Merr.] cultivars with [Less than or equal to] 80 mg seed−1 are used to produce grain for export to Japan for the production of natto, a fermented food. The purpose of the study was to compare three population types for the recovery of lines that would have adequately small seed size for natto. Two small-seeded and one normal-size conventional cultivar or line were used to produce a small-seeded x small-seeded two-parent population, a small-seeded x normal-size two-parent population, and a small-seeded x (small-seeded x normal-size) three-parent population. Five sets of the three population types were developed with different parents in each set. The seed size of 100 random F2 plants from each of the 15 populations and 1O plants of each parent was determined. For each of the five sets, the progeny of the 100 F2 plants of each population type were compared as F2-derived lines with 10 entries of each of the three parents at two Iowa locations. The average percentage of lines with a seed size equal to or smaller than one of the parents in a cross was 90% for the small-seeded x small-seeded populations, 4% for the small-seeded x normal-size populations, and 20% for the three-parent populations. An average of 10% of the lines from the small-seeded x small-seeded populations had significantly smaller seed size than either of the parents, and no transgressive segregation for small seed size was observed in the other two population types. For the development of small-seeded cultivars, small-seeded x small-seeded and three-parent populations would provide lines with acceptable seed size. A small-seeded x normal-size population may provide suitable lines if seed size of the conventional cultivar is sufficiently small and adequate resources are available to select for the limited number of small-seeded segregates.

Merr.] cultivars with large seed size and high protein content are desirable for the production of tofu and other food products. The objectives of this study were to evaluate the amount of genetic variability and the effectiveness of single plant selection for seed size and protein in two- and three-parent soybean populations. Two parents with large seed and high protein (LSHP) and one parent with normal seed size and protein (N) were used to produce three population types: LSHP x N, LSHP x LSHP, and LSHP x (LSHP x N). Four sets of the three population types were evaluated with different parents in each set. For each of the populations, 100 random F2 plants and 10 plants of each parent were harvested at Ames, IA, and their seed size and protein measured. The F3 progeny of the 100 F2 plants and 10 entries of the three parents of each set were evaluated in replicated tests at two Iowa locations. The percentage of F[Subscript 2:3] lines with seed size equal to the smallest LSHP parent and protein equal to the lowest LSHP parent in the set averaged 4% for the LSHP x N, 88% for the LSHP x LSHP, and 3l% for the three-parent crosses. Single-plant selection was not considered cost effective in the LSHP x N populations because the percentage of acceptable segregates was so small or in the LSHP x LSHP populations because the percentage of acceptable segregates was so high. For the three-parent populations, single-plant selection was most effective when the F2 plants were ranked for the two traits and those with the most favorable rank were selected for evaluation as F[Subscript 2:3] lines. LSHP x LSHP and three-parent populations should be the most useful for developing LSHP cultivars.

Book excerpt: Prior to reproductive development, the rate of leaf appearance is relatively uniform for the main-stems of soybean genotypes evaluated under unstressed field experiments. A recent discovery of a soybean breeding line with faster leaf appearance demonstrates the existence of phenotypic variation for this trait, which is supported by recent functional genomics studies that have identified a number of genes that regulate the rate of leaf appearance in other plant species. Subsequent characterization revealed that the genotype with faster node appearance requires approximately 0.5 fewer days to produce a new node on the main-stem under irrigated field conditions. Soybean yield potential is theoretically dependent upon the generation of nodes as a source of additional flowers and leaf biomass that support seed production. An increased rate of node accrual could benefit yield potential through earlier canopy closure, greater seasonal light interception, and increased harvest index. Overall, characterization of the faster node accrual trait will help to improve understanding of the molecular mechanisms controlling the rate of leaf appearance and to evaluate the importance of node production as it pertains to the development of elite, high-yielding soybean cultivars.

Book excerpt: Abstract: Food-grade soybeans are a specialty crop with unique chemical and physical seed quality requirements. The tofu production market requires large, round soybean seed with a clear hilum and high protein content. The aim of this project was to detect QTL responsible for traits important in food-grade soybeans, and to provide information for the selection of food-grade soybeans in a breeding program. This study assessed seed shape, size, density, weight, protein content, and oil content in two independent populations of breeding lines and cultivars as well as textural traits of the tofu produced from a subset of those. By identifying the relationship between seed traits and tofu texture, the previously established positive correlation between seed protein content and the stiffness and gel strength of tofu were confirmed. Little to no correlations between seed size and shape measurements and the texture of tofu were detected, indicating that this consumer preference for large, round seeds may not relate to tofu quality. The effects of selection for specialty traits on the genetic differentiation within a breeding program were assessed, results shown that both parental and progeny selection contributed to genetic differentiation and population structure. Association mapping was conducted with 504 markers in a population of 242 breeding lines and cultivars, leading to the identification of 50 significant marker-trait associations. Of the 27 significant associations tested in another independent confirmation population consisting of 152 lines or cultivars, 12 were confirmed. These results can be directly applied to increase selection efficiency in a breeding program.
