Book Matrix assisted Laser Desorption Fourier Transform Mass Spectrometry for Biological Compounds

Download or read book Matrix assisted Laser Desorption Fourier Transform Mass Spectrometry for Biological Compounds written by and published by . This book was released on 1990 with total page 9 pages. Available in PDF, EPUB and Kindle. Book excerpt: The recent development of matrix-assisted UV laser desorption (LD) mass spectrometry has made possible the ionization and detection of extremely large molecules (with molecular weights exceeding 100,000 Daltons). This technique has generated enormous interest in the biological community for the direct examination of large peptides and oligonucleotides. Although this matrix-assisted ionization method has been developed and used almost exclusively with time-of-flight (TOF) mass spectrometers, research is currently in progress to demonstrate this technique with trapped ion mass spectrometers, such as Fourier transform ion cyclotron resonance mass spectrometry (FTMS). The potential capabilities of FTMS for wide mass range, high resolution measurement, and ion trapping experiments suggest that this instrumental technique should be useful for the detailed structural characterization of large ions generated by the matrix-assisted technique. We have recently demonstrated that matrix-assisted ultraviolet laser desorption can be successfully used with FTMS for the ionization of small peptides. The objective of this report is to summarize the application and current limitations of matrix-assisted laser desorption FTMS for the characterization of peptides and oligonucleotides at the isomeric level. 4 refs., 3 figs., 2 tabs.

Book Mass Spectrometry in the Biological Sciences A Tutorial

Download or read book Mass Spectrometry in the Biological Sciences A Tutorial written by M.L Gross and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 462 pages. Available in PDF, EPUB and Kindle. Book excerpt: The developments in mass spectrometry over the past fifteen years have been impressive in their implications in bioanalytical chemistry. The achievements begin with the inventions of Cf-252 Plasma Desorption Mass Spectrometry by Macfarlane and Fourier Transform Mass Spectrometry by Comisarow and Marshall in the mid 1970s. The former showed the feasibility of producing large gas-phase ions from large biomolecules whereas the latter enhanced the capabilities for ion trapping especially in analytical mass spectrometry. A major achievement was the development by Barber of Fast Atom Bombardment (FAB) mass spectrometry, an advance that heralded a new era in biological mass spectrometry. Contemporary and routine instruments such as magnetic sectors and quadrupoles were rapidly adapted to F AB, and nearly the entire universe of small molecules became amenable to study by mass spectrometry. The introduction of FAB also paved the way for improvement of instrument capability. For example, the upper mass limit of magnet sector mass spectrometers was increased by nearly an order of magnitude by the instrument manufacturers. Furthermore, the technique of tandem mass spectrometry (MS/MS) was given new meaning because important structural information for biomolecules could now be produced for ions introduced by FAB into the tandem instrument. The evolution of MS/MS continues today with the development of ion traps, time-of-flight, and sector instruments equipped with array detection.

Book Development and Characterization of an Atmospheric Pressure Ionization Source Matrix Assisted Laser Desorption Electrospray Ionization Coupled to Fourier Transform Ion Cyclotron Resonance Mass Spectrometry for Analysis of Biological Macromolecules

Download or read book Development and Characterization of an Atmospheric Pressure Ionization Source Matrix Assisted Laser Desorption Electrospray Ionization Coupled to Fourier Transform Ion Cyclotron Resonance Mass Spectrometry for Analysis of Biological Macromolecules written by and published by . This book was released on 2004 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: The field of mass spectrometry has grown tremendously over the past few decades due in large part to the continued application to new and interesting areas of exploration. The advent of soft ionization sources such as electrospray ionization and matrix-assisted laser desorption/ionization have dramatically increased the capabilities of mass spectrometry and increased the amount of information derived from biological samples. As a result, there has been tremendous growth in the development of new ionization source technology in recent years. One of the main driving forces behind the development of new ionization technology is for the reduction of sample preparation required prior to analysis, yielding high throughput sample analysis. Demonstrated herein is the development and characterization of an atmospheric pressure ionization source called matrix-assisted laser desorption electrospray ionization (MALDESI). MALDESI is a hybrid combination of MALDI and ESI which utilizes laser desorption with electrospray postionization for the generation of multiply-charged ions. Multiply-charged ions are of particular importance when using Fourier transform mass spectrometry, due to the increase in resolving power and mass accuracy with increasing charge on the molecule. Top-down characterization of intact polypeptides is demonstrated as well as high mass accuracy utilizing internal calibration. A newly designed highly robust and versatile atmospheric pressure ionization platform is developed and described in detail. Solid- and liquid state analysis of three out of the four classes of biological molecules is demonstrated utilizing the newly developed versatile ionization platform. MALDESI utilizing ultraviolet and infrared laser desorption is demonstrated at various wavelengths (UV, 337 nm and 349 nm and IR, 2.94 Ã'Âæm and 10.6 Ã'Âæm) with and without ESI postionization for the generation of multiply-charged ions. The characteristics of liquid-state samples are described as a mac.

Book Experimental Mass Spectrometry

Download or read book Experimental Mass Spectrometry written by David H. Russell and published by Springer Science & Business Media. This book was released on 2013-06-29 with total page 324 pages. Available in PDF, EPUB and Kindle. Book excerpt: Mass spectrometry underwent dramatic changes during the decade of the 1980s. Fast atom bombardment (F AB) ionization, developed by Barber and coworkers, made it possible for all mass spectrometry laboratories to analyze polar, highly functionalized organic molecules, and in some cases ionic, inorganic, and organometallic compounds. The emphasis of much of this work was on molecular weight determination. Parallel with the development of ionization methods (molecular weight mass spectrometry) for polar biological molecules, the increased mass range of sector and quadrupole mass spectrometers and the development of new instruments for tandem mass spectrometry fostered a new era in structural mass spectrometry. It was during this same period that new instrument technologies, such as Fourier transform ion cyclotron resonance, radio frequency quadrupole ion trap, and new types of time-of-flight mass spectrometers, began to emerge as useful analytical instruments. In addi tion, laser methods useful for both sample ionization and activation became commonplace in almost every analytical mass spectrometry laboratory. In the last 5 years, there has been explosive growth in the area of biological mass spectrometry. Such ionization methods as electrospray and matrix-assisted laser desorption ionization (MALDI) have opened new frontiers for both molecular weight and structural mass spectrometry, with mass spectrometry being used for analysis at the picomole and even femto mole levels. In ideal cases, subfemtomole sample levels can be successfully analyzed. Sample-handling methods are now the limiting factor in analyz ing trace amounts of biological samples.
