Intramolecular cyclization strategies of donor-acceptor (D-A) cyclopropanes and alkylidene malonate monoamides have excellent potential for synthesis as they offer easy access to structurally-diverse compounds. The work described in this thesis accesses the scope of the In(OTf)3-catalyzed cyclization reaction of cyclopropanes and alkylidene malonate monoamides. In(OTf)3-catalyzed reactions of alkenyl and heteroaryl cyclopropyl ketones were examined in the synthesis of functionalized cyclohexenone-based derivatives (Chapter 2). Subsequent efforts to utilize a tandem cyclopropane ring-opening/Friedel-Crafts alkylation sequence of methyl 1-(1H-indolecarbonyl)-1-cyclopropanecarboxylates to prepare functionalized hydropyrido[1,2-a]indole-6(7H)-ones is discussed in Chapter 3. The extension of this tandem protocol towards the total synthesis of (±)-deethyleburnamonine is the subject of Chapter 6. Intramolecular Friedel-Crafts alkylation of N-indolyl alkylidene malonate monoamides was also examined. An In(OTf)3-catalyzed cyclization of substituted methyl 2-(1H-indole-1-carbonyl) acrylates afforded a series of 1H-pyrrolo[1,2-a]indole-3(2H)-ones (Chapter 4), whereas substrates with the indole 2-position blocked provided access to substituted 4H-pyrrolo[3,2,1-ij]quinolin-4-ones (Chapter 5).

Book excerpt: Organisms across all domains of life have developed elegant strategies to produce specialized metabolites, also referred to as natural products, for defense, structure, and communication. Natural products provide novel inspiration for pharmaceutical development given their potent biological activities and structural diversity. Recent advancements in DNA sequencing technology and the development of predictive bioinformatics tools have unveiled a vast collection of biosynthetic machinery responsible for generating this structural diversity that may be harnessed by synthetic chemists. Many of the enzymes encoded by biosynthetic genes catalyze chemical reactions that are difficult to replicate with the same selectivity by traditional synthetic methods. An example of this discrepancy is illustrated by the current enzymatic and chemical methods known to generate a highly reactive ortho-quinone methide (o-QM) intermediate. The utility of o-QMs in total synthesis has been hampered by complications that surround the preparation of their precursors, the harsh generation methods, and poor chemo-, regio-, and stereoselective control. In contrast, multiple enzymes have been reported to catalyze o-QM formation under mild conditions with remarkable selectivity. Chapter 1 of this dissertation examines several different enzymatic strategies developed to generate o-QMs. Chapter 2 describes progress towards the biomimetic total synthesis of (-)-chlorizidine A that aims to synthetically replicate the final biosynthetic intramolecular cyclization via an o-QM intermediate. Chapter 3 reports the discovery of tetrachlorizine, a novel tetrachlorinated marine natural product, and the identification of its associated biosynthetic gene cluster. Biochemical characterization of a pivotal flavin-dependent oxidase revealed a dehydrogenation reaction that proceeds via an unprecedented o-QM mechanism. This oxidase was then repurposed to generate an isolable o-QM, an extremely rare chemical motif. Chapter 4 investigates the structure-function relationships of two homologous flavin-dependent oxidases with divergent reaction selectivities and highlights their favorable biocatalytic properties. This chapter also discloses the first reported bacterial enzymes capable of performing oxidative cyclization reactions to produce cannabinoids.

Book excerpt: Abstract: An efficient approach to the synthesis of bicyclo [3.3.1] nonane-1, 3, 5-trione core of polycyclic polyprenylated acylphloroglucinol natural products (PPAPs) has been developed previously in our laboratory. The key transformation involves base-mediated dearomatization-annulation of a fully substituted acyl phloroglucinol. Application of this key transformation has been successful for the total synthesis of type B PPAPs 7- epi -clusianone.Synthesis efforts towards the type A PPAPs, including the adamantane-bearing natural product plukenetione A were evaluated. Initial attempts involved adapting the similar methodology for the previous synthesis of 7-epi -clusianone and this method successfully constructed the target molecules' core framework. Alternative approaches for plukenetione A synthesis were evaluated and a new method involving tandem base-mediated dearomatization and acid catalyzed annulation of acyl phloroglucinol was identified. This key methodology along with modified retro-aldol condensation Grignard addition and stereoselective cyclization led to the successful synthesis of type A adamantane plukenetione A.We have also developed a unified strategy for the synthesis of the type A PPAP 7-epi -nemorosone. Beginning with the same adamantane intermediate obtained in the plukenetione A synthesis, the natural product 7-epi -nemorosone was prepared in ten chemical steps. Preliminary evaluation of oxidative cyclizations of 7-epi -nemorosone to produce polycyclic structure has been conducted.
