Book excerpt: Development and maintenance of the prostate is dependent on androgens and the androgen receptor. The androgen pathway continues to be important in prostate cancer. Here, we evaluated the transcriptome of prostate cancer cells in response to androgen using Long Serial Analysis of Gene Expression (L0ngSAGE) libraries. We identified 35 genes with novel associations to androgen signalling and validated 24 of these genes using quantitative real time-polymerase chain reaction (qRT-PCR). These genes were: ARL6IF5, BL VRB, C]9orf48, C]orfJ22, C6orf66, CAMK2NJ, CCNI, DERA, ERRFI], GLUL, GOLFH3, HMJ3, HSP9OB], MANEA, NANS, NIPSNAP3A, SLC4JA], SOD], SVIF, TAOK3, TCP], TMEM66, USP33, and VTAJ. The physiological relevance of these expression trends was evaluated in vivo using the LNCaP Hollow Fibre model. There is no cure for castration-recurrent prostate cancer (CRPC), and the mechanisms underlying the disease are not known. To address this problem, we assayed the transcriptome of LNCaP human prostate cancer cells as they progress to castration-recurrence in vivo using replicate L0ngSAGE libraries. We identified 96 novel genes consistently differentially expressed in CRPC. The expression profiles support a role for the transcriptional activity of the androgen receptor genes (CCNH, CUEDC2, FLNA, and FSMA 7), steroid synthesis and metabolism genes (DHCR24, DHRS7, ELOVL5, HSDJ 7B4, and OPRKJ), neuroendocrine cell genes (ENO2, MAOA, OPRK], SJOOA]O, and TRPM8), and proliferation genes (GAS5, GNB2L], MT-ND3, NKX3-], PCGEM], PTGFR, STEAFJ, and TMEM3OA) in castration-recurrence. Screening for prostate cancer using serum levels of prostate-specific antigen has resulted in the over-treatment of indolent disease. Novel diagnostic and prognostic markers for prostate cancer are required. To address this need, the levels of 27 transcripts were investigated with qRT-PCR. Inter vening Prouts Neck Meetings in 1986, 1987 and 1988 were devoted to treatment, image cytometry and clinical markers. As before, the Prouts Neck tradition of bringing together an international, multidiscipli nary group of experts for 3 days to exchange ideas and new data, in the relaxed atmosphere of an old iun on the scenic Maine coast, proved to be an ideal combination for a highly successful conference. Accordingly, the Organ System Program of the National Cancer Institute plans to use the Prouts Neck model for future conferences on other solid tumors (bladder in 1990 and kidney in 1991) and will return to the prostate in 1992. A new dimension was added to the current program through the inclusion of a poster session to recognize the research of pre-and postdoctoral investigators. The posters were judged by Drs. Collette Freeman, Frank French, Shutsung Liao, Robert Matusik and Henry Sun. The three winners, in alphabeti cal order, were John Fabian, Robert Getzenberg and Ming Fong-Lin.

The five-year patient survival ranges from nearly 100% for local and regional stage cancers to 28% once the cancer becomes distantly invasive. This suggests that the lethal and non-lethal prostate cancers are likely separate diseases and may possess different early expression profiles. We compared the gene expression of early prostate cancer cells of varying Gleason scores using transcriptome analysis and confirmed the expression of certain overexpressed genes with Western blotting. Of the approximately 40 differentially expressed genes found in our RNAseq study, 21 were singled out due to their pronounced (greater than 2.5-fold) over- or under- expression in other cancers. SERPINB2, PI3, SPRR2D, RRAD, S100P, SLPI, LCN2, EDN1, CLDN4, LAMP3, ANGPTL4, and COL1A2 were upregulated. SOX2, MELK, CENPF, TOP2A, CDC20, MCM3, DLGAP5, ANLN, and RRM2 were downregulated. The magnitude of gene up- or down-regulation, with the exception of COL1A2, were much lower in passaged PrCa. Lipocalin-2 protein was highly expressed by PrCa 87 and NEp 21 with PrCa 87 expressing four times the amount of NEp 21. Sox-2 protein was expressed highest in PrCa 109. SerpinB2 was expressed the highest in PrCa 87, then PrCa 109, then PrCa 76 while NEp 21 and NEp 83 had negligible amounts of SerpinB2. These results, if repeated in large numbers of prostate cancer short-term cultures, may facilitate the prevention of overtreatment due to uncertainty about lethality of the patient's prostate cancer.

Book excerpt: Our general objective is to use completely defined cell-free transcription systems both to identify novel AR-associated cofactors and to investigate the mechanism of action of AR and both novel and previously identified candidate coactivators. To this end, we have expressed and purified 5 AR-associated cofactors and established a chromatin assembly system to study AR function with chromatin template. We have shown an enhancement of AR function by the TRAP/Mediator coactivator complex that is generally utilized by a number of different nuclear receptors. Relevant to the mechanism involved, we have identified two TRAP/Mediator subunits as potential targets for the liganded AR receptor. The expression of AR and 10 cofactors by quantitative in situ RNA hybridization in 44 primary prostate cancers with different degree of differentiation was investigated. Our results revealed near constant expression of AR and heterogeneous expression of AR cofactors. Expression of PIAS1 and Ran/ARA24 was increased and expression of ELEl/ATRA70 was decreased. In addition, we demonstrated that the human prostate tumor cell proliferation and colony formation are markedly reduced by over-expression of ELEl/ARA70. Together, these findings indicate that the change of expression levels of AR cofactors may play important roles in prostate growth and tumorigenesis.
