidney disease (CKD), and the survival risk ratio in such patients is independent. This book examines the molecular mechanisms, treatments and clinical implication of cardiac remodeling. The first chapter discusses risk factors for cardiovascular disease in patients on continuous ambulatory peritoneal dialysis. The following chapters examine the impacts tropomyosin, vitamin D, and coffee have on cardiac remodeling.

Book Cardiac Remodeling and Failure

Download or read book Cardiac Remodeling and Failure written by Pawan K. Singal and published by Springer Science & Business Media. This book was released on 2003-02-28 with total page 570 pages. Available in PDF, EPUB and Kindle. Book excerpt: The importance of the developmental approach for experimental and clinical cardiology is indisputable. Clinical-epidemiological studies have clearly shown that the risk factors of serious cardiovascular diseases, such as atherosclerosis and ischemic heart disease, are already present during the early phases of ontogenetic development. Furthermore, congenital cardiovascular malformations remain the single largest cause of infant mortality from congenital defects in industrial countries. It is therefore not surprising that the interest of theoretical and clinical cardiologists in the developmental approach keeps increasing. Advances in molecular biology accelerated this trend substantially. This book is based on contributions presented at the international symposium The Developing Heart in Prague in May 2000. It is our contention that the biological, electrophysiological, morphological, functional, biochemical and functional approaches employed by distinguished scientists worldwide will provide the reader with a global picture for changes characterizing the developing heart. It should stimulate the curiosity of cardiovascular scientists in gaining insight into the mechanisms of normal and pathological development.

Download or read book The Breathless Heart written by Michele Emdin and published by Springer. This book was released on 2016-11-25 with total page 288 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book systematically focuses on central sleep apneas, analyzing their relationship especially with heart failure and discussing recent research results and emerging treatment strategies based on feedback modulation. The opening chapters present historical background information on Cheyne-Stokes respiration (CSR), clarify terminology, and explain the mechanics and chemistry of respiration. Following a description of the physiology of respiration, the pathophysiology underlying central apneas in different disorders and particularly in heart failure is discussed. The similarities and differences of obstructive and central apneas are then considered. The book looks beyond the concept of sleep apnea to daytime CSR and periodic breathing during effort and contrasts the opposing views of CSR as a compensatory phenomenon or as detrimental to the failing heart. The diagnostic tools currently in use for the detection of CSR are thoroughly reviewed, with guidance on interpretation of findings. The book concludes by describing the various forms of treatment that are available for CSR and by explaining how to select patients for treatment.

Book Regulation of Cardiac Contractility

Download or read book Regulation of Cardiac Contractility written by R. John Solaro and published by Biota Publishing. This book was released on 2011-04-01 with total page 46 pages. Available in PDF, EPUB and Kindle. Book excerpt: Contractility describes the relative ability of the heart to eject a stroke volume (SV) at a given prevailing afterload (arterial pressure) and preload (end-diastolic volume; EDV). Various measures of contractility are related to the fraction as the SV/EDV or the ejection fraction, and the dynamics of ejection as determined from maximum pressure rise in the ventricles or arteries or from aortic flow velocities determined by echocardiography. At the cellular level, the ultimate determinant of contractility is the relative tension generation and shortening capability of the molecular motors (myosin cross-bridges) of the sarcomeres as determined by the rates and extent of Ca activation, the turnover kinetics of the cross-bridges, and the relative Ca responsiveness of the sarcomeres. Engagement of the regulatory signaling cascades controlling contractility occurs with occupancy and signal transduction by receptors for neurohumors of the autonomic nervous system as well as growth and stress signaling pathways. Contractility is also determined by the prevailing conditions of pH, temperature, and redox state. Short-term control of contractility is fully expressed during exercise. In long-term responses to stresses on the heart, contractility is modified by cellular remodeling and altered signaling that may compensate for a time but which ultimately may fail, leading to disorders.
