Available in PDF, EPUB and Kindle. Book excerpt: Electron diffraction data and high resolution images can now be used to obtain accurate, three-dimensional density maps of biological macromolecules. These density maps can be interpreted by building an atomic-resolution model of the structure into the experimental density. The Cowley-Moodie formalism of dynamical diffraction theory has been used to validate the use of kinematic diffraction theory, strictly the weak phase object approximation, in producing such 3-D density maps. Further improvements in the preparation of very flat specimens and in the retention of diffraction to a resolution of 0.2 nm or better could result in electron crystallography becoming as important a technique as x-ray crystallography currently is for the field of structural molecular biology.

Book excerpt: Ultra high resolution X-ray crystallography allows for analyzing the charge distribution in the molecules and provides methods to study the intermolecular interactions at a deeper level. Structural studies of several thiophene based compounds have been carried out and the phenomenon of disorder has been discussed. Experimental and theoretical charge density analysis of two important molecules was performed using a multipolar atom model. A new virtual atom model is also tested which allows for a rapid calculation of the electrostatic properties. The hydrogen bonding with oxygen atom acceptor is studied through an extensive survey of more than 500,000 crystal structures. The stereo chemical results are compared with the electron density of the oxygen atoms in different chemical environments which give conclusive evidences for the dependence of directionality of hydrogen bonds on the shape and orientation of the electrons lone pairs. Finally, it has been shown that how in the absence of high resolution X-rays data, principle of transferability of electron density parameters between molecules can be used to study the electrostatic properties and the intermolecular interactions. This principle has been successfully applied to a small thiophene based molecule and the large FAD binding protein Cholesterol oxidase.

Book excerpt: Maximum Entropy (ME) techniques have found widespread applicability in the reconstruction of incomplete or noisy data. These techniques have been applied in many areas of data analysis including imaging, spectroscopy, and scattering [Gull and Skilling, 1984]. The techniques have proven particularly useful in astronomy [Narayan and Nityanada, 1984]. In many of these applications the goal of the reconstruction is the detection of point objects against a noisy background. In this work we investigate the applicability of ME techniques to data sets which have strong components which are periodic in space or time. The specific interest in our laboratory is High Resolution Electron Micrographs of beam sensitive materials. However, ME techniques are of general interest for all types of data. These data mayor may not have a spatial or temporal character. Figure 1 shows an HREM image of the rigid-rod polymer poly(paraphenylene benzobisoxazole) (PBZO). The 0.55 nm spacings in the image correspond to the lateral close-packing between the extended polymer molecules. Near the center of this crystallite there is evidence for an edge dislocation. In HREM images both the frequency and position of the infonnation is important for a proper interpretation. Therefore, it is necessary to consider how image processing affects the fidelity of this information in both real and Fourier space.
