Liquid chromatography, being a powerful universal separation technique, is widely available in laboratories and easily hyphenated with other spectroscopic and biochemical techniques. A number of LC-based strategies—such as multicomponent quantitation, fingerprinting, bioaffinity chromatography, and on-flow assays for screening—have been used for the comprehensive QC of TCMs, lead finding, and identification of active constituents. In this chapter, the applications and development of such LC-based methods for TCMs and other herbal drugs are presented and discussed. Some future trends are given.

An analytical methodology is employed to detect the presence of Aristolochic Acid-1 in Chinese herbal medicine. 9 different samples were extracted and fingerprinted using HPLC-DAD.Data treatment was performed using SIMCA-P, a chemometric analytical software and the output obtained was transformed into a PLS-DA model which was capable of distinguishing between the samples which contains AA-I and does not. Hence, the study provides evidence of identifying the toxic AA-I in Chinese herbal medicines.

Having a good quality control system for Traditional Chinese Medicine ( TCM ) can ensure reproducibility of therapeutic effects from the same herbal mixture bought at different locations and can reduce the potential of drug interactions due to the presence of impurities in the decoctions. Nine herbs with analgesic effect were purchased from a large TCM shop in Shatin. These herbs were processed individually into decoctions and tested on mice for analgesia with the hot plate method. Fang Feng ( Divaricate Saposhnikovia ), Bai Zhi ( Bahurian Angelica ), Gui Zhi ( Cassiabarktree Twig ), Pu Huang ( Longbract Cattail Pollen ) and Ru Xiang ( Olibanum ) were found to have good analgesic effects and a dose-response curve was established for equal proportion of this herbal mixtures. Crude extracts of these herbal mixtures were separated into different molecular fractions using dialysis membranes at different molecular cut off ( MWCO ) points. 3These molecular fractions were tested on mice to identify the most effective molecular fraction ( biological fingerprint ). A wavelength scan from 210nm to 700nm for this molecular fraction was performed using a High Performance Liquid Chromatography system to obtain its chemical fingerprint. The same procedures were repeated on the same herbal mixtures purchased from another TCM shop in Jordon. The fingerprints obtained from the most effective molecular fraction of the herbal mixtures purchased in these two different TCM shops were compared for biological and chemical equivalences. Results suggested that they had similar quality and analgesic efficacy.
