Le professeur parvient à convaincre le jeune journaliste et l'invite à assister à une conférence le soir même: là, le professeur Challenger propose que soit montée une expédition destinée à prouver la véracité de ses dires. Ses membres en seront son contradicteur, le professeur Summerlee, le célèbre aventurier et chasseur Lord John Roxton, et enfin, Edward Malone, qui représentera la presse. Summerlee est un zoologiste réputé; quant à Roxton, il connaît bien l'Amazonie, où il a contribué à mettre fin à un trafic d'esclaves. Bientôt, les trois hommes traversent l'océan Atlantique, prennent pied au Brésil et engagent quelques hommes pour leur caravane, parmi lesquels Zamba, un grand homme noir à la force de colosse et d'une loyauté à toute épreuve, et un indien du nom de Gomez. C'est au moment de découvrir les instructions écrites par Challenger que les trois Anglais ont la surprise de le voir paraître en personne devant eux: il va prendre la tête de l'expédition.
