It is the end result of several years of cooperative work between scientists of three countries: the United States, Germany, and Portugal. The first two chapters of this book are devoted to provide a clear, understandable explanation of the theoretical basis for chlorophyll fluorescence analysis. The definitions and terminology that are specific to chlorophyll fluorescence analysis are included in this discussion, which leads to the bridging of chlorophyll fluorescence analysis to plant tissue condition status. Subsequent chapters focus on the monitoring of stress in the natural terrestrial and aquatic environments, assessing seedling quality in forestry, and postharvest quality in fruits and vegetables. A final chapter is devoted to a newly emerging use for the technique in plant breeding programs. It is the hope of the editors and authors that readers who have not used this technique will be encouraged to explore the possibilities in their area of study. Porphyrins (also referred to as tetrapyrroles) are essential for life in the biosphere. Chlorophyll catalyzes the conversion of solar energy to chemical energy via the process of photosynthesis. Organic life in the biosphere is made possible by consumption of the chemical energy generated by photosynthesis. Hemes are the prosthetic groups of cytochromes which are involved in electron transport during oxidative phosphorylation and photosynthetic phosphorylation which generate ATP and NADPH. The latter are essential for many cellular functions. Chlorophyll on the other hands catalyzes the process of photosynthesis. Indeed, life in the biosphere depends on the process of photosynthesis which converts light energy, carbon dioxide and water into the chemical energy, required for the formation of food and fiber. Photosynthetic efficiency is controlled by extrinsic factors such as the availability of water, CO2, inorganic nutrients, ambient temperature and the metabolic and developmental state of the plant, as well as by intrinsic factors (Lien and San Pietro, 1975). The most important intrinsic factor is the efficiency of the photosynthetic electron transport system (PETS). Conventional agriculture is one of the few human activities that have not undergone a revolution to join other activities such as overcoming gravity by flying, and landing on the moon, crossing underwater the polar cap, and communicating wirelessly over long distances via electromagnetic waves. We now feel that enough biochemical and molecular biological knowledge has accumulated to render this dream amenable to experimentation. We believe that the time has come to bioengineer chloroplasts capable of synthesizing a short chain carbohydrate such as glycerol at rates that approach the upper theoretical limits of photosynthesis [Rebeiz, C. In order to achieve this goal a thorough knowledge of the Chl biosynthetic pathway is needed along with knowledge in other domains (Rebeiz 2010). In this context, this monograph is devoted to an in depth discussion of our present knowledge of the Chl biosynthetic pathway. The complexity and biochemical heterogeneity of the Chl biosynthetic pathway and the relationship of this complexity to the structural and biosynthetic complexity of photosynthetic membranes will be emphasized. We will also emphasize in historical perspective, key stages in our understanding of the Chl biosynthetic heterogeneity. The reader should keep in mind that a complex biosynthetic process is only fully understood when it becomes possible to reconstitute in vitro every step of the process. We are not yet at this stage of understanding of thylakoid membrane biogenesis. Book excerpt: As editor of the two-part Volume V on photosynthesis in RUHLAND'S Encyclopedia, the forerunner of this series published in 1960, I have been approached by the editors of the present volume to provide a short preface. The justification for following this suggestion lies in the great changes which have been taking place in biology in the two decades between these publications, changes which are reflected in the new editorial plan. Twenty years ago it appeared convenient and formally easy to consider photo synthesis as a clearly separated field of research, which could be dealt with under two major headings: one presenting primarily photochemical and biochemical prin ciples, the other physiological and environmental studies. Such a partition, however, as far as aims and opinions of the authors were concerned, resulted in a rather heterogeneous volume. Today, the tendency in experimental biology is towards a merger of previously distinct disciplines. Biochemists and biophysicists have developed their methods to such an extent that, over and above the analysis of individual reaction sequences, work on the manifold interrelationships among cellular activities has become in creasingly possible. Joining them in growing numbers are the physiologists and ecologists with their wealth of information on activity changes in vivo and on the variability and efficiency of the organisms concerned. Furthermore, biochemists, biophysicists and physiologists also now share a lively interest in ultrastructure research, the results and implications of which, through continually improving methodology, have generated important stimuli for the work in the field of cell function.

Book excerpt: Results of in situ fluorescence investigations on chlorophyll-a (chl-a) of phytoplankton in the Sea of Azov (May to June 2018) and in the Don River estuary (September to November 2019) are presented. Continuous data series of chl-a fluorescence were obtained with flow-through fluorometers. Discrete reference measurements of chl-a concentration and phytoplankton biomass were performed by standard methods for the sake of comparison and eventual corrections. The fluorescence intensity values measured in a lateral surface salinity gradient of the Sea of Azov were found correlating with the obtained data on the chl-a concentration (R2 = 0.88, n = 27) and phytoplankton biomass (R2 = 0.90, n = 11). Instead, there was a weak correlation (R2 = 0.40, n = 33) between the fluorescence and phytoplankton biomass found in the estuary. This disparity in correlations was explained by the difference in conditions during measurements, which affected the fluorescence. There were no significant changes in both temperature and coenotic composition of phytoplankton in the course of the marine expedition. The measurements on the river covered a period characterized by the seasonal variations in the phytoplankton composition and by noticeable temperature fluctuations.
