Book Laser Ablation Inductively Coupled Plasma Mass Spectrometry LA ICP MS as a Biomonitoring Tool

Download or read book Laser Ablation Inductively Coupled Plasma Mass Spectrometry LA ICP MS as a Biomonitoring Tool written by Brianne Duncan and published by . This book was released on 2019 with total page 98 pages. Available in PDF, EPUB and Kindle. Book excerpt: Using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) this research analyzed human cadaver hair for gadolinium and mice hair samples for cadmium. This project combined fundamental bench science (analytical chemistry and animal studies), and insight into exposures to an emerging contaminant. This study demonstrated the feasibility of LA-ICP-MS to measure time resolved exposure profiles in hair samples, potentially opening up a transformative new exposure analysis method for investigators conducting both bench toxicology and human epidemiology studies. Consecutive autopsy cases that presented at the University of Washington's medical department, who had received a gadolinium-based contrasting agent (GBCA) to enhance magnetic resonance imaging procedures constituted the human population component of this research. Multiple hairs from numerous subjects were analyzed using LA-ICP-MS along with calibration gels (bovine-skin based) and a standard reference material glass disc with trace metals. Overall analytical method accuracy and precision was acceptable. There was moderate correlation between gadolinium dose and area under the curve as well as gadolinium peak position and GBCA injection day. Hair concentrations were also compared to the other tissue (brain, bone, and skin) concentrations using linear regression. This analysis showed good correlation between hair gadolinium concentrations and brain and skin gadolinium concentrations. These results suggest that hair may serve as a safe and effective biomonitoring tissue for patients who receive GBCA injections. A mouse animal model was used to evaluate LA-ICP-MS as a biomonitoring tool in assessing cadmium exposure. Hair samples from mice that were part of a mechanistic toxicology study at the University of Washington were opportunistically obtained for inclusion in this analytical methods-focused element of the research. The mice were dosed with cadmium-spiked water at three levels (0, 0.6, and 3 mg/L) for 14 weeks before they were sacrificed. Hairs from seven mice were analyzed using LA-ICP-MS. This research showed that there were increasing concentrations of cadmium in hair shafts with increasing doses of cadmium in drinking water among mice.

This book was released on 2017 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: A quantitative reduction strategy consisting of Ratioing, Standardization, and Normalization (RSN) was proposed to process the LA-ICP-MS transient signal of NIST, MPI-DING, USGS and CGSG glass reference materials. The RSN strategy allows the quantitative reduction without knowing the concentration of internal standard prior to LA-ICP-MS analysis. The down-hole fractionations of the investigated glasses in initial 5-35s ablation period were insignificant and independent of the chosen internal standards under the given laser conditions. The analytical accuracy obtained from internal standardi...

Download or read book Laser Ablation Inductively Coupled Plasma Mass Spectrometry and Raman Spectroscopy Imaging of Biological Tissues written by Emma Lee Gorishek and published by . This book was released on 2016 with total page 43 pages. Available in PDF, EPUB and Kindle. Book excerpt: Laser Ablation Inductively coupled plasma mass spectrometry (LA-ICP-MS) and Raman spectroscopy are both powerful imaging techniques. Their applications are numerous and extremely potential in the field of biology. In order to improve upon LA-ICP-MS an in-house built cold cell was developed and its effectiveness studied by imaging Brassica napus seeds. To further apply LA-ICP-MS and Raman imaging to the field of entomology a prong gilled mayfly (Ephemeroptera: Leptophlebiidae) from the Róbalo River, located on Navarino Island in Chile, was studied. Analysis of both samples showcased LA-ICP-MS and Raman spectroscopy as effective instruments for imaging trace elements and larger molecules in biological samples respectively.

Book Analysis of Solids by Laser Ablation Inductively Coupled Plasma Mass Spectrometry

Download or read book Analysis of Solids by Laser Ablation Inductively Coupled Plasma Mass Spectrometry written by David Brian Aeschliman and published by . This book was released on 2003 with total page 188 pages. Available in PDF, EPUB and Kindle. Book excerpt: This dissertation manuscript concentrates on methods to improve the utility of laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) as a rapid and accurate technique for quantitative analysis, forensic matching, and source attribution of solid samples. A solution calibration method for LA-ICP-MS is described that does not require matrix-matched solid standards or prior knowledge of sample composition. Desolvated particles from a microconcentric nebulizer and particles ablated from a localized position on a solid sample are mixed and introduced simultaneously to the ICP-MS. The transport of sample particulates from the ablation cell is measured online with piezoelectric microbalance to provide signal normalization. The relative accuracy of the quantitative analysis of NIST 612 glass and NIST 1264a steel is 3% and 12%, respectively, except for the elements Pb and Bi in steel. LA-ICP-MS is also used with principal components analysis (PCA) to simplify routine comparisons of forensic materials without time-consuming and destructive sample dissolution. The relative concentrations of trace elements in a solid sample are examined to provide a "fingerprint" composition that can be used for definitive identification and source matching of the material. Residue samples are matched to bulk materials using PCA. Variation of laser focus and PCA are also used to diagnose the presence and severity of elemental fractionation, which remains the most significant limitation for accurate quantitative analyses by LA-ICP-MS. High-speed photographs and video of an ICP are used to examine the validity of solution calibration for LA-ICP-MS and to study the trajectories, lifetimes, and emission behavior of particles traversing the plasma under a variety of conditions. No red emission clouds from YO are visible in the ICP when a 20-[mu]L min−1 nebulizer is used to spray a 2000 ppm aqueous yttrium solution, suggesting that the wet droplets from this nebulizer desolvate almost completely before entering the ICP. These desolvated particles appear to atomize and ionize like the small dry particulates from laser ablation. However, some large ablated particulates are observed to fly through the plasma intact, possibly contributing to elemental fractionation. Nevertheless, accurate calibration results using solution standards can be obtained for elements not affected by fractionation.

Book Evaluation of Inductively Couple Plasma time of Flight Mass Spectrometry for Laser Ablation Analyses

Download or read book Evaluation of Inductively Couple Plasma time of Flight Mass Spectrometry for Laser Ablation Analyses written by D. P. Baldwin and published by . This book was released on 2003 with total page 158 pages. Available in PDF, EPUB and Kindle. Book excerpt: The purpose of this trip to LECO Corporation was to test the non-matrix matched calibration method and the principal component analysis (PCA) method on a laser ablation-inductively coupled plasma-time of flight mass spectrometry (LA-ICP-TOFMS) system. An LA-ICP-TOFMS system allows for multielement single-shot analysis as well as spatial analysis on small samples, because the TOFMS acquires an entire mass spectrum for all ions extracted simultaneously from the ICP. The TOFMS system differs from the double-focusing mass spectrometer, on which the above methods were developed, by having lower sensitivity and lower mass resolution.

Book Laser Ablation inductively Coupled Plasma mass Spectrometry

Download or read book Laser Ablation inductively Coupled Plasma mass Spectrometry written by and published by . This book was released on 2011 with total page 192 pages. Available in PDF, EPUB and Kindle. Book excerpt: This dissertation provides a general introduction to Inductively coupled plasma-mass spectrometry (ICP-MS) and laser ablation (LA) sampling, with an examination of analytical challenges in the employment of this technique. It discusses the origin of metal oxide ions (MO+) in LA-ICP-MS, as well as the effect of introducing helium and nitrogen to the aerosol gas flow on the formation of these polyatomic interferences. It extends the study of polyatomic ions in LA-ICP-MS to metal argide (MAr+) species, an additional source of possible significant interferences in the spectrum. It describes the application of fs-LA-ICP-MS to the determination of uranium isotope ratios in particulate samples.
