Book Host Interactions in HIV Infection

Download or read book Host Interactions in HIV Infection written by Chris Stinson and published by . This book was released on 2015-03-23 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: There is no vaccine or cure available for HIV; hence it is a promintent global health threat. To beat the challenges of HIV/AIDS, one must have sufficient knowledge of HIV infection. This book discusses various forms of HIV-host interactions in vitro and in vivo, like the interrelation between cellular components and HIV proteins. Integrase, Tat, and Nef, as well as the clinical pertinence of HIV superinfection. The book also examines the function of innate immunity along with dendritic cells and defensins in HIV infections and the effects of host factors on HIV pathogenesis. Topics like co-infection are also included that talk about the influence of Human herpesvirus 6 and Trichomonas vaginalis on HIV infection. The book also includes a discussion on the generations of HIV molecular clones that can be applied in macaques and the potential usage of cotton rats for HIV studies.

Book excerpt: In Chapter five of this dissertation, I showed that LY6E showed a distinct phenotype in low-CD4-expresisng Jurkat cells, i.e., LY6E inhibits HIV-1 entry by downregulating CD4. LY6E was found to be co-localized with CD4 on plasma membranes and further down-modulated CD4 on cell surface; this leads to compromised viral binding and subsequent viral entry. Ectopic expression of CD4 was able to rescue the phenotype of LY6E in Jurkat cells. The CD4 dependent inhibitory phenotype of LY6E can be recapitulated in low-CD4 primary cells human Monocyte-derived macrophages (MDMs). Taken together, this study reveals that LY6E likely modulates HIV-1 infection in a cell type dependent manner, and the CD4 surface expression level determines the sensitivity of HIV to LY6E. Overall, in this thesis work, I have investigated the role of IFITMs and LY6E in HIV-1 infection and discovered some multifaceted functions of these two families of proteins in modulating the different stages of the viral life cycle. My data highlight the more complex functions of type I IFNs and ISGs in HIV infection and AIDS pathogenesis. Information obtained in this thesis provides insights into understanding of the modes of action of type I IFNs and virus-host interaction in general, and could have therapeutic implications for prevention and treatment of HIV/AIDS.

Book HIV Interactions with Host Cell Proteins

Download or read book HIV Interactions with Host Cell Proteins written by Paul Spearman and published by Springer Science & Business Media. This book was released on 2009-12-11 with total page 210 pages. Available in PDF, EPUB and Kindle. Book excerpt: The study of viruses necessarily involves dissecting the intimate details of cellular pathways. Viruses have often been employed as tools in studying cellular pathways, as was done by early retrovirologists such as Peyton Rous in attempting to understand the mechanism of cellular transformation and oncogenesis. On the other side of the coin, virologists seek to de?ne those cellular elements interacting intimatelywiththeir virus ofinterestinorder to better understand viral replication itself, and in some cases to develop antiviral strategies. It is in the intersection of virology and cell biology that many of us ?nd the most rewarding aspects of our research. When a new discovery yields insights into basic cellular mechanisms and presents new targets for int- vention to ?ght a serious pathogen, the impact can be high and the excitement intense. HIV has been no exception to the rule that viruses reveal many basic aspects of cellular biology. In recent years, in part because of the importance of HIV as a major cause of human suffering, numerous cellular processes have been elucidated through work on processes or proteins of this human retrovirus. The excitement in this ?eld is especially well illustrated by the discovery of new innate means of resisting viral replication, such as the work on APOBEC3G, TRIM5a, and BST-2/ tetherin presented in this volume.
