The productive potential has been shown to increase in response to strategic supplementation programs. It has been speculated that production from growing and lactating cattle is limited by inadequate supply of metabolizable amino acids. Deficiencies are heightened when cattle are exposed to cold environments. Increasing metabolizable amino acids is commonly accomplished by implementing one of two supplementation regimens: (1) protein sources which escape ruminal digestion (UIP) or (2) ruminally protected amino acids (RPAA). Three experiments were conducted to evaluate the effects of increasing postruminal amino acid supply to cattle. In experiment 1, 30 lightweight heifers were used to determine the influence of supplemental soybean hulls plus UIP on intake, growth performance, and serum metabolites. Intake was not influenced by supplementation during cold ambient conditions, but growth performance was greatest for heifers offered UIP. Serum glucose, urea N, and non-esterified fatty acids were increased in heifers receiving supplemental UIP compared to those fed the isocaloric equivalent amount of soybean hulls. Six multi-cannulated Holstein steers were used in experiment 2 to evaluate the effects of the same supplements on site and extent of digestion at thermoneutrality and exposure to cold conditions. Fluid and particulate passage from the rumen increased during cold exposure, which did not affect microbial flow to the duodenum. Digestibility of N shifted from the small intestine to the large intestine in response to the cold environment, which allowed greater OM and NDF digestibilities in the lower and total tract of steers supplemented with UIP. From the results of these two experiments, it is suggested that addition of UIP to soybean hulls increased growth performance by maintaining nutrient digestion and enhancing metabolic status in the cold. Experiment 3 was conducted to determine how primiparous beef cattle respond to increasing level of postruminal methionine and lysine. Total milk protein exhibited a quadratic response with increasing level of RPAA, while cow weight gain yielded an inverse response to RPAA. Growth hormone, insulin, and insulin-like growth factor-I were not affected by dietary treatment, but did respond to changes in physiological state. Therefore, shifts in production away from body weight gain to increased milk production did not appear to be associated with changes in metabolic regulators.

Rumen bacteria and protozoa were analyzed for amino acid (AA) composition and digestibility, and values were incorporated into the Cornell Net Carbohydrate and Protein System (CNCPS). Two in vivo studies were performed to evaluate the effects of a commercial fermentation byproduct on nutrient digestion, microbial growth, omasal flow of nutrients, and milk production in lactating dairy cattle. The objectives of the first portion of the first study (Chapter 2) was to evaluate nutrient digestion in the rumen and total tract, along with omasal flows of N. Inclusion of fermentation byproduct decreased dietary N degradation in the rumen, with no detrimental effects on microbial N flows. The second part of the experiment (Chapter 3) involved measurment of microbial growth and turnover, partitioning of protozoa and bacterial N, and determination of omasal AA flow. Cows fed the fermentation byproduct had increased flows of non-microbial AA at the omasal canal. Comparison of the observed results against model predicted values indicated that the rumen sub-model of CNCPS v.7 was able to provide accurate estimations of total microbial N. The second trial (Chapter 4) was performed to evaluate the effects of the fermentation byproduct on milk production in groups of lactating cows. Cows fed fermentation byproduct had increased energy-corrected milk yield and protein yield compared with cows fed regular and rumen protected soybean meal. In Chapter 5, two methods not previously applied to microbial samples were used to characterize the AA profile and digestibility of bacterial and protozoal protein. The values obtained, along with other updates to protozoal growth and turnover parameters from Chapter 3 were incorporated into the CNCPS. A comparison of model predicted vs. observed value from a larger literature dataset was used to evaluate improvements in the model's predictive capability. Overall, the use of omasal sampling, improved isolation of protozoa, and more accurate determination of post ruminal flows of digestible AA allowed for a more complete understanding of microbial protein synthesis and more accurate model predictions of post ruminal AA supply.

Book excerpt: The beef industry is in a constant search for new feedstuffs to be used in the production process. This occurs for several reasons. Nutritional support of gain in growing beef cattle needs to, at minimum, match improvements realized in the genetic capability for growth. Feed costs constitute one of the largest beef production costs and economics has dictated that feed cost per kilogram of gain be reduced to remain competitive in the world market. Manufacturing of goods for human consumption has created numerous by-products with little economic value for human use. Dried distillers grains with solubles (DDGS), a by-product of ethanol production, represents a product which could be used as a replacement for soybean meal (SBM) in cattle rations. A national need for fuel-grade ethanol has driven dramatic increases in its production and subsequent increases in DDGS supply. The state of Missouri has been targeted as an area for development of ethanol production. The by-product of the process, DDGS, is high in protein, digestible fiber, and lipid content. This makes it a potential Nitrogen and energy source for ruminant rations. A series of experiments was conducted to evaluate the impact of DDGS as a protein and energy source in beef cattle. We compared DDGS and SBM in growing heifers consuming corn-silage, soyhull, and corn-based diets to evaluate the performance and cost-effectiveness of the two supplements. Similar dietary N levels were evaluated. Inclusion of DDGS in the diet resulted in increased final weight, ADG, DMI, and lower cost of gain as compared to heifers consuming SBM. Grazing is utilized in a large portion of Missouri cattle production, with summer generally requiring supplementation. A study was conducted which evaluated increasing levels of DDGS for heifers grazing tall fescue. Performance criteria were evaluated for animals receiving 0.45, 0.91, or 1.36 kg DDGS and a 0.1 kg soyhull negative control. Average daily gain was quadratically improved as level of DDGS in the diet was increased, with 0.91 kg DDGS providing the greatest ADG. Intake of forage DM and cost of gain were not affected by treatment. The final experiment examined the post-ruminal amino acid flow of mature steers consuming timothy hay and supplemented with DDGS, SBM, a SBM/DDGS blend, or negative control. Supplementation increased duodenal flow of total essential and non-essential AA, while Met, Leu, and Trp were linearly increased by greater inclusion of DDGS in the diet. Dried distillers grains with solubles appears to be a valid replacement of SBM in highforage growing diets.

Book excerpt: Amino acids (AAs) are not only building blocks of protein, but are also signalling molecules as well as regulators of gene expression and the protein phosphorylation cascade. Additionally, AAs are key precursors for syntheses of hormones and low-molecular weight nitrogenous substances with each having enormous biological importance. For example, physiological concentrations of AA metabolites (e.g., nitric oxide, polyamines, glutathione, taurine, thyroid hormones, and serotonin) are required for cell functions. Growing evidence shows that humans and animals have dietary requirements for all proteinogenic AAs. Mammals, birds and fish also have species- and age-dependent needs for some AA-related substances. However, elevated levels of other products (e.g., ammonia, homocysteine, H2S, and asymmetric dimethylarginine) are pathogenic factors for neurological disorders, oxidative stress, and cardiovascular disease. Thus, optimal amounts of AAs and their ratios in diets and circulation are crucial for whole body homeostasis and health. Adequate provision of one or a mixture of functional AAs or metabolites may be beneficial for ameliorating health problems at various stages of the life cycle (e.g., fetal growth restriction, neonatal morbidity and mortality, weaning-associated intestinal dysfunction and wasting syndrome, obesity, diabetes, cardiovascular disease, the metabolic syndrome, and infertility). Dietary supplementation of these nutrients can also optimize the efficiency of metabolic transformations to enhance muscle growth, milk production, and athletic performance, while preventing excess fat deposition and reducing adiposity. Therefore, functional AAs hold great promise in improving the growth, health and well-being of individuals.
