As our work progressed, the rigidity of basic, previous embryological principles was broken down as scientific knowledge advanced. At the same time, the molecular, biological characterization of the cell surface receptor systems progressed enormously with the invention of NUMEROUS monoclonal antibodies. Thus, thymology became once again very important because the thymus is the first and central organ of the human immunological system. Then, the question of immuno-neuroendocrine regulation arose and has only been partially answered. Our book seeks to explore what has not been explored. The topic of thymic epithelial cells is a unique one and has never been explored in any previous book as it is explored in this one. Only a handful of great thymologists remain in the world today, especially after the great loss the medical community suffered with the passing of Dr. Good, the list includes but is not limited to: Dr. Ritter and Dr. Kendall in England, Dr. Savino in Brazil, Dr. Dardenne in France, Dr. von Gaudecker in Germany, a few others in Belgium and Holland, and it is our hope that Dr. Bodey is among them. Nonetheless, a book on the thymus has not been written in the last five years and a book such as this one has never been. This book is based on a 30-year period of research and includes references from a broad range of sources spanning the globe and all sources, even those that were the beginning of thymic research. The book, thus, is uniquely well rounded, more so that previous works.

There should be specificity, so that tumour cells alone are destroyed whilst normal tissues are unaffected. Provided the tumour is uni form and all of the cells have appropriate antigens, every malignant cell should be destroyed and even distant metastases dealt with. So far these speculative advantages are unfulfilled and the initial optimism that surrounded im munotherapy has not been sustained. Acceptance of the precepts of tumour im munology continues but these disappointing observations had led to increasing scrutiny of certain aspects. The purpose of this chapter is to review the prin ciples which underly tumour immunology and immunotherapy, so that the more detailed studies that follow can be considered in perspective. TUMOUR ANTIGENS (Chapter 2) For a tumour to initiate an immunological response, it must possess distinctive antigens. Much of the early work in tumour immunity was confused because it was not appreciated that tumours, like other tissues, exhibit transplan tation antigens. Only when syngeneic tumours are used can tumour antigens alone be studied and it was the introduction of inbred mouse strains which allowed Foley in 1953' to produce the first evidence for specific an tigenicity of experimental tumours. Demonstration of these antigens requires that pretreatment with syngeneic tumour will influence the growth of a sub sequent challenge with the same neoplastic cells.

As a result of technologic developments and new insights in the realm of molecular biology, new and important approaches to an understanding of neoplasia are now possible. Several aspects of neoplasia are clearly of microbiologic interest, including the role of viruses in the etiology of cancer, control of the immune response to tumor cells, and the susceptibility of tumor-bearing hosts to overwhelming infection as a result of immuno deficiency. Recent advances in these areas led us to organize this symposium, and, through this publication, to record some of the progress being made in laboratories around the world in understanding some of the basic aspects of the cancer problem. This symposium was held as part of the commemoration of the twentieth anniversary of the Waksman Institute of Microbiology. Dr. Waksman's devo tion to the study of the smallest forms of life and the commitment of the Waksman Institute to the free pursuit of knowledge are the underpinnings of the institute's research eHorts in the broad area of microbiology, including the problem of neoplasia. It is of interest to note that actinomycin, one of the earliest antibiotics discovered in Waksman's laboratory, was also one of the first compounds found to be clinically useful in the treatment of certain types of cancer.

An antigen of cross-reacting specificity was revealed in tissue extracts of many other animal species. We next wanted to learn more about the antigen responsible for induction of postoncolytic immunity. Extracts from virus-infected tumors were immunogenic, and both active and inactive fractions of such extracts were obtained. The growth of the virus in the tumor cells was studied with the electron microscope, in the hope that this might shed some light on the manner in which viral infection transforms a poorly immunogenic tumor into a highly immunogenic one. We consider none of the questions which our work has raised as definitely solved. In fact, we are still working on many of the aspects alluded to above. When we embarked on a study of postoncolytic immunity, we were supported in this endeavor by our lack of experience in the fields of transplantation and tumor immunology.
