Book Biological Reactive Intermediates III

Download or read book Biological Reactive Intermediates III written by James J. Kocsis and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 1028 pages. Available in PDF, EPUB and Kindle. Book excerpt: This volume contains the proceedings of the third in a series of conferences entitled, The International Symposium on Biological Reactive Intermediates. The first was held at the University of Turku in Finland, in 1975, the second at the University of Surrey in the United Kingdom, in 1980 and the most recent at the University of Maryland in the United States, in 1985. The significance of these conferences has been emphasized by the rapid growth of mechanistic toxicology over the last decade. These conferences were initially stimulated by the attempt to uncover the significance behind the observations that the toxicity of carcinogenic responses produced by many chemicals was associated with the observation that their metabolism led to the formation of chemcially reactive electrophiles which covalently bound to nucleophilic sites in cells such as proteins, nucleic acid or fats. Recently, newer concepts have arisen which have necessitated the expansion of subjects covered by the conference. For example, the application of newer knowledge of the role of active oxygen species in reactive metabolite formation, the concept of suicide substrates, examination of the function of glutathione in cells, application of immunological techniques and molecular biological probes to the solution of toxicological problems all had an impact on the study of the biological reactive intermediates.

Book Distributed in space Glutathione Conjugation Kinetics of Ethacrynic Acid in Liver

Download or read book Distributed in space Glutathione Conjugation Kinetics of Ethacrynic Acid in Liver written by Rommel Giongco Tirona and published by . This book was released on 2000 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: The factors affecting the hepatocellular glutathione (GSH) conjugation of ethacrynic acid (EA), a model substrate, in the formation of the glutathione adduct EA-SG were assessed for their influence on local GSH conjugation rates from the inlet to the outlet of the liver. These included plasma protein binding, sinusoidal uptake, spontaneous and enzyme-catalyzed conjugation, co-substrate (GSH) availability and EA-SG efflux at canalicular and sinusoidal membranes and their zonation within the acinus. The spontaneous coupling of EA with GSH was rapid, whereas the 'in vitro' conjugation catalyzed by the glutathione 'S'-transferases (GSTs) in rat liver was even faster. The conjugation rates greatly exceeded the EA uptake rate by saturable and non-saturable processes in isolated rat hepatocytes. When these ' in vitro' data were viewed with respect to events in whole liver (single-pass perfused rat liver), transport and not GSH conjugation was found to be rate-limiting in the rapid conversion of EA to EA-SG. In studies with isolated, enriched periportal and perivenous hepatocytes, EA uptake and EA-SG efflux were functionally similar among hepatocytes throughout the acinus. However, a shallow and increasing (portal to venous) GST activity gradient was observed in rat liver. When these zonal, uptake and metabolic 'in vitro' data were utilized to predict EA disposition in liver perfusions in the presence and absence of bovine serum albumin (BSA), a series compartment model was found to be most consistent with data obtained in the absence of albumin. The model predicted both a temporal and zonal depletion of GSH among hepatocytes and a change in the rate-limiting step with increasing perfusion time from uptake to co-substrate availability. The model was less capable of predicting the influence of high albumin protein binding on uptake and metabolism, unless a role of BSA in facilitating drug uptake was assumed. This investigation showed for the first time that, in addition to metabolic enzymes, flow and substrate binding, other important factors such as zonal transport, co-substrate, and GSH conjugate efflux further influence the distributed-in-space processing of acceptor substrates in GSH conjugation.

Book Structure and Function of Glutathione S Transferases

Download or read book Structure and Function of Glutathione S Transferases written by Kenneth D. Tew and published by CRC Press. This book was released on 1993-08-26 with total page 354 pages. Available in PDF, EPUB and Kindle. Book excerpt: Structure and Function of Glutathione S-Transferases provides some of the latest information available on a variety of structural and functional components of glutathione S-transferases, a family of isozymes involved in many endogenous and exogenous functions in cells. Molecular studies presented in the book focus on the regulation of these enzymes and identify important response elements. X-ray crystallographic structures show detailed information on the structural aspects of these proteins. Metabolism of a number of carcinogens and drugs is covered in detail, and the role that these enzymes play in governing drug resistance at the preclinical and clinical levels is discussed. The book will be excellent for biochemists, pharmacologists, oncologists, experimental therapeutic specialists, and others interested in glutathione S-transferases.
