Book Microbial DNA and Host Immunity

Download or read book Microbial DNA and Host Immunity written by Eyal Raz and published by . This book was released on 2002-09-26 with total page 428 pages. Available in PDF, EPUB and Kindle. Book excerpt: The immunostimulatory prospects of bacterial DNA have attracted the interest and attention of scientists and physicians and become a major focus in immunobiology and biomedicine. These activities are the product of immunostimulatory DNA sequences (ISS, also known as GpG motifs), which are rare in the mammalian genome. ISS were shown to enhance immunological responses and were used to confer protection to a wide variety of tumors, allergic inflammation, and infections. In Microbial DNA and Host Immunity, leading researchers review the activation of the mammalian immune system by bacterial DNA and consider the applications of ISS in clinical medicine. The authors survey the latest findings concerning the receptor-recognition and signaling pathways triggered by ISS, the process of cell activation, and potential vaccination strategies using ISS. Specific pharmaceutical applications discussed include infectious disease (Hepatitis B, HIV, and mycobacterial infections), allergy (asthma and conjunctivitis), cancer (lymphoma), and inflammation and autoimmunity (arthritis and colitis). Up-to-date and informative, Microbial DNA and Host Immunity illuminates the immunobiology of bacterial DNA and its promise of powerful new vaccines to provide protective immunity against infections, tumors, and chronic disease.

Book Immunobiology of Bacterial CpG DNA

Download or read book Immunobiology of Bacterial CpG DNA written by H. Wagner and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 249 pages. Available in PDF, EPUB and Kindle. Book excerpt: Bacterial CpG-DNA sequences not only serve for genetic information but act as "danger" signal to alert immune cells such as macrophages and dendritic cells. CpG-DNA directly causes activation of these members of the innate immune system yielding in up-regulation of co-stimulatory molecules and pro-inflammatory cytokines. As such they act as "natural" adjuvants to promote protective Th1 polarized T cell immune responses. This book is the first comprehensive review of the data emerging in this field.

Download or read book DNA Vaccines written by Douglas B. Lowrie and published by Springer Science & Business Media. This book was released on 2008-02-01 with total page 530 pages. Available in PDF, EPUB and Kindle. Book excerpt: The field of DNA vaccines has undergone explosive growth in the last few years. As usual, some historical precursors of this approach can be d- cerned in the scientific literature of the last decades. However, the present state of affairs appears to date from observations made discreetly in 1988 by Wolff, Malone, Felgner, and colleagues, which were described in a 1989 patent and published in 1990. Quite surprisingly, they showed that genes carried by pure plasmid DNA and injected in a saline solution, hence the epithet “naked DNA,” could be taken up and expressed by skeletal muscle cells with a low but reproducible frequency. Such a simple methodology was sure to spawn many applications. In a separate and important line of experimentation, Tang, De Vit, and Johnston announced in 1992 that it was indeed possible to obtain humoral immune responses against proteins encoded by DNA delivered to the skin by a biolistic device, which has colloquially become known as the “gene gun. ” The year 1993 saw the publication of further improvements in the me- ods of naked DNA delivery and, above all, the first demonstrations by several groups of the induction of humoral and cytotoxic immune responses to viral antigens expressed from injected plasmid DNA. In some cases, protection against challenge with the pathogen was obtained. The latter result was - questionably the touchstone of a method of vaccination worthy of the name.
