Book excerpt: One of the ways to make consistent progress in a particular field of biology consists in choosing a good model system on which to focus the experimental efforts of the scientific community. It has taken a long time for scientists interested in various aspects of the life of plants to reach some sort of consensus. With the advent and impact of molecular biology, the small weed Arabidopsis is now the object of rapidly growing scientific attention. Since it is reasonable to assume that the general molecular mechanisms that are responsible for the physiological, cellular and biochemical properties of plants will be essentially conserved in all plants, it follows that these mechanisms should also operate in Arabidopsis and hence that its genome should contain most of the genes that we need to know about if we want to understand the genetic determination of the life processes in plants.Arabidopsis has a small genome and well documented genetic studies are available. It is easy to grow in large numbers and mutants defining important genetically controlled mechanisms are either available, or can readily be obtained. Various methods to introduce and express isolated homologous or heterologous genes are available. It is therefore realistic and desirable to aim at exploring the genome of this plant in very great detail. As will be illustrated in this book all the elements for such a grand strategy are in place.More and more scientists are therefore willing to accept the obvious and very real practical disadvantages resulting from its small size when experiments call for the isolation of proteins, membranes, subcellular fractions etc, in order to benefit from its extraordinary experimental advantages as a model system in molecular genetics. One can safely predict that in the next decade studies with Arabidopsis will provide major breakthroughs in our understanding of most aspects of plant physiology and developmental biology. The importance of this knowledge for plant breeding and therefore for a sustainable highly productive agriculture cannot be overestimated. Our aim in choosing the topics and contributors was to demonstrate common approaches to studies of fungal-plant and fungal-animal interactions, particularly at the biochemical and molecular Ievels. For example, the initial events of adh«sion of fungal spores to the exposed surface tissues of the host are essential for subsequent invasion of the plant or animal and establishment of pathogenesis. A point of consensus among investigators who have directed their attention to such events in plants, insects, and vertebrates isthat spore adhesion to the host cuticle or epithelium is more than a simple binding event. lt is a complex and potentially pivotal process in fungal-plant interactions which "may involve the secretion of ftuids that prepare the infection court for the development of morphological stages of the germling" and subsequent invasion of the host (Nicholson and Epstein, Chapter 1). The attachment of the fungal propagule to the arthropod cuticle is also "mediated by the chemical components present on the outer layer of the spore wall and the epicuticle . . . . Initial attachment may be reinforced further by either the active secretion of adhesive materials or the modification of spore wall materiallocated at the [fungal spore arthropod] cuticle interface (Boucias and Pendland, Chapter 5).

Topics discussed include the history, biology and genetic diversity of Moniliophthora species (which cause witches’ broom and frosty pod rot) and Phytophthora species (which cause black pod rot) that cause diseases resulting in major losses to cacao production. Emerging pathogens such as Cacao swollen shoot virus and Ceratobasidium theobromae (which causes vascular streak dieback) are also discussed in detail, along with many pathogens of significant local concern. Most of these pathogens represent major risks to global cacao production should they expand into new areas, breaking out of their current limited distributions. By considering cacao diseases as a group, similarities in the available tools and techniques used in their management become apparent, as do their limitations. Gaps in our current knowledge of cacao pathogens and the management of the diseases they cause are detailed, and suggestions for future research directions are provided. This insight allows readers to consider cacao disease threats from a more comprehensive, global perspective and paves the way for an improved synergy of efforts between the various research programs, agencies, and industries, both private and public, with vested interests in cacao production, and cacao farmers.

The demand for increased food production needs to be met from ever reducing resources of land, water and other environmental constraints. Rice remains the staple food source for a majority of the global populations, but especially in Asia where ninety percent of rice is grown and consumed. Climate change continues to impose abiotic and biotic stresses that curtail rice quality and yields. Researchers have been challenged to provide innovative solutions to maintain, or even increase, rice production. Amongst them, the ‘green super rice’ breeding strategy has been successful for leading the development and release of multiple abiotic and biotic stress tolerant rice varieties. Recent advances in plant molecular biology and biotechnologies have led to the identification of stress responsive genes and signaling pathways, which open up new paradigms to augment rice productivity. Accordingly, transcription factors, protein kinases and enzymes for generating protective metabolites and proteins all contribute to an intricate network of events that guard and maintain cellular integrity. In addition, various quantitative trait loci associated with elevated stress tolerance have been cloned, resulting in the detection of novel genes for biotic and abiotic stress resistance. Mechanistic understanding of the genetic basis of traits, such as N and P use, is allowing rice researchers to engineer nutrient-efficient rice varieties, which would result in higher yields with lower inputs. Likewise, the research in micronutrients biosynthesis opens doors to genetic engineering of metabolic pathways to enhance micronutrients production. With third generation sequencing techniques on the horizon, exciting progress can be expected to vastly improve molecular markers for gene-trait associations forecast with increasing accuracy. This book emphasizes on the areas of rice science that attempt to overcome the foremost limitations in rice production. Our intention is to highlight research advances in the fields of physiology, molecular breeding and genetics, with a special focus on increasing productivity, improving biotic and abiotic stress tolerance and nutritional quality of rice.

Book excerpt: The development of new plant varieties is a long and tedious process involving the generation of large seedling populations for the selection of the best individuals. While the ability of breeders to generate large populations is almost unlimited, the selection of these seedlings is the main factor limiting the generation of new cultivars. Molecular studies for the development of marker-assisted selection (MAS) strategies are particularly useful when the evaluation of the character is expensive, time-consuming, or with long juvenile periods. The papers published in the Special Issue “Plant Genetics and Molecular Breeding” report highly novel results and testable new models for the integrative analysis of genetic (phenotyping and transmission of agronomic characters), physiology (flowering, ripening, organ development), genomic (DNA regions responsible for the different agronomic characters), transcriptomic (gene expression analysis of the characters), proteomic (proteins and enzymes involved in the expression of the characters), metabolomic (secondary metabolites), and epigenetic (DNA methylation and histone modifications) approaches for the development of new MAS strategies. These molecular approaches together with an increasingly accurate phenotyping will facilitate the breeding of new climate-resilient varieties resistant to abiotic and biotic stress, with suitable productivity and quality, to extend the adaptation and viability of the current varieties.
