Book excerpt: In an attempt to isolate a temperature sensitive allele of ftsN, chemical random mutagenesis was performed using hydroxylamine. SMM 12 a temperature sensitive filamentous strain was successfully isolated. However, it did not carry an ftsN(Ts) allele. Transforming with different pasmids carrying cloned genes and random genomic plasmid libraries have yielded no complementation. Using P I -mediated transduction, the location of the mutation was found to be at the 88-89 minute region on the E. coli chromosome. This finding represents the discovery of an unprecedented gene at the 88- 89 minute region that blocks cell division. in addition, we report in this work the isolation of the first temperature-sensitive mutant that is suppressed by L-glutamate. Although the precise identity of the mutated gene remains illusive, all data produced by this work point out that we may have isolated a previously unknown player in the cell- division protein machinery in E. coli.

This book was released on 2003 with total page 156 pages. Available in PDF, EPUB and Kindle. Book excerpt: Temperature-sensitive (Ts) mutants are considered a valuable tool to study the functional properties of many essential proteins in Escherichia coli. These include protein-protein interactions, DNA-protein interactions, protein localization and the characterization of functional domains in proteins. Using P I-mediated localized mutagenesis, we were able to generate a number of Ts mutants by targeting genes at the 54.4 minutes on Escherichia coli chromosome. One of which, termed LS3533 was isolated for further testing. This 54.4 minutes locus on the Escherichia coli chromosome carries two essential genes that are involved in cell cycle. The genes are: zipA (a cell division gene) and ligA (encoding DNA ligase gene). Characterization of the mutant gene was carried out by two steps. First, through mapping the mutation using P-I transductions and second by complementing the Ts phenotype (observed at 42 C) through transforming the mutant strain with a plasmid that carries a wild type copy of the affected gene. Parallel to the identification process, the isolated strain was subjected to several physiological studies including phenotypic observation, chromosome patterns visualization, FtsZ localization and SOS response testing. Our findings demonstrated that the temperature-sensitive phenotype in isolate Ls3533 was due to a mutation in the ligA gene. The block to division at 42 C in this strain was shown to be a consequence of the induction of the SOS-response in a lexA-dependent but sfiA-independent fashion, in which the localization of the essential division protein FtsZ, to the division site is prohibited.
