Book excerpt: In this research, a recombinant inbred line (RIL) population derived from cross between a leaf rust- and stripe rust-susceptible spring wheat 'Avocet S' and a slow leaf- and stripe-rusting resistant spring wheat 'Amadina' was used to postulate and map leaf rust seedling resistance genes, identify quantitative trait loci (QTL) for slow-rusting resistance against leaf and stripe rust, and study slow leaf-rusting components, latent period and infection frequency. Two known Lr genes (Lr23, and Lr26) were identified to be present in 'Amadina' through gene postulation, pedigree, cytogenetic, and polymerase chain reaction analyses. One unknown gene associated with seedling resistance was also mapped on chromosome 1BL. In greenhouse experiment, it was estimated that at least five genes conditioning final disease severity (FS) and latent period (LP), and four genes conditioning infection frequency (IF), segregated in the population. Correlations between LP and FS, and LP and IF were moderately negative, and that between IF and FS was moderately positive, indicating inter-dependence of the traits. Two QTL on chromosomes 1BL and 6BL were associated with LP and FS, and three QTL on chromosomes 1BL, 6BL and 2DS were associated with IF. Segregation of the RIL population in field experiment indicated that there were at least four and three adult plant resistance (APR) genes involved in resistance for leaf and stripe rust. Six QTL on chromosomes 3AL, 4AL, 1BL, 5BL, and 7BL were associated with APR for leaf rust, and seven QTL on chromosome 4AL, 5AL, 1BL, 2BL, 4BL, 5BL, 2DL, and 4D were associated with APR for stripe rust. Our results indicated that the major portion of genetic variability for slow-rusting resistance was additive gene action, and, to some extent, epistasis. In this research, we also explored the utility of remote sensing and geographic information systems (GIS) and analytical operations to discriminate leaf rust pustules from other parts of leaf and to accurately determine pustule size in 'Amadina' and 'Avocet S'.

I was then invited by Dr. Watson to share in its writing. Unfortunately, shortly there after, recurring heart problems forced him to curtail his activities and with draw from the project. He died before the book could be completed. Dr. Watson's intention was to produce a very practical book that would provide wheat breeders with all of the information necessary to breed success fully for resistance to the three wheat rusts: leaf rust, stem rust, and yellow rust. It was intended to be very specific in describing procedures to be used and at the same time provide all of the necessary theoretical background. I hope that I have been successful in meeting these objectives. The book assumes that the reader has some knowledge of plant pathology, genetics, and plant breeding. Extensive use has been made of the literature, but it was not possible to cite all of the papers on a given topic. In making a choice, an attempt was made to choose key papers or more recent papers that provided references to the earlier literature. Acknowledgements This book was written partly at the University of Saskatchewan and partly at the University of California, Davis, while I was on a sabbatical leave.

Book excerpt: Wheat leaf rust, stem rust, and stripe rust are major fungal diseases of wheat caused by Puccinia triticina, P. graminis f. sp. tritici, and P. striiformis f. sp. tritici, respectively. Development of wheat varieties with genetic resistance to pathogens is a common strategy to protect against significant yield loss. However, incorporating resistance is not always straightforward, and understanding the mechanisms responsible for triggering host resistance is key. Here, were characterized two different genetic mechanisms involved in the rust resistance defense response in wheat. Two spring wheat cultivars and their respective EMS mutants, which displayed enhanced resistance to leaf, stem, and/or stripe rust, were studied. The cultivar Canthatch possesses a stem rust resistance suppressor on chromosome 7DL. We characterized the spectrum of resistance of Canthatch and the mutant CTH-NS, confirming the specificity of this suppressor is stem rust-specific. The cultivar Alpowa mutant MNR220 displays broad-spectrum resistance to a number of wheat rust races at the seedling stage. We characterized the functional resistance of MNR220 to a race of leaf rust by sampling inoculated tissues at various time points and tracking the disease progress microscopically. The mutant MNR220 confers resistance at the pre-haustorial stage; formation of haustoria is delayed or inhibited in MNR220 relative to the wild type Alpowa.
