Book excerpt: The distribution in Oregon of two viruses affecting salmonid fishes was determined by examination of hatchery and feral fish. Infectious pancreatic necrosis virus (IPNV) was isolated from fish at the Oregon Department of Fish and Wildlife Research Laboratory, Corvallis, Roaring River, Fort Klamath, and Wizard Falls Hatcheries. Fish collected at Elk, Cache, and Sparks Lakes were also found to be infected with IPNV. An epizootic caused by IPNV occurred at Wizard Falls Hatchery in 1974. Infectious hematopoietic necrosis virus (IHNV) was isolated from fish at Round Butte, Wizard Falls, and Elk River Hatcheries, and the Upper Metolius River. Epizootics due to IHNV occurred at Round Butte and Wizard Falls Hatcheries in 1975. The fish dying from IHNV at Wizard Falls Hatchery were also infected with IPNV, the first reported case of double virus infection in fish. The isolation of IHNV at Elk River Hatchery extended the range of the fish viruses into the Oregon coastal waterways for the first time. The occurrence of IPNV at hatcheries was controlled by destruction of infected stocks, sanitation of facilities and identification of a source of virus-free eggs. After sanitation the hatcheries were restocked with virus-free eggs and fish. The infection of hatchery fish by IHNV was not controlled due to its presence in trout and salmon brood stocks and in feral fish in waters above the hatcheries. Isolates of IPNV from sites in Oregon were compared serologically. Determination of 100% endpoint titers were found to be too variable for accurate comparisons; 50% endpoint titers were less variable and were used in this study. Only one serotype of IPNV was found in Oregon, but considerable antigenic variation was seen. There were two antigenic subtypes among the IPNV isolates examined: a Willamette Valley subtype and a Deschutes River subtype. The viruses isolated from the 1974 and 1975 Wizard Falls Hatchery IPNV epizootics were found to be antigenically dissimilar, indicating a separate source of infection for each epizootic. The 1975 epizootic had as its source eggs from Roaring River Hatchery broodstock. The origin of the virus in the 1974 epizootic was not determined. The source of infection of the 1973 Fall River Hatchery epizootic was shown to be eggs taken from virus-infected Eastern brook trout in Elk Lake.

Infectious pancreatic necrosis virus (IPNV) was isolated and identified from asymptomatic adult coho (Oncorhynchus kisutch) and chinook (0. tshawytscha) salmon returning to the Columbia River system. Infectious pancreatic necrosis virus was also identified as the causative agent of severe losses of eastern brook trout juveniles (Salvelinus fontinalis) at the Fall River trout hatchery in central Oregon. This was the first documented IPNV epizootic in fish from Oregon. A high incidence of IPNV was detected in eastern brook trout populations from central Oregon indicating that this virus poses a threat to the rearing of that species in Oregon. An attenuated strain of infectious hematopoietic necrosis virus (IHNV) was developed for use as a water-borne vaccine for the control of IHNV disease. This water-brone vaccine is easily administered as an immunizing agent by direct addition to the water containing fish. The attenuated IHNV water-borne vaccine strain was determined to be one hundred three times less virulent than the wild type strain from which it was derived. This vaccine was proven to be efficacious against high levels of both water route and injected challenges of virulent wild type virus in kokanee (0. nerka) and sockeye salmon (0. nerka). Other parameters for the development of an effective vaccine preparation were also tested. An optimal exposure period to the immunizing virus of 48 hours was determined as were the minimum immunizing dose for sockeye and chinook salmon. The attenuated IHNV water-borne vaccine produced immunity with a duration of at least 110 days and was shown to provide protection to sockeye salmon challenged with four isolates of virulent IHNV from different geographic locations. Another method of immunization, vacuum infiltration, was tested using a second strain of attenuated IHNV as an immunizing agent. Even though this method of immunization was effective in eliciting a protective immune response in chinook and kokanee salmon, it did not produce the level of protection provided by the attenuated IHNV water-borne vaccine.

The virus was isolated from landlocked Atlantic salmon (Salmo salar) and/or brook trout (Salvelinus fontinalis) at four of seven widely separated collection sites of different habitat types. The fish appeared to be only carriers of the virus since clinical signs of disease were absent in fish sampled. It was concluded that IPNV occurs naturally in salmonids of this drainage system. The consequences that this IPNV might have on policies of future enhancement programs and fish farming in the province are briefly discussed.
