During fibrogenesis, pathologic stimuli cause hepatocyte apoptosis, and these apoptotic bodies can be cleared by hepatic stellate cells (HSC) by phagocytosis. Upon phagocytosis, HSC, the key fibrogenic cells of the liver become activated and transdifferentiate into myofibroblasts, meanwhile produce extra amount of ECM molecules. Therefore HSC phagocytosis is a major event in the initial process of fibrosis. To study HSC phagocytosis in vivo, we have developed a mouse model, in which a lenti viral-green fluorescence protein expressing vector (LV-GFP) and an adeno viral-tumor necrosis factor-related apoptosis-inducing ligand (Ad-TRAIL) were applied to mice to induce hepatocyte apoptosis and to label the apoptotic bodies. Using this model, observing HSC phagocytosis and activation in vivo has been achieved. Galectin-3 (Gal3), a [beta]-galactoside-binding lectin, is a regulator of the phagocytic process and the expression of Gal3 is related to fibrotic diseases. In this study, we have focused on the mechanism by which Gal3 modulates HSC phagocytosis and activation. We found that HSC from Gal3[superscript -/-] mice displayed decreased phagocytic activity compared to wild type cells, and decreased expression of TGF-[beta] and procollagen [alpha] 1(I). This suppression was partially reversed by recombinant Gal3 suggesting that extracellular Gal3 is required for HSC activation by phagocytosis. HSC expressing wild type integrin [beta]3, but not binding deficient mutant, bind to recombinant Gal3, suggesting that Gal3 mediates HSC phagocytosis via crosslinking to this integrin and facilitating the tethering of apoptotic cells. Blocking integrin [alpha]v[beta]3 resulted in a decrease of HSC phagocytosis. On the other hand, we discovered that Gal3 expression and release were induced in HSC by phagocytosis, via the activation of nuclear factor kappa B (NF-[kappa]B) signaling. The upregulation of Gal3 in active HSC was further confirmed in vivo on the bile-duct ligated animals. Gal3[superscript -/-] mice displayed significantly decreased fibrosis, reduced expression of alpha smooth muscle actin ([alpha]SMA) and procollagen [apha] (I) upon the challenge of bile duct ligation (BDL). Consistently, using the phagocytosis model, we discovered that Gal3[superscript -/-] mice showed less HSC phagocytosis and fibrotic activity following the viral injection. In summary, Gal3 plays a key role in liver fibrosis by mediating HSC phagocytosis and activation in a feed-forward pattern.
