Book excerpt: The objective of this review is to suggest that the advances being made in our understanding of the molecular events surrounding chromosome segregation in non-mammalian and somatic cell models be considered when designing experiments for studying aneuploidy in mammalian germ cells. Accurate chromosome segregation requires the temporal control and unique interactions among a vast array of proteins and cellular organelles. Abnormal function and temporal disarray among these, and others to be inidentified, biochemical reactions and cellular organelles have the potential for predisposing cells to aneuploidy. Although numerous studies have demonstrated that certain chemicals (mainly those that alter microtubule function) can induce aneuploidy in mammalian germ cells, it seems relevant to point out that such data can be influenced by gender, meiotic stage, and time of cell-fixation post-treatment. Additionally, a consensus has not been reached regarding which of several germ cell aneuploidy assays most accurately reflects the human condition. More recent studies have shown that certain kinase, phosphatase, proteasome, and topoisomerase inhibitors can also induce aneuploidy in rodent germ cells. We suggest that molecular approaches be prudently incorporated into mammalian germ cell aneuploidy research in order to eventually understand the causes and mechanisms of human aneuploidy. Such an enormous undertaking would benefit from collaboration among scientists representing several disciplines.

This Symposium developed as a consequence of the concern of the Environmen tal Protection Agency with the support of the National Institute of Envi ronmental Health Sciences about human exposure to environmental agents that cause aneuploidy. The program was chosen to explore what is currently known about the underlying causes, the origins, and the extent of the prob lem of human aneuploidy, and whether exposure to environmental agents is assodated with an increased incidence of aneuploidy in humans. Basic research findings in the area of mitosis and meiosis were presented and related to possible mechanisms of how aneuploidy may be produced spontane ously and chemically. A survey of data regarding the chemical induction of aneuploidy in experimental organisms was presented. Outstanding scientists from different fields were invited to cover a broad perspective of aneu ploidy from the molecular aspects to the human situation. We hope that the publication of the proceedings will share the enthu siasm of the meeting and its scientific content. The topic of aneuploidy has received little attention and it is the purpose of this Volume to establish a scientific basis for assessing health risks posed by environ mental exposures to aneuploidy-inducing chemicals. Book excerpt: The cause of many of the adverse reproductive outcomes and developmental diseases among offspring is not well understood. Most of the epidemiologic and experimental animal research has focused on the relationship between maternal exposures including medications, tobacco smoke, alcohol, infections, and occupation and the occurrence of spontaneous abortion, low birth weight, and birth defects. The potential role of paternal exposures has not been investigated as extensively despite long-standing animal research that demonstrates the induction of mutations in the male germ cell after exposure to certain agents and subsequent reproductive failure or early pregnancy loss. Given this relative lack of interest, acquisition of epidemiologic data and the development of a definitive model or mechanism for potential male-mediated effects has been hindered. However, recent laboratory and epidemiologic investigations have suggested that paternal exposures may be more important than previously suspected. This topic has been termed by some as "male-mediated developmental toxicity. " This is meant to refer to the effects of exposures and other factors relating to the male parent that result in toxicity to the conceptus and abnormal development. The developmental endpoints of interest can include fetal loss, congenital abnormalities, growth retardation, cancer, and neurobehavioral effects. These effects may operate through a variety of mechanisms including gene mutation, chromosomal aberrations, seminal fluid transfer of toxicants and epigenetic events.
