These were usually based on the physical or chemical characteristics of the substances to be measured. However in recent years there has been an explosion of interest in analytical methods which made use of the high specificity and sensitivity of im munological reactions. These methods can be very simple in terms of technical procedures and can usually be performed on minute samples of biological fluids - factors which have ensured their ready acceptance in most laboratories. Recently there have been numerous meetings on technical aspects of particular immunoassays and on their application in specific diseases. We felt however that the time was ripe for an 'overview' of the whole field. To this end a conference on 'Immunoassays for the 80s' was held at the Zoological Society of London in 1980, and this book is largely based on that meeting. Both the immunoassay techniques and their numerous applications were discussed and are dealt with at length in this volume. The editors wish to thank all the contributors for their chapters and to acknowledge the debt they owe to Jean Ryan (NLCM) without whose organization and assistance this volume would not have been completed. The most widely used immunoassay technique is radioimmunoassay (RIA) which was first developed by Yalow and Berson in 1959. The principle of RIA is elegantly simple. It utilizes a competitve binding reaction between analytes and a radio-labeled analog of the analytes (the tracer) for anti-analyte antibodies. In addition to its exquisite specificity, extraordinary sensitivity, good accuracy and precision, ease and rapidity of assay and simplicity of assay development, the applicability of RIA to a wide variety of substances has made it one of the most powerful and versatile analytical methods of the 20th century and beyond. Millions of RIA's are being performed annually on clinical, biological and environmental samples in licensed laboratories. In order to expand the use of RIA beyond the confines of these laboratories to areas like physician's offices, patients' homes, economically less developed countries, agricultural fields, large scale and continuing screening tests for infectious diseases, it has become necessary to develop non-isotopic labels. Indeed the last fifteen years have seen the development of a great number of ingenious non-isotopic labels in immunoassay so that a whole new industry capitalizing on the potential market for non isotopic immunoassays has appeared. It is the purpose of this volume to present in depth, state-of-the-art reviews on techniques used in non-isotopic immunoassays. Current methods have 1 2 arisen from the pioneering work ofYalow and Berson in the U. S. A. and Ekins in the u. K. Much of the early development was concerned with the analysis of protein hormones, and nearly a decade passed before attention focussed also on small molecules such as steroids and drugs. The potential of immunoassay methods for drug monitoring in clinical and forensic laboratories and in addict treatment programmes resulted in the commercial production of immunoassays for various therapeutic and abused drugs, making the technique available to laboratories lacking the facilities to raise their own antisera and synthesise labelled compounds. However, commercial assays are not only expensive but are restricted in range, and so it is advantageous for a forensic laboratory to have the capability to devise "in-house" immunoassays suited to its particular requirements. This chapter describes the theory and practice of RIA in forensic drug analysis. Much of the theory and some of the practice are applicable to immunoassays in which non isotopic labels are used, but such assays are not described in detail since, to date, the versatility and sensitivity of RIA have made it the immunoassay technique of choice in forensic toxicology. The particular advantages of RIA are its sensitivity and the fact that samples such as haemolysed blood can be assayed with little or no prior preparation.

The development of methods in environmental virology became a focus of growing interest about two decades ago. Progress has been significant since that time in pure experimental systems, where there are no interferences, consistent high recoveries of viruses from environmental waters has been achievable for some time. In the natural environment, however, in relatively clean waters, substances such as humic and fulvic acids interfere with viral recoveries and average recovery rates probably do not reach 20%. With sewage sludges and shellfish, recoveries are undoubtedly much lower. Yet, even relatively low viral recovery rates have made possible the detection of viral hazards in drinking waters. The hazards that exist are undoubtedly much greater than those demonstrated with the relatively inefficient methods inefficient methods developed thus far. Improving methods, as they are developed in the years to come, will undoubtedly bring the true extent of the hazards into better perspective.
