Book excerpt: Hormones, Lipoproteins and Atherosclerosis, Volume 35, emerged from the international symposium on ""Hormones, Lipoproteins and Atherosclerosis"" held in Bratislava, Czechoslovakia. The symposium was devoted to the problems of hormonal effects on lipoprotein metabolism and atherosclerosis, and was also concerned with problems of composition, structure, synthesis, and degradation of liporoteins, as well as with genetical and clinical aspects. The participants of the symposium have contributed to this volume not only by presenting new data but also new ideas and stimulating new trends in the research and therapy of metabolic derangements of lipoproteins and the resulting pathological conditions. The 51 contributions are organized into five parts. Part I includes papers on the metabolism of lipoproteins. Part II presents studies on hormones and lipoproteins. Part III is devoted to atherosclerosis and lipoproteins. Part IV takes up the clinical aspects of hyperlipoproteinemias. Part V on methods contains two studies—the first on the isolation of human plasma high density lipoproteins; the second on the separation of subfractions of the serum triacylglycerols.

Book excerpt: As the acute infectious diseases recede in importance, and as the number of people surviving into middle and old age increases, the chronic 'degenerative' diseases assume greater importance as causes of death and disability. Of these diseases, atherosclerosis is by far the most prevalent and its consequences the most devastating. The search for the cause of atherosclerosis is consuming large amounts of resources of both money and research talent. As yet, the cause remains unknown. Much of the research effort into atherosclerosis has been concerned with lipid metabolism. This is based on the knowledge that abnormalities of certain lipids and lipoproteins predispose to cardiovascular disease. Often the research has not been directly related to atherosclerosis and it is only recently that widespread attention has been paid to the artery. The development of methods of growing vascular endothelial and smooth muscle cells in culture has made possible detailed studies of the biology of the arterial wall. There are a number of reasons why investigations of lipid metabolism alone will not identify the cause of atherosclerosis. First, only a minority of patients with cardiovascular disease have abnormal circulating lipids and lipoproteins. Second, there are three major predisposing factors for atherosclerosis which cannot be entirely explained by abnormal lipid metabolism - age, sex, and diabetes mellitus. Third, it is now clear that lipid is only one component of the atheromatous plaque, and incorporation of lipid may be a late feature of the development of the lesion.

The present book summarizes data discussed at an International Meet ing at Marburg, Germany, on "Hormones in Lipoprotein Metabolism". It was an attempt to pull together knowledge in basic science from the mode of action of hormones all the way to clinical appearance of hor monal disorders involving lipoprotein metabolism. Thus data on molec ular biology, on hormonal regulation of apolipoprotein synthesis, on lipoprotein receptors and enzyme induction are discussed together with the large field of oral contraceptive use, postmenopausal estrogen sub stitution, lipid disorders in diabetes mellitus, in thyroid dysfunction, in adrenal insufficiency and in glucocorticoid application. We are aware of the fact that such an overview cannot be complete but should serve as a collection of data and ideas for those interested in hormonal regulation of lipid metabolism. We hope that this volume of edited papers, a consequence of the third international colloquium on atherosclerosis, conducted at the University of Brussels, Belgium through the support of the "Fondation de Recherche sur l' AtherosclE,rose" will contribute to this goal. Among the topics discussed the major ones were the mechanism of action of lipolytic enzymes, the deficiency or dysregulation of cellular receptors, the genetic deficiencies of apolipoproteins, and the panoply of external factors as diet, physical exercise, drugs, which mOdify the lipoprotein metabolism. Special interest was also devoted to potent techniques as kinetic analysis of metabolic tracers and use of monoclonal antibodies. Their contribution to the detection and treatment of atherosclerosis will be obviously essential in the future.
