Available in PDF, EPUB and Kindle. Book excerpt: Fusarium Head Blight (FHB) is a devastating disease of Wheat (Triticum aestivum L.) caused primarily by Fusarium graminearum Schwabe in Canada. Wheat FHB results in yield and produce quality losses. The clearest symptoms of FHB are premature bleaching of spikelets in the field, and fusarium damaged kernels in the harvested seed. Insufficient and costly disease control strategies make breeding for FHB resistance in wheat an ideal choice. There is a lack of FHB resistance sources in the Canadian wheat germplasm and Genome Wide Association Studies (GWAS) can be utilized to identify such sources. Furthermore, as no Quantitative Trait Loci (QTL) provides absolute resistance to FHB, identification of novel sources of resistance is desired. Genomic Selection (GS) has great potential in crop improvement, specifically for quantitative traits. A diversity panel that represents the genetic diversity of the Canadian, i.e., high latitude North American winter wheat was assembled for this thesis and the genetic diversity, population structure, and linkage disequilibrium of the germplasm was studied. To identify QTL associated with FHB, a GWAS study was conducted using the phenotypic data collected at three locations over two years. For understanding the role of number of genotypic markers, population structure, and choice of model in trait prediction modelling, a GS study was conducted. The Canadian Winter Wheat Diversity Panel is a diverse collection of winter wheat capturing diversity both in time and geography from Canada. The panel comprises of seven subpopulations with different LD, allele frequencies, and genetic diversity parameters. Multiple QTL associated with FHB related traits were identified on 13 chromosomes which may harbor genes involved in plant defense and stress response mechanisms. For FHB resistance improvement, the GS study demonstrated that the modelling parameters can be determined based upon the genotypic information available. Deoxynivalenol contamination was one of the traits with the highest prediction accuracy. GS can account for minor effect QTL, which is beneficial when breeding for quantitative traits. FHB resistant varieties are needed for an effective and economical disease control strategy in wheat and GS can complement current breeding efforts to develop FHB resistant wheat varieties.

Book excerpt: Wheat and canola are two major economically important food crops grown in western Canada, accounting for billions of dollars in revenue. Fusarium head blight (FHB) in wheat and blackleg in canola are the most destructive diseases that cause economic losses annually. Both diseases are caused by fungal pathogens, where FHB is primarily caused by Fusarium graminearum, while Leptosphaeria maculans causes the blackleg disease. This project's first objective is to evaluate the Fusarium species diversity and deoxynivalenol (DON) levels in western Canadian wheat fields in 2019 and 2020. The analysis of deoxynivalenol (DON) revealed that spring wheat grain contained higher DON levels than winter wheat grain samples. Additionally, for spring wheat, a significantly lower DON content was found in the grain than in the chaff collected from the same wheat heads for both years. The species diversity analysis showed that F. graminearum was the most frequent Fusarium species found in the infected samples except the samples from Alberta, while the highest percentage of F. graminearum was found in the spring wheat samples from Manitoba. The analysis of chemotype diversity of infected samples showed that 3ADON is the dominant chemotype in FHB disease. For the blackleg disease, a growing concern among blackleg researchers is that resistance (R) genes are identified, the same gene as two different genes by two independent laboratories, as not all research laboratories use a standard set of well-characterized isolates. To standardize the R gene identification and novel R gene discovery, the second objective of this thesis is to develop a procedure to generate L. maculans isolates that only carry a single avirulent (Avr) gene. Mating between two L. maculans isolates showed less efficiency in achieving the above objective. Thus, gene editing with the clustered regularly interspaced short palindromic repeat (CRISPR)/Cas9 was utilized. Seven transformed isolates displayed reduced virulence on the canola cultivar Westar, even though they were not mutated at the target gene.

One of the most important fungal diseases of wheat and other cereals in the world is Fusariumhead blight caused by the fungal pathogens Fusarium graminearum and Fusarium culmorumand others. In addition, these fungi produce mycotoxins, contaminating food and feed. The most important Fusarium mycotoxins include trichothecenes, zearalenone and fumonisins, primarily because of their prevalence, but also because of the toxic effect to humans and animals. However, these fungi produce also other mycotoxins such as moniliformin, beauvericin, enniantin or fusarins. Food and feed can be contaminated with mycotoxins at various stages in the production chain resulting in serious problems with health, safety and economic losses. It is estimated that 25% of the crop in the world each year are contaminated with these metabolites, the problem affects both industrialized countries and developing countries. The aim of this Research Topic of Frontiers in Microbiology is to publish state of the art research about occurrence and genomics of Fusarium species and their mycotoxins in the whole food and feed chain starting from the crops as well as implications for health and economic aspects. This research topic highlights the current knowledge on the plant diseases caused by Fusarium fungi as well as all aspects of Fusarium mycotoxin contamination of crops, food and feed, taking into account decontamination methods.
