Book excerpt: The Major Histocompatibility Complex (MHC) was discovered as a con sequence of the chronic problem encountered by cancer biologists in the early years of this century: the failure to maintain tumor lines by serial passage in outbred mice. A number of observations pointed to genetic similarity being a prerequisite for successful transplantation and they were incorporated into a genetic theory of transplantation by C.C. Little. This prompted scientists like Little to initiate breeding experiments designed to test his hypothesis and produce genetically identical mice which would permit the growth of trans planted tumors. Most inbred strains of mice commonly used in immunology derive from those efforts. Transplantation of normal tissues obeyed the same rules found for malignant tissues and rejection was shown to be an immunological phenomenon. G.D. Snell showed that a single genetic locus determined rapid rejection of skin grafts. This was initially called the Major Histocompatibility Locus but was subsequently shown to include many functionally related genes and renamed the Major Histocompatibility Complex (MHC). Book excerpt: The Role of Products of the Histocompatibility Gene Complex in Immune Responses documents the proceedings of a conference held on 3-7 November 1975, which brought together an international group of scientists spanning three independent disciplines—genetics and immunogenetics, molecular biochemistry, and immunobiology—with clinical medicine overlapping these disciplines. This volume contains 42 papers organized according to the eight sessions held at the conference. The papers in Session I examined the genetics of the major histocompatibility complex. Session II presented studies on the biology of mixed lymphocyte interactions and cell-mediated cytotoxicity reactions. Session III discussed immune response gene systems while Session IV dealt with the genetic control of cell interactions. The papers in Session V covered idiotypic determinants on T cell receptors. Session VI investigated the properties of histocompatibility gene products involved in regulation of immune responses. Session VII focused on the biochemistry and immunocytology of cell surface products of the major histocompatibility complex. Finally, Session VIII discussed interrelationships between products of the major histocompatibility complex and their relevance to disease.

Book excerpt: After the discovery of the function of MHC molecules, namely to provide the context for T cell recognition of foreign antigens, in 1974 Zinkernagel and Doherty made the first drawing of MHC+X (Fig. 1 from Zinkernagel and Doherty, Nature, 251: 547, 1974). Over the next 18 years a very large number of similar drawings ensued, some of real artistic beauty. One side of the problem, the nature of the T cell receptor, was unraveled; however, we still do not know exactly what kind of a structure the T cell receptor recognizes, al though in 1987 we learned so much about the structure of MHC molecules and antigen presentation. In schematic presentations no one is now placing the foreign antigen beside the MHC molecule, but rather on top of it, as pointed out by J. L. Strominger at the MHC + X meeting in Paris. The complex of MHC and antigen is named MHC + X, but the precise meaning of this formula remains a "problem perplex", as illustrated in these proceedings by Peter Par ham. When planning the Ommen/Amsterdam meeting at the begin ning of 1987, its major aim was seen as to discuss the question of whether MHC + X can induce antibodies and, consequently, their specificity. In other terms, whether - in analogy to antigen specific MHC restricted T cells - MHC restricted antibodies also exist.
