Il concerne l'utilisation de l'aspartate aminotransférase (AAT) pour la production d'analogues de l'acide L-glutamique (Glu). L'importance biologique de cet acide aminé et notamment ses propriétés de neurotransmetteur constituent une motivation importante de ce travail de recherche. Une étude bibliographique articulée en 3 parties éclaire les principaux aspects de ce travail : les principales méthodes de synthèse des analogues de Glu sont tout d'abord évoquées. Après une description des principales caractéristiques biochimiques des aminotransférases, les procédés de bioconversions mettant en jeu ces enzymes sont décrits en détail. Enfin, les méthodes de synthèse développées pour accéder à des analogues de l'acide 2-oxoglutarique (KG) sont présentées. Ces molécules, substrats potentiels de l'AAT, constituent les précurseurs immédiats des analogues de Glu. Les résultats obtenus sont présentés en 3 points : la synthèse d'analogues de KG, l'étude de la réaction enzymatique et enfin la production des analogues de Glu. Des méthodes de synthèses générales et efficaces ont été mises au point et ont permis de préparer de nombreux analogues de KG, substitués en position 4 pour la plupart d'entre eux. L'étude de la réaction enzymatique nous a permis de montrer que l'AAT de coeur de porc et d'E. coli possèdent une large spécificité de substrat et qu'elles manifestent une énantiosélectivité marquée en faveur des analogues (R)-4-alkyl-KG. Le procédé de bioconversion a été amélioré après une étude concernant l'inhibition de l'enzyme liée à l'utilisation de l'acide cystéine sulfinique (ACS). Plusieurs 4-alkyl-L-Glu ont été préparés et isolés avec une très grande pureté stéréochimique. Ce travail offre de nombreuses perspectives pour la production de nouveaux analogues de Glu mais aussi d'autres acides aminés avec les aminotransférases
