Book excerpt: Advances in the Biosciences 6: Schering Symposium on Intrinsic and Extrinsic Factors in Early Mammalian Development, Venice, April 20 to 23, 1970 is a collection of papers presented at the Schering Symposium on Intrinsic and Extrinsic Factors in Early Mammalian Development, held in Venice, Italy, on April 20-23, 1970. Contributors explore intrinsic and extrinsic mechanisms underlying early development in mammals and cover topics ranging from transmission of maternal proteins into oocytes to normal and abnormal fertilization in mammals; experimental early parthenogenesis in mammals; and nutrient requirements for the culture of preimplantation embryos in vitro. This book is comprised of 33 chapters and begins with an overview of the oocyte and the egg, touching on subjects such as electron microscopy of the primary and secondary oocyte; experimental early parthenogenesis in mammals; and laparoscopic recovery of pre-ovulatory human oocytes after priming of the ovaries with gonadotrophins. The next section is devoted to intrinsic and extrinsic influence on the metabolism of preimplantation embryos and includes chapters dealing with the composition of oviductal and uterine fluids; the role of uterine proteins in embryonic development; sex chromosome markers as indicators in embryonic development; and manipulations of the blastocyst. The remaining chapters examine placental and fetal physiology, immunology and teratology, and differentiation of tissues. This monograph will be of interest to biologists and physiologists.

Book excerpt: It is two years since a general meeting of the Gesellschaft fur Biologische Chemie first requested us to organize the 21 st Mosbach Colloquium on mammalian reproduction, and one year since we received final authorization to do so. The present volume contains the papers read at the Colloquium, but the discussions have been omitted because writing and proof reading them would have delayed the appearance of this volume for an unjustifiable long time. Besides, in most cases the discussion was of a relatively specific nature and we did not consider it essential, bearing in mind that the purpose of the Mosbach Col loquia is to provide advanced further education for the non specialist. One of us has referred to this and to the topical structure of the 21 st Colloquium in the introductory and final remarks. Chapter 1 describes gene transfer by micro injection of cloned recombinant DNA into zygotes. Although the factors that affect transformation frequencies and integration sites are still unknown, such techniques offer a number of exciting prospects. Research models for human disease coula be artificially created and desirable characteristics in agricultural animals could be - hanced. . The theme of cell-to-cell transfer is continued in Chapters 2 and 3. Chapter 2 describes pronuclear transplantation by Sendai virus-induced fusion of the karyoplast with the enucleated embryo. Using this procedure, it has been dem onstrated that both male and female genomes are essential for normal develop ment, although the reason for this is not yet understood. Chapter 3 describes studies on the fusion of whole oocytes. .

Book excerpt: With a few notable exceptions, mammalian preimplantation embryos grown in vitro are likely to exhibit sub-optimal or retarded development. This may be manifested in different ways, depending on the species and on the stage(s) of embryonic development that are being examined. For example, bovine embryos often experience difficulty in cleaving under in vitro conditions, and usually cease development at about the 8-cell stage (Wright and Bondioli, 1981). The block to development is stage-dependent; embryos cultured for 24 hr from the I-cell stage are much more capable of developing into viable blastocysts after transfer to oviducts than embryos cultured for 24 hr from the 4-cell stage prior to transfer (Eyestone et oZ. , 1985). Similar problems with in vitro embryo development are encountered in other species. Pig embryos can be grown up to the 4-cell stage in vitro but usually no further (Davis and Day, 1978). In the golden hamster, in the rat and in many outbred strains of mice, development of zygotes in vitro is blocked at the 2-cell stage (Yanagimachi and Chang, 1964; Whittingham, 1975). Even with some inbred mouse strains, embryo development is reduced if very early cleavage stages are used as the starting point for in vitro culture (Spielmann et oZ. , 1980). A common finding is that embryos grown in vitro have reduced cell counts (Harlow and Quinn, 1982; Kane, 1985) and their viability is reduced (Bowman and McLaren, 1970; Papaioannou and Ebert, 1986) compared to equivalent developmental stages recovered from mated animals.

The term developmental biology is really a new name for embryology; it is, however, used to denote the molecular approach to the study of developing systems. In this book we have tried wherever possible to blend the older information of classical embryology and in particular organogeny with the newer concepts of developmental biology. The original intention was to cover all the tissues of the body in this book. However, it soon became obvious that it was not possible to do this within one volume. Therefore we decided to have two general chapters, one on the basic concepts of cellular development and an other on the ageing of cells (this being considered part of the normal growth process). In addition to these two general chapters we have included chapters on some of the major tissues. These were chosen not just to illustrate the points made in the general chapters but because there is enough information available on the development of these tissues for the expert in that field to present a good, readable account. It is hoped that at a later date when more information is available, we will be able to extend this work, probably as several volumes, and to include the other tissues of the body which are not dealt with in this volume.
