Book excerpt: The concept that hormones influence tumor growth originated in 1889 with the proposal of Albert Schinzinger who suggested that breast cancer is related to the ovaries. Several years later, Sir George Beatson observed that remission of disseminated breast cancer could be achieved in premenopausal patients by performing bilateral oophorectomy. As a result of the contri butions of Hedley Atkins, Charles Huggins and others, additive and ablative hormonal therapies have been widely used for the treatment of advanced breast cancers for several decades. Model systems to study the effects of hormones on growth and regression of breast tumors have been available for many years; however, the complexities of the hormonal environment have rendered in vivo studies difficult in man and experimental animals. Recently, the availability of long-term cultures of breast cancer cells has stimulated many investigators to use these cell lines to unravel the mechanisms of hormone action. Because of the extreme diversity and complexity of advances regarding the endocrinology of the breast and breast cancers, a multi-authored review was deemed necessary. It has been gratifying to receive contributions from many noted scholars. In Volume I of this monograph, the influence of steroid hormones and their antagonists upon normal and neoplastic tissues of the mammary gland are presented. In Volume II, the effects of peptide and other hormones are reviewed.

Book excerpt: One of the principal objectives of our laboratory is to understand the mechanisms by which hormones and growth factors regulate normal mammary gland development and how these same regulatory pathways become altered in breast cancer. Specific emphasis has been placed upon studying the mechanisms by which the lactogenic hormones, prolactin, hydrocortisone and insulin, regulate milk protein gene expression. The CCAAT/enhancer binding protein B (c/EBPB), Yin Yang-i (YY- 1), signal transducers and activators of transcription S (STATS), and the glucocorticoid receptor (OR) all have been shown to mediate the hormonal and developmental regulation of B-casein gene expression. However, the mechanism by which these transcription factors as well as coactivator proteins coordinately function to promote normal mammary gland development and breast cancer remains undefined. To understand the role of C/EBPj3, STATS, OR and comodulatory factors in hormonally-regulated chromatin remodeling at the 3-casein promoter and enhancer, we have initiated chromatin immunoprecipitation assays in HCl 1 cells and in the mammary gland and liver from lactating mice. In addition, we have analyzed mammary gland development in several specific mouse knockouts. The examination of mammary transplants from OR* and CBP mice suggested that MR may compensate for the loss of OR in mammary gland development during pregnancy, lactation and involution, and that haploinsufficiency of CBP does not appear to affect mammary gland development.

Book excerpt: Changes in signal transduction networks occur during mammary tumor progression that affect the proliferative response of the cells to exogenous factors. These alterations may be related to the loss of hormonal regulation of breast cancer which has a major impact on prognosis and therapy of breast cancer. The goal of this research was to identity the mechanisms underlying the difference in the proliferative response to 3'-5' cyclic adenosine monophosphate (cAMP) that we observed in our rodent model systems (Imagawa et aL 1992). cAMP is a potent mitogen for normal mammary gland epithelium, weakly nitogenic for hormone-dependent mammary tumors and growth inhibitory to hormone-independent mammary tumors. cAMP-stimulated proliferation was inhibited by pertussis toxin (PT) in normal mammary epithelium but neither the inhibitory effect of cAMP nor basal cAMP-independent proliferation of hormone-independent mammary tumors was affected by PT (Imagawa et al. 1995). PT is a bacterial toxin that ADP-ribosylates G(Delta)i(Beta)y subunits and blocks activation of receptor-coupled heterotrimeric G(Delta)i(Beta)y proteins.
