This makes kisspeptin a fundamental component of the reproductive axis. Kisspeptin has been deemed the single most potent stimulator of GnRH neurons yet known. The importance of kisspeptin has been documented in humans as well as non-human animal models, ranging from monkeys, sheep, and rodents to numerous fish species, thus signifying a highly conserved nature of its reproductive function. Importantly, kisspeptin neurons seem to mediate many of the regulatory effects of other signals, whether they are metabolic, circadian, hormonal, or stress. This places kisspeptin neurons in a unique position to be key nodal points and conduits for conveying numerous endogenous and exogenous signals to the reproductive axis.

At the beginning of the 20th century, when the British physiologists, Bayliss and Starling, first proposed endocrinology to be an independent field of inquiry, they went to great lengths to establish the autonomy of chemical secretions in general and their independence from nervous control in particular (Bayliss, W. M. , and Starling, E. H. , 1902, The mechanism of pancreatic secretion,]. Physiol. 28:325). They argued with Pav lov, who said that there was a strong influence of the nervous system on the gastrointestinal phenomena the endocrinologists were studying. For several decades, the English physiologists prevailed, at least in the West; and Pavlov's critique was not taken to heart by the practitioners of the newly emerging discipline of endocrinology. Through the work of Harris, the Scharrers, Sawyer, Everett, and others, there has been something of a scientific detente in the latter half of this century; the hybrid field of neuroendocrinology is now regarded as one of the corner stones of modern neural science and is of fundamental importance in basic and clinical endocrinology.

Book excerpt: One of the major breakthroughs of the last decade in the understanding of energy homeostasis is the identification of a reciprocal control between circadian rhythmicity and cellular metabolism. Circadian rhythmicity is a fundamental endogenous process of almost every organism living on Earth. For instance, the alternation of hunger and satiety is not continuous over 24 h, but is instead structured in time along the light/dark cycle. In mammals, the temporal organization of metabolism, physiology and behavior around 24 h is controlled by a network of multiple cellular clocks, synchronized via neuronal and hormonal signals by a master clock located in the suprachiasmatic nuclei of the hypothalamus. This central circadian conductor in the brain is mainly reset by ambient light perceived by the retina, while secondary circadian clocks in other brain areas and peripheral organs can be reset by meal timing. Chronic disruption of circadian rhythms, as seen in human shift-workers (up to 20% of the active population), has been associated with the development of a number of adverse mental and metabolic conditions. Understanding of the functional links between circadian desynchronization and overall health in animal models and humans, however, is still scarce. Interactions between circadian clocks and metabolism can occur at different levels: the molecular clockwork, internal synchronization via neuro-hormonal signals, or external synchronization via photic or feeding cues. This Research Topic comprises a number of reviews as well as research and methods articles that feature recent advancements in the mechanisms linking circadian clocks with energy metabolism, and the pathophysiological implications of these interactions for metabolic health.

Its 54 amino-acid product, Kisspeptin-54 (Kp-54), was originally called metastin for its ability to inhibit cancer metastasis through the activation of a G coupled receptor, previously known as GPR54 and currently renamed the Kisspeptin receptor (KISS1R). Shorter active peptides all capable of binding to KISS1R have been identified and the physiological activities of these Kisspeptins are now known to extend not only to the suppression of metastasis. Kisspeptins currently represent evolutionarily conserved biological modulators, with a recognized role in the central control of sex maturation, reproduction and fertility. Consequently, the focus on the central role of the Kisspeptins has led to neglecting their possible activities in peripheral tissues. Increasing data reveals that Kisspeptins and KISS1R have a wider expression and possibly a broader spectrum of action in several peripheral tissues such as the gonads, adipose tissue, and liver with direct consequences on gamete quality and fertility rate, pregnancy, energy homeostasis and body weight control. In this respect, the Kisspeptin system may represent a promising prognostic/diagnostic biomarker and therapeutic target for cancer and other human diseases such as infertility and metabolic disorders. The 29-amino acid peptide (named after its N-terminal glycine and C-terminal alanine) was identified in 1983 by its C-terminal amidation. This 'reverse' approach, that is to discover a substance through a distinct chemical feature, and only subsequently to characterize its biological activity, was novel and has been successful in the identification of several other peptides. After the structure of galanin was determined in 1983, functional studies were performed with material purified from natural sources until the synthetic form of the peptide became available. Galanin can act as transmitter, modulator and trophic factor, and is involved in a number of physiological processes such as hormone secretion, cardiovascular mechanisms, feeding and cognition. This peptide may also be of significance for a number of pathological processes/disorders including pain, depression, Alzheimer's disease, epilepsy, addiction and cancer. This wide diversity of actions is mediated by three galanin receptor subtypes. The studies reviewed in this volume give a fairly complete overview of the spectrum of the biological actions and functions of galanin and its receptors and on possible therapeutic applications in a number of pathological conditions.
