Book excerpt: These studies were undertaken in preparation for an extensive investigation of the association of cell-mediated immunity to chlamydial agents to clinical nongonococcal urethritis. Spleen cells from guinea pigs immunized with the meningopneumonitis strain of Chlamydia psittaci (MN) derived from infected chicken embryos were made to react with MN antigen derived from cell cultures. Lymphocytic blastogenic transformation, production of lymphotoxin, and of migration inhibition factor were demonstrated. The results indicate that these tests can be performed with considerable simplicity with chlamydial antigens. These studies were not applied to nongonococcal urethritis because of the untimely death of Dr. Gordon. (Author).

Book excerpt: It is almost axiomatic that an explosive increase in knowledge and interest concerning host immune defense mechanisms and the immune response system in general is occurring. Many new developments are evident at the interphase between the areas of immunology, infectious diseases, and host defenses to microorganisms. It is now widely rec ognized that a functioning host immune defense system consists of a variety of lymphoid cells. These include effector and affector cells consisting of both Band T cells and their progeny. Mononuclear phago cytes which are widely distributed throughout the body are also intimately involved in these processes. These cells are involved not only in the clearance of particulate matter including bacteria, viruses and fungi, but also presumably in host defense against neo plasia. Among the many triumphs of the biological revolution over the past decade or so is the rapidly developing understanding of how these cells and their products, including antibody, lymphokines, monokines, etc. , function and interact. It is now recognized that although the immune defense system is involved in defense against infectious agents, it is also now be coming quite clear that many infectious agents, including bacteria, viruses and fungi, as well as their products, can interact with the immune response system. For example, it has been long recognized that many of the "adjuvants" which enhance the immune responses in nonspecific ways are derived from microorganisms. Microbial products have marked effects on the immume response system.

Book excerpt: Staphylococcus aureus infection is not associated with the development of protective immunity, and disease relapses occur frequently. We hypothesize that protein A, a factor that binds immunoglobulin Fc[gamma] and cross-links VH3 clan B cell receptors (IgM), is the staphylococcal determinant for host immune suppression. To test this, vertebrate IgM was examined for protein A cross-linking. High VH3 binding activity occurred with human and guinea immunoglobulin, whereas mouse and rabbit immunoglobulins displayed little and no binding, respectively. Establishing a guinea pig model of S. aureus bloodstream infection, we show that protein A functions as a virulence determinant and suppresses host B cell responses. Immunization with SpAKKAA, which cannot bind immunoglobulin, elicits neutralizing antibodies that enable guinea pigs to develop protective immunity. Importance: Staphylococcus aureus is the leading cause of soft tissue and bloodstream infections; however, a vaccine with clinical efficacy is not available. Using mice to model staphylococcal infection, earlier work identified protective antigens; however, corresponding human clinical trials did not reach their endpoints. We show that B cell receptor (IgM) cross-linking by protein A is an important immune evasion strategy of S. aureus that can be monitored in a guinea pig model of bloodstream infection. Further, immunization with nontoxigenic protein A enables infected guinea pigs to elicit antibody responses that are protective against S. aureus. Thus, the guinea pig model may support preclinical development of staphylococcal vaccines.

Direct macrophage migration inhibition was observed in cultured peritoneal exudate cells as early as 3 days after exposure. Maximum inhibition of macrophages cultured with phase I or II antigen occurred 14 to 21 days post-exposure and persisted through 35 days. This inhibitory action was no longer detectable at 42 days. Serum antibody to the phase II antigen of C. burnetii was detected at 14 days and to phase I antigen at 21 days, 18 days after the cell-mediated immune response. (Author).

Book excerpt: The ability of a killed phase I Coxiella burnetii vaccine to induce cell-mediated immune (CMI) responses in guinea pigs was studied. Cell-mediated immune responses were assessed by the inhibition of macrophage migration (IMM) and lymphocyte transformation (LT) assays. The IMM response occurred rapidly, was detected at high levels, but was relatively short-lived. In contrast, the LT response developed more slowly, and persisted for a longer period. The vaccine given in a single dose, or two doses one week apart, protected guinea pigs from a subsequent virulent challenge.
