Book excerpt: Excerpt from Laboratory Guide in Soil Bacteriology Bacteriology has contributed, within the past few years, some important facts to our rapidly expanding knowledge of soils. It has shed a new light on the transformation and accumulation of nitrogen compounds in tilled land, as well as in uncultivated forest and prairie soils. It has taught us to understand more clearly the decomposition of cellulose and allied compounds, and the conservation of the potential energy of hydrogen and of methane. It has given the student of soils and of soil fertility a number of new methods for measuring differences in the quality and quantity of plant-food. It bids fair to supply us with still other methods that would be more adequate than any chemical methods yet devised, for the investigation of fundamental problems of soil fertility. In view of the past achievements of soil bacteriology and of the promise of even greater usefulness in the future, no apology is needed for the publication of this little volume. The authors have felt, for some time, that there is room in every well-arranged course in soils for a series of exercises in soil bacteriology, and have, therefore, made the attempt to provide them. It is their hope that after preliminary instruction in general bacteriology and laboratory technique the "Guide" will prove useful to students of soils, and that time and service will permit the elimination of the imperfections and inadequacies that undoubtedly exist in it. Toward this end, friendly criticism will be appreciated. Meanwhile, the authors wish to express their indebtedness to Dr. R. E. Buchanan, of the Iowa State College, for many helpful suggestions. The author has taken the opportunity to make changes which, he believes, will increase the usefulness of the book. The principal change has come in the new form which will certainly be found more convenient. The sequence of the exercises in Chap. I. has been somewhat changed and a few new subjects have been introduced. The order in which the various bacteria is taken up has been altered so that they are now to be studied in groups. This arrangement is in keeping with recent tendencies, and it is hoped that it will impress the student with the similarity between closely related forms, and also emphasize certain minute but important differences. The addition of a number of new cuts will tend to greater clearness. The general plan of the book remains the same. Valuable suggestions have been received from several investigators and teachers. Especial mention is due Mr. John F. Nicholson, Assistant Bacteriologist to the Wisconsin Experiment Station, for valuable assistance. Suggestion or criticisms will be gladly received. Book excerpt: Excerpt from Laboratory Manual of General Microbiology: With Special Reference to the Microorganisms of the Soil Microorganisms are divided broadly into two large groups, on the basis of their energy requirements: (1) The autotrophic organisms, which can obtain the energy required for their activities from the oxidation of inorganic elements or their compounds or from simple compounds Of carbon, their carbon from carbon dioxide, and their nitrogen and other minerals from inorganic compounds. In addition to those organisms which can obtain their energy from the Oxidation of inorganic substances or simple compounds of carbon (chemosynthetic), the chlorophyll-bearing plants, which obtain their energy photo synthetically are also classified with this group. (2) The heterotrophic organisms which obtain their energy and carbon from complex Organic substances. Among the heterotrophic organisms, however, there is also very considerable specificity. Some organisms can Obtain their energy and carbon only from celluloses, while the great majority of microorganisms cannot attack celluloses and cannot utilize them either as sources Of energy or of carbon. Some organisms can Obtain their nitrogen from inorganic compounds, such as ammonium salts or nitrates; others require only organic nitrogen sources; still others can use even gaseous atmospheric nitrogen. Some can grow at a wide range of reactions; for the activities of others only a very limited range of hydrogen-ion concentration exists. Some organisms are able to grow on a great variety of media, liquid or solid; others will develop only on very specific media, when the particular nutrients are available.
