These have laid the foundations of a new generation of vaccines exemplified by subunit vaccines produced through gene cloning and by synthetic peptides mimicking small regions of proteins on the outer coat of viruses. Such peptide~ are capable of eliciting virus-neutralizing antibodies. Unfortunately, subunit and peptide vaccines are only weakly or non immunogenic in the absence of immunological adjuvants that are known to augment specific cell-mediated immune responses to the antigens and to promote the formation of protective antibodies. This book contains the proceedings of the 4th NATO Advanced Studies Institute (ASI) "Vaccines: New Generation Immunological Adjuvants" held at Cape Sounion Beach, Greece, during 24 June -5 . July 1994 and deals in depth with both theoretical and practical aspects of vaccinology. These include the role of antigen presenting cells in the induction of immune responses. immunopotentiation by a variety of new generation immunological adjuvants and vaccine carriers. and recent advances and perspectives in experimental vaccines as well as vaccinatioll with nucleic acids. Within months, it spread worldwide from its “birthplace” in Guangdong Province, China, affecting over 8,000 people in 25 countries and territories across ?ve continents. SARS exposed the vulnerability of our modern globalised world to the spread of a new emerging infection. SARS (or a similar new emerging disease) could neither have spread so rapidly nor had such a great global impact even 50 years ago, and arguably, it was itself a product of our global inter-connectedness. Increasing af?uence and a demand for wild-game as exotic food led to the development of large trade of live animal and game animal markets where many species of wild and domestic animals were co-housed, providing the ideal opportunities for inter-species tra- mission of viruses and other microbes. Once such a virus jumped species and attacked humans, the increased human mobility allowed the virus the opportunity for rapid spread. An infected patient from Guangdong who stayed for one day at a hotel in Hong Kong led to the transmission of the disease to 16 other guests who travelled on to seed outbreaks of the disease in Toronto, Singapore, and Vietnam, as well as within Hong Kong itself. The virus exploited the practices used in modern intensive care of patients with severe respiratory disease and the weakness in infection control practices within our health care systems to cause outbreaks within hospitals, further amplifying the spread of the disease. Health-care itself has become a two-edged sword.

Book excerpt: In his introduction to the Ciba Foundation Symposium on "Immunopotentia tion," Sir Peter Medawar stated that, "for the past twenty years the control of the immune response has been virtually equated to immunosuppression because the great goal of applied immunology has been the transplantation of tissues," and that, "with the discovery of tumor immunity-the focal point of immuno logical control has changed from immunosuppression to immunpotentiation and correspondingly the great prize of applied immunology has become the preven tion and control of malignant growth." Corynebacterium parvum is now univer sally recognized as being the most potent presently known immunopotentiator. C parvum is at the center of almost all papers presented at this conference and it has been analyzed from various viewpoints: taxonomy, pharmacology, immunopotentiation, stimulation of host resistance to tumor invasion in experi mental models and in human therapeutics. Almost all data reported in this volume are original and recently obtained. The discussions were spontaneous, stimulating, and highly enriching, and the results already established and re ported here are in many respects unique. Discussions at the conference revealed many other aspects which require further exploration. Nonspecific immunopotentiation has strongly penetrated into immunology, immunopathology, and, recently, oncology. Undoubtedly, this area of research will witness important developments in the near future. This book offers the first synthesis of presently available knowledge and emphasizes future prospects.
